Description
一种HPV多肽/DC混合疫苗及其制备
技术领域
[0001] 本发明属于医学生物工程技术领域,具体地说涉及Toll样受体(TLR)的激动 剂,作为一种HPV11E7表位肽特异性DC疫苗的佐剂,以期用于尖锐湿疣和宫颈癌的防 治。
背景技术
[0002] 人类乳头瘤病毒(HPV)可引起我国目前高发的性传播性疾病一尖锐湿疣以及 HPV高相关性宫颈癌。尖锐湿疣的病原体主要为HPV6和11型,复发率高,顽固难治; 而宫颈癌至今疗效和预后不佳,且逐年出现年轻化趋势,两者均成为严重危害患者、家 庭、乃至社会的重大隐患。
[0003] 尽管宫颈癌预防性疫苗已在多个国家上市,并显示了较理想的疗效。然而,目 前多数HPV治疗性疫苗对人类机体细胞免疫的诱导激活较有限,临床试验也并未达到预 期结果。由于HPV感染患者机体免疫力往往比较低下,不足以激发有效的细胞免疫清除 病毒感染,而临床上又缺乏高效、特异性的抗HPV药物,因此,如何增强HPV蛋白的免 疫原性、促进抗原的呈递以及寻找高效的佐剂等来提高机体的特异性抗病毒免疫是HPV 疫苗研究中需迫切解决的关键问题。
[0004] Toll样受体(TLR)是存在于树突状细胞(DC)等抗原提呈细胞上的病原相关模 式识别受体。当病原体感染机体时,TLR识别相应的配体而活化,通过不同的细胞内信 号转导通路进而调节基因表达,从而在机体抗感染的天然免疫应答过程中发挥至关重要 的作用。参与机体抗病毒免疫的主要有四种TLRs :以polyI:C (PIC)为激动剂的TLR3 ; 以咪喹莫特等为激动剂的TLR7/8 ;以CpG为激动剂的TLR9。近来发现某些TLR激动 剂可明显上调病毒抗原肽引起DC分化及功能成熟,从而引发更强的抗原特异性细胞免疫 应答,因此认为TLRs在激发机体特异性抗病毒免疫、清除病毒感染的过程中也可能起着 关键作用。而其激动剂也可能作为重要的免疫佐剂来上调各种预防性或治疗性疫苗的作 用。
发明内容
[0005] 本发明的目的在于提供一种HPV多肽/DC混合疫苗,所述的混合疫苗,主 要由一份HPV11E77_15,一份体外培养的小鼠骨髓来源的树突状细胞(BM-DC),一份 CpG(TLR9激动剂)作为佐剂组成。
[0006] 本发明的另一个目的是提供一种HPV多肽/DC混合疫苗的制备方法,通过以下 技术方案实现:
[0007] (1)从小鼠骨髓提取树突状细胞(BM-DC),进一步体外培养;
[0008] (2)先与免疫原性最强的一段HPV11E7CTL表位肽(HPV11E77_15, TLKDIVLDL)共孵育;
[0009] (3)再分别与TLR配体CpG等共孵育,以促进DC成熟;[0010] (4)检测孵育后DC表面标志改变以确认其成熟度;
[0011] (5)上述刺激成熟的DC疫苗可用于免疫小鼠。
[0012] 本发明将TLR配体与HPV11E7多肽联合使用可促进DC的成熟度,尤其联用 CpG后可提高T细胞分泌细胞因子TNF- α和IFN_ Y的水平。因此认为TLR9激动剂 CpG对HPV11E7CTL表位肽在小鼠模型中的免疫应答具有一定程度的上调作用,可作为 免疫佐剂以提高HPV11E7表位肽特异性的DC疫苗的免疫效应。本发明针对现有疫苗的 不足,用TLR激动剂(CpG)作为免疫佐剂,以提高HPV11E7表位肽特异性的DC疫苗的 免疫效应,用于尖锐湿疣和宫颈癌的防治。

具体实施方式

[0021 ] 实施例1小鼠骨髓来源的HPVl 1Ε7多肽/DC混合疫苗的制备
[0022] 1)方法:
[0023] 将小鼠胫骨和股骨的骨髓吹打成细胞悬液,加入氯化铵裂解红细胞,用含 murine GM-CSF (10ng/ml)、muring IL-4 (lng/ml)及 10 %胎牛血清的 RPMI1640 培养液 3ml/孔重悬后培养于六孔板。48小时后,弃去全部培养液,加2ml/孔培养液和细胞因 子。48小时后,收集全部细胞,重悬后分为5组,各加不同刺激剂共孵育48小时:(1) 肽(20 μ g/ml)和咪喹莫特(2 μ g/ml),(2)肽(20 μ g/ml)和 PIC (20 μ g/ml),(3)肽 (20 μ g/ml)和 CpG (20 μ g/ml),(4)肽(20 μ g/ml),(5))空白对照(不加任何成分), 其中TLR配体均为肽加入2小时之后再加入。
[0024] 2) DC形态观察:
[0025] DC分离培养第3天时倒置显微镜下即可见DC呈典型的集落样生长,经第5天 加入抗原肽及TLR配体冲击48小时后,于第7天倒置显微镜下可见DC悬浮生长,突触 明显增多,呈典型的树突状突起。
[0026] 实施例2流式细胞术检测DC的表面分子标记变化的鉴定:
[0027] 1)方法:
[0028] DC 培养的第 7 天,将 DC (1 X IO5 个 /IOOul)与 FITC 标记的鼠 CD40、CD80、CDllc和PE标记的CD86、Iab抗体共孵育30分钟后,进行流式细胞术检测。
[0029] 2)结果: [0030] 经流式细胞仪分析,CDllc阳性细胞占81.3%。于第5天和第7天行流式细胞 仪分析DC表型,结果参见图1,图1是BM-DC与HPVl 1E7多肽及TLR激动剂共同孵 育48小时后,用流式细胞仪检测DC表面标志CD40、CD80、CD86及MHCII(I_Ak)以
确定其成熟度。
[0031] 该结果提示仅用抗原肽冲击后,DC成熟度均较未冲击组稍有增高,但加入TLR 配体处理后,各处理组较对照组及抗原肽组均提高显著,尤其以CD40和CD86较明显。 其中:
[0032] CD40 :咪喹莫特组和PIC组与肽组及对照组相比均有显著差异(P < 0.05)
[0033] CD86 :咪喹莫特组、PIC组和CpG组与对照组相比均有显著统计学差异(P <0.05),其中咪喹莫特组、PIC组与肽组相比亦有显著统计学差异(P <0.05)。
[0034] 而CD80与MHC II(I-Ak)各组之间相比较均未见明显统计学差异。
[0035] 实施例3小鼠接种疫苗及免疫学特性观察
[0036] 1)接种方法:
[0037] 将5组不同处理后的DC用PBS洗至少2次后,调整细胞浓度为3X 105/200 μ 1/ 只,注射入小鼠腹腔(6只Χ5组)。之后每隔1周注射1次,共计注射3次。
[0038] 2)小鼠荷瘤的免疫治疗模型方案(体内实验):
[0039] 将小鼠用DC免疫3次,于第3次免疫后的第2天,在每只小鼠左侧腹股沟皮下 接种Β16细胞(1Χ 105/200 μ1/只)。接种肿瘤后的第19天,处死全部小鼠,制备脾细 胞悬液,贴壁2小时后,收集悬浮细胞,用含mUringIL-2(20U/ml)和10%胎牛血清的 RPMI1640培养液重悬后培养于六孔板(约7X 106/ml),每孔加入肽(20 μ g/ml),以行 细胞因子的检测。
[0040] 3)体外实验:
[0041] 将HPV11E7CTL表位肽和TLR配体处理后的5组DC分别与未经任何处理的小 鼠脾细胞(约2.1 X 1073ml/孔)以1 : 17混合培养。
[0042] 4)检测小鼠脾细胞在体内外分泌细胞因子水平(TNF- α和IFN_ Y ):
[0043] 取脾细胞培养48小时后的培养液上清,用ELISA方法检测细胞因子,具体 步骤参照试剂盒说明书(mTNF- α ELISA Kit, BIOSOURCE ; mIFN_ Y ELISA Kit, BIOSOURCE),ELISA板在450nm处读取,并参照标准曲线确定细胞因子的浓度。
[0044] 5)免疫学检测结果:

[0046] 将各组小鼠处死,取脾细胞培养,并加入HPVl 1E7CTL表位肽刺激,48小时后 取小鼠脾细胞培养上清检测细胞因子分泌水平,结果参见图2,图2是用经HPV11E7多 肽及TLR激动剂刺激的BM-DC免疫小鼠,再用B16肿瘤细胞皮下荷瘤,然后分离脾脏 T细胞,与相应多肽重新孵育后用ELISA测定细胞因子分泌情况。
[0047] 在分泌TNF- α水平上,PIC和CpG组显著高于对照组(P < 0.05),但与肽组相 比均未见统计学差异。在分泌IFN-Y水平上,PIC、CpG和抗原肽组均显著高于对照组 (P < 0.05),同时CpG组与抗原肽组相比亦有明显差异(Ρ<0.05)。说明在促进TNF-a分泌上,单用肽效果较微弱,但联合PIC和CpG后效果即较为明显,以CpG组为著,可 见PIC和CpG可上调肽促进脾细胞TNF-α分泌的水平;而在促进IFN_ Y分泌上,单用 肽即可见明显促进效果,联用PIC后未见效果有明显增加,但联用CpG后发现,效果较 单用肽亦有明显增加,因此,可见CpG可显著上调肽促进脾细胞IFN-Y分泌的水平。而 咪喹莫特组,在检测TNF-α和IFN-Y的水平上,均未见明显效果。

[0049] 将各不同处理组的DC与未经任何处理小鼠的脾细胞混合培养,取培养48小时 后的上清行ELISA检测,结果参见图3,图3是小鼠BM-DC用HPVl 1Ε7多肽及TLR激 动剂刺激后,在体外与未处理的新鲜脾细胞共同孵育,再用ELISA测定细胞因子分泌情况。
[0050] 从分泌IFN- Y水平上看,咪喹莫特和CpG组效果较为明显(P < 0.05),同时在 分泌TNF-α水平上,咪喹莫特组效果仍较为明显(P < 0.05),其次以CpG组效果为较好。
[0051] 综上所述,TLR配体与HPV11E7多肽联合使用可促进DC的成熟度,尤其联用 CpG后可提高T细胞分泌细胞因子TNF- α和IFN_ Y的水平。因此认为TLR9激动剂 C pG对HPV11E7CTL表位肽在小鼠模型中的免疫应答具有一定程度的上调作用,可作为 免疫佐剂以提高HPV11E7表位肽特异性的DC疫苗的免疫效应。
