本发明涉及使用包含过乙酸的蒸气对表面进行杀菌的方法。所述蒸气通过以下方式产生:将包含稀的过乙酸溶液的溶液引入热气流中。使用这种真的蒸气(true vapor)的结果是实现所需的对基材的灭菌,同时不会在基材表面上沉积冷凝液滴。
发明背景
长期以来,人们已认识到了为了健康和卫生对物件表面进行杀菌的必要性。人们需要有效的杀菌方法用于各种目的,包括无菌包装,医疗设备杀菌,生物杀灭带菌体以保护环境,熏蒸消毒,容器杀菌,食物处理等。
现有技术已知的可以用于杀灭细菌的化合物是过乙酸。一般来说,过乙酸被用于水基体系,例如包含过乙酸、乙酸和过氧化氢的平衡混合物。虽然已经证明该体系是有效的,但是在许多情况下,需要进行独立的冲洗和/或干燥步骤。这样会给杀菌工艺增加相当大的费用和时间,尤其当加快干燥过程所需的高温可能对基材造成不利影响的时候。
因此人们非常希望具有使用过乙酸作为杀菌剂的方法,该方法不需要耗能或耗时的干燥步骤。
发明概述
本发明涉及一种对材料进行杀菌的方法,该方法包括以下步骤:
a)将包含过乙酸的溶液引入热气流中,产生过乙酸蒸气;以及
b)使该过乙酸蒸气与待杀菌的材料接触。
通过该方法能够对材料进行有效的杀菌,同时因为在这样处理的材料的表面上不会形成和沉积溶液的液滴,因此不需要随后的干燥步骤。因此,可以使用本发明的方法对很多种材料进行快速而经济的杀菌。
发明详述
本发明涉及一种对材料进行杀菌的方法,该方法包括以下步骤:
a)将包含过乙酸的溶液引入热气流中,产生过乙酸蒸气;以及
b)使该过乙酸蒸气与待杀菌的材料接触。
在本发明中,术语“蒸气”是用来表示过乙酸基本完全处于气体形式的状态。这与薄雾或雾不同,薄雾和雾均包含显著部分的液滴悬浮在空气中。与使用薄雾或雾状态的过乙酸的情况不同,发现使用蒸气形式的过乙酸能够对材料进行极佳的杀菌,同时不会在材料表面上伴随形成水滴。
过乙酸通常以乙酸、过氧化氢和过乙酸的水性平衡混合物[比例为35∶10∶15]的形式使用。该组合物通常还可以包含稳定剂,例如膦酸或膦酸盐,即Degquest2010或螯合剂,例如吡啶二羧酸,以及其它的组分,如:无机酸催化剂(硫酸,硝酸或磷酸);表面活性剂,例如阴离子型月桂酸盐,脱水山梨糖醇以及它们的相应的酯,即聚亚乙基脱水山梨糖醇单月桂酸酯;以及在溶液中形成混合的过酸的短链脂肪酸酯(C6-C12)。
在引入加热的气流中之前,优选通过加入水将所述过乙酸稀释至浓度约小于10,000ppm,优选约小于4,000ppm。
所述加热的气流通常是无菌空气,但是也可以使用其它的气体,例如氮气,CO2,或惰性稀有气体载体。所述气流通常可以至少加热至约300℃,优选最低温度约为250℃,可以超过350℃,只要其能够充分进行冷却以供应用即可。通常随后将其冷却至约80-120℃,然后引入过乙酸溶液。在加入过乙酸的时候,加热的气流的温度应当至少比过乙酸的露点(约46.5°-49.9℃)高5℃;也就是说,至少约为55℃,从而确保将过乙酸转化为蒸气,而不转化为薄雾或雾。
可以通过本领域技术人员众所周知的任意方法将过乙酸引入加热的气流中。一种优选的方法是直接注入溶液。
然后使过乙酸蒸气与待杀菌的材料接触一段足以杀灭所关心的污染物的时间。该段时间将根据一些变量变化,这些变量包括使用的过乙酸蒸气的浓度;待杀菌的材料的表面的性质;待杀菌的具体污染物;待杀菌的污染物的浓度;等等。一般来说,所述接触通常保持约15-40分钟的时间。
可以使用本发明的方法对很多种材料进行杀菌,包括金属、塑料、聚合物和弹性体的硬表面。
该方法可以用来对被细菌污染的材料进行杀菌,而人们通常用液体形式的过乙酸对这些细菌进行抑制。这些包括杆菌类的细菌和孢子,用苏云金杆菌(B.thuringiensis)和萎缩芽孢杆菌(B.atrophaeus)作为致病性更强的种类(形式)例如肉毒芽孢杆菌(C.botulinum)的替代物;以及更常规种类的经常用PAA进行抑制的细菌、真菌和病毒以及原生动物,例如(但不限于):葡萄球菌(Staphlococcus),肠球菌(Enterococcus),沙门氏菌(Salmonella),弯曲杆菌(Capmylobacter),假单胞菌(Pseudomonas),假丝酵母菌(Candida),根霉菌(Rhizopus),毛霉菌(Mucor),流感病毒(Influenza)等。
提供以下实施例来进一步举例说明本发明的方法。
实施例
使用DA 2000无菌袋式填料,在17分钟的时间内将1.5升过乙酸的稀溶液(浓度为4000PPM)注入无菌空气流中,所述无菌空气流此前已经加热至300℃并冷却至90℃。没有观察到薄雾、雾或冷凝,表明发生了完全的蒸发。将用所示浓度的所示孢子形成有机体接种的采样管放在机器的“头部”无菌区域内,使其接触过乙酸蒸气。在任何被测的采样管上都没有观察到冷凝。在过乙酸溶液注入完成之后,立刻将采样管从机器中取出。将样品从采样管中取出,转移到合适的生长培养基上,培养并监控其生长。该测试的结果见表1。

Claims (12)
1.一种对材料进行杀菌的方法,所述方法包括以下步骤:
a)将包含浓度小于10,000ppm的过乙酸的溶液引入热气流中,产生过乙酸蒸气;以及
b)使该过乙酸蒸气与待杀菌的材料接触,同时不会在所述材料表面上沉积冷凝水滴。
2.如权利要求1所述的方法,其特征在于,所述材料选是金属。
3.如权利要求1所述的方法,其特征在于,所述材料是塑料。
4.如权利要求1所述的方法,其特征在于,所述材料是聚合物。
5.如权利要求1所述的方法,其特征在于,所述材料是弹性体。
6.如权利要求1所述的方法,其特征在于,将所述过乙酸稀释至浓度小于4,000ppm,然后引入所述热气流中。
