Abstract
本发明公开了一种促进烟草漂浮苗生长发育的方法,属于植物培养方法领域。本发明中将烟草种子播在漂浮苗专用育苗盘的混合基质中,在由4mmol/LCO(NH2)2,5mmol/LKNO3,0.5mmol/LCa(NO3)2·4H2O,1mmol/LKH2PO4,1mmol/LMgSO4·7H2O,0.06mmol/LEDTA-Fe,0.125mg/LCu,2.86mg/LB,0.4mg/LZn,0.1mg/LMo,2mg/LMn组成的正常培养基中生长到四叶一心时进行完全缺磷胁迫处理20天,然后换到正常营养液中生长17天,再脱水脱肥炼苗三天获得烟草漂浮苗。本发明能够较好的促进烟草漂浮苗移栽前和移栽后根系和地上部生长发育,增加根系与地上部生物量,可为培育强根壮苗提供指导。

技术领域
[0001] 本发明属于植物培养方法领域,特别涉及一种促进烟草漂浮苗生长发育的方法。
背景技术
[0002] 烟草漂浮育苗是我国一种新的和重要的育苗方式,它具有提高育苗效率、能有效 控制土传病害、烟苗素质好等优点,在我国很多地区已被推广应用。烟草漂浮苗从温室苗 床移栽到田间时,由于环境条件的变化,原有主根死亡,大部分侧根不能适应新的环境而凋 亡,烟苗必须在较短时间内长出新的根系才能维持生长,因此有必要在移栽前采取一些措 施,以促进移栽后烟草的正常生长。
[0003] 生产中常在移栽前进行剪叶和炼苗以达到培育发达根系、促进移栽后烟草快 速还苗的目的,二者能够增加烟苗生物量、提高根系活力、缩短还苗期,并且以苗期剪叶 2-3次,剪叶面积1/3-1/2,脱水脱肥炼苗4-6天为最佳(周米良,烤烟漂浮育苗及配套 技术研究.2003;周仰全等,不同断水炼苗时间对烤烟湿润育苗的影响.现代农业科 技.2012,(2) :52-53)。目前,这两项措施已成为烟草漂浮苗育苗方式中必不可少的一部 分。然而,在漂浮育苗实际生产中,部分地区的烟苗在初春移栽时易遭受低温、阴雨天气的 影响,造成烟苗存活率下降、还苗期延长(黄泽春等.不同农艺措施对烟草根系生长影响的 研究进展.作物研究.2008, 22(5) :466-469)。人们虽常采用调整移栽期、覆盖地膜等农艺 措施来缓解外界环境对烟苗生长的影响,但那些措施需要花费较高的人力、物力和财力(罗 新斌等.烤烟覆盖栽培现状与展望.现代农业科技,2010,(24) :47-49),因此仅靠上述两种 技术促根壮苗是不够的,需要探索新的、切实可行的增强烤烟根系发育的办法和措施。
[0004] 磷是植物体必需的一种矿质元素,植物生长环境中供磷量的高低可改变其根系 生长发育状况。植物在低磷下,根系为适应磷胁迫环境,在根系形态、构型及生理等方面 发生一系列的变化,如主根缩短、浅根层根系分布增多、激素含量改变、根系分泌物增加等 (Chiou Tzyy-Jen, Lin Shu-I. Signaling network in sensing phosphate availability in plants. Plant Biology. 2011. 62:185-206;廖红,严小龙·菜豆根构型对低磷胁迫的适 应性变化及其基因型差异·植物学报.2000, 42 (2) :158-163 ;Rengel Z. Genetic control of root exudation. Plant and Soil. 2002, 245 (1) : 59-70)。烟草在低憐胁迫下,根系生 长变化表现为主根伸长,根体积和一级侧根的生长受抑制,生长素/细胞分裂素比值增 大,根冠比增大等(徐敏等,磷对不同基因型烤烟幼苗生长和生理特性的影响,华北农学 报.2006, 21 (3) : 23-26),但也有报道称烟草短期缺磷能够促进根系发育,随时间的延长,缺 磷症状才会逐渐表现出来(刘国顺等,不同供磷能力的土壤施磷对烤烟根体积和根冠比以 及根系伤流组分的影响,中国烟草学报.2009, 15(2) : 28-32),因此适度磷胁迫可能会发挥 烟草耐低磷潜力,并且又能促进根系与地上部生长。苏以荣等(2012)采用砂培培养烟苗得 出烟苗苗期前30天,用0. 02mmol/L的磷胁迫浓度最有利于根系根毛的生长,30天以后到 移栽前烟苗根系最适合在〇. lmm〇l/L的磷胁迫下生长(专利申请号:201210103599. 2),这 种方法虽能够缩短烟草还苗期,促进根系生长,但因其是在砂培中培养的,砂子对磷的吸收 能力强,低浓度的磷胁迫是否为均匀胁迫,并不清楚,而且从砂培中移苗,工序多,劳动强度 大,还可能会伤根。烤烟漂浮育苗是近年来大面积推广的一种育苗方式,因其彻底摆脱了土 壤介质,杜绝了苗期土传病害的发生;而且漂浮育苗在育苗棚内进行,减轻了不良环境对烟 苗的影响,烟苗整齐性高;另外,漂浮育苗在移栽时运苗工作量大幅下降,保证了连片区内 烟株生长的一致性和均匀性(李章海,丁伟.烟草生产理论与技术.中国科学技术大学出 版社.2002:88-89)。然而,如何培育出根系发达、粗状的漂浮苗还需要不断研究。
[0005] 因此,为解决漂浮育苗实际生产中的促根壮苗问题,我们研究了磷饥饿对烟草漂 浮苗生长发育的影响,并结合生产中常用的炼苗措施进一步探讨了一种新型漂浮苗培养方 式,旨在为节约磷素资源,培育壮苗,促进移栽前后烟苗的生长提供一种新方法。
发明内容
[0006] 本发明所解决的技术问题在于提供一种促进烟草漂浮苗生长发育的方法。具体为 提供一种在烟草漂浮苗移栽前磷饥饿与炼苗结合的培育方式,以达到强根壮苗、促进烟苗 根系与地上部生长的目的。
[0007] 本发明所解决的技术问题采用以下技术方案来实现:
[0008] -种促进烟草漂浮苗生长发育的方法,包括以下步骤:
[0009] (1)烟草漂浮苗播种前的准备:将烟草漂浮苗专用育苗盘放在质量份数为 0. 1%ΚΜη04溶液中浸泡15分钟,再盖膜密封2天,然后用清水冲洗干净,在自然光下晾晒3 天;3天后,装上基质和蛭石比为1:1配成的混合基质,混合基质湿度为45%-55% ;
[0010] (2)烟草漂浮苗的播种:将烟草种子播到混合基质中,每穴两粒,播种完毕,在浮盘 表面撒一层1. 5-2. 5_厚的蛭石,以隐约能看见烤烟种子为宜;然后把烟草漂浮苗专用育 苗盘放在含营养液的漂浮池中;每7天换一次营养液。保持育苗棚内温度在22-30°C,若白 天温度高,可揭开薄膜;
[0011] (3 )漂浮烟苗在混合基质中生长到四叶一心时用缺磷培养液代替正常营养液进行 完全缺磷胁迫处理20天,结束后转移到正常营养液中生长17天,然后脱水脱肥炼苗3天, 完成促进烟草漂浮苗生长发育的培植,所得的烟草漂浮苗用于移栽到土壤中培养。
[0012] 步骤(1)中所述的基质是由草炭、膨胀珍珠岩、腐殖质、蛭石、碳化谷糠和泥炭按质 量比为1 :1 :1 :1 :1 :1的比例配成的基质;

[0018] 本发明能够很好的促进烟草漂浮苗生长发育,其创新之处在于本方法根据烟草漂 浮苗的培育及生长特点,用0. 5mmol/L K2S04R替培养液中的lmmol/LKH2P04,直接在烟草漂 浮苗池进行完全缺磷胁迫,并且结合移栽前脱水脱肥炼苗,能够充分发挥烟草耐磷潜力,可 为漂浮育苗实际生产提供指导。

[0022] 将烟草漂浮苗专用育苗盘放在质量分数为0. 1%ΚΜη04水溶液中浸泡15分钟,再 盖膜密封2天,然后用清水冲洗干净,在自然光下晾晒3天。3天后,装上基质(广州三力园 艺公司提供)和蛭石比为1:1配成的混合基质,基质湿度为45%-55%。然后将K326包衣种 子(中国烟草中南农业试验站永州基地提供)播到基质中,每穴两粒,播种完毕,在种子表面 撒一层大约2mm厚的蛭石。最后将播完种的漂浮盘放在含营养液(营养液组成为:4_〇1/ L C0(NH2)2,5mmol/LKN03,0. 5mmol/L Ca(N03)2 · 4H20, lmmol/L KH2P04, lmmol/L MgS04 · 7H20, 0. 06mmol/L EDTA-Fe,0· 125mg/L Cu,2. 86mg/L B,0· 4mg/L Zn,0· lmg/L Mo, 2mg/L Mn,其中 Cu、B、Zn、Mo、Μη 分别由 CuS04 · 5H20、H3B03、ZnS04 · 7H20、(NH4)2M〇704 · 4H20、MnS04 · H20 提 供)的漂浮池中。每7天换一次营养液。10-12月份在华南农业大学资源环境学院网室大 棚中进行实验。网室采用自然光照,保持平均温度24°C。

[0034] 每个处理完成后移栽到土培盆栽中。每盆栽一株,每个处理6个重复。移栽后10 天内观察烟苗长势,并在移栽后的第10天取样,测定根系干重、地上部干重等指标。
[0035] 表2和表3分别为完全缺磷胁迫不同时间对烟草漂浮苗移栽前和移栽10天后烟 苗生长的影响。由表2结果可知,处理VI(完全缺磷胁迫25天)已出现缺素症状,总根数、 根体积及植株生物量均下降。与对照处理I (未进行缺磷胁迫处理)相比,移栽前处理V的 总根数、根体积分别增加157. 50%、225. 24%,根冠比增加183. 15% (见表2),移栽10天后, 处理V的叶片数、根系干重及根冠比分别增加112. 50%、145. 45%、131. 32% (见表3)。处理 II、处理III、处理IV随缺磷时间的延长,总根数、根体积、根干重、地上干重及根冠比均逐 渐增加,但除地上部干重外,处理V (漂浮苗四叶一心时完全缺磷胁迫20天)的总根数、根体 积、根干重及地上干重等在这4个处理中增幅最大,促进烟草生长效果最显著(见表2)。图 1为完全缺磷胁迫不同时间对移栽前烟草漂浮苗根系生长影响的根系图,由图1可以看出, 在烟草漂浮苗四叶一心时,进行完全缺磷胁迫20天能够明显促进烟苗生根,最有利于烟苗 移栽前和移栽后根系和地上部的生长。

[0044] 生产中烟草漂浮苗在移栽前通常会进行炼苗处理,以提高烟苗的抗逆性和移栽后 烟苗的返青速度。因此,结合实施例1的结果,探讨了完全缺磷胁迫与炼苗对烟草漂浮苗生 长发育的影响。

[0047] 处理II :进行完全缺磷胁迫20天,处理结束,转移到正常营养液中再生长17天,然 后断水断肥炼苗3天,结束后移栽到土培盆栽中;
[0048] 处理III :烟苗在正常营养液中长到六叶一心时,断水断肥炼苗6天,处理结束直接 移栽到土培盆栽中;
[0049] 处理I和处理II中的完全缺磷胁迫处理与实施例1中的完全缺磷胁迫处理相同。 处理完成后,将烟苗移栽到土培盆栽中。每盆栽一株,每个处理6个重复。移栽后10天内 观察烟苗长势,并在移栽后的第10天取样,测定根系干重、地上部干重等指标。
[0050] 表4和表5探讨了完全缺磷胁迫与炼苗结合对烟草漂浮苗移栽前和移栽后10天 生长发育的影响。移栽前,处理II的根冠比分别是处理I和处理III的107. 69%、187. 63%,总 根数、根体积、根干重及地上部干重等处理II和处理I相差不大(见表4);移栽10天后,处 理II的叶片数、根干重、地上部干重分别是处理I的111. 11%、137. 5%、104. 02%,是处理III的 123. 08%、169. 23%、111. 73%(见表5)。可见,在烟草漂浮苗移栽前,进行完全缺磷胁迫后,再 进行炼苗3天,最能促进烟草移栽后烟叶的生长及根系干重的增加。

1. 一种促进烟草漂浮苗生长发育的方法,其特征在于包括以下步骤: (1) 烟草漂浮苗播种前的准备:将烟草漂浮苗专用育苗盘放在质量份数为0. 1% ΚΜη04 溶液中浸泡15分钟,再盖膜密封2天,然后用清水冲洗干净,在自然光下晾晒3天;3天后, 装上基质和蛭石比为1:1配成的混合基质,混合基质湿度为45% -55%;所述的基质是由草 炭、膨胀珍珠岩、腐殖质、蛭石、碳化谷糠和泥炭按质量比为1 :1 :1 :1 :1 :1的比例配成的基 质; (2) 烟草漂浮苗的播种:将烟草种子播到混合基质中,每穴两粒,播种完毕,在浮盘表 面撒一层1. 5-2. 5_厚的蛭石,以隐约能看见烤烟种子为宜;然后把烟草漂浮苗专用育苗 盘放在含营养液的漂浮池中;每7天换一次营养液;保持育苗棚内温度在22-30°C,若白天 温度高,可揭开薄膜; (3) 漂浮烟苗在混合基质中生长到四叶一心时用缺磷培养液代替正常营养液进行完全 缺磷胁迫处理20天,结束后转移到正常营养液中生长17天,然后脱水脱肥炼苗3天,完成 促进烟草漂浮苗生长发育的培植,所得的烟草漂浮苗用于移栽到土壤中培养。
