Die Erfindung bezieht sich auf neue heterocyclisch substituierte Phenoxycarbonsäuren und Derivate mit pharmakologischer Wirksamkeit zur Behandlung von Hypercholesterin ämie und Hyperlipidämie in der Humanmedizin und insbesondere auf Verbindungen der genannten Art, die am Phenoxyteil und gegebenenfalls auch im Alkoxylatteil einen heterocyclischen Substituenten tragen.

wird klinisch zur Verminderung überhöhter Cholesterinkonzentrationen im Blut bzw. Blutserum eingesetzt. Die relativ hohe Toxizität von Clofibrat (LDso = 2350 mg/kg, oral, Mäuse) und die Notwendigkeit, diese Verbindung wegen ihrer relativ geringen therapeutischen Wirksamkeit (ED2s = 165 mg/kg) in entsprechend hohen Dosen verwenden zu müssen, sind nachteilig. Der Verhältniswert Toxizität: therapeutische Wirksamkeit wird auch als therapeutischer Index bezeichnet und beträgt bei Clofibrat etwa 14; Verbindungen mit vergleichsweise geringeren Toxizitätswerten (höheren LDso-Werten) oder/und vergleichsweise höheren therapeutischen Wirksamkeiten (kleinere ED2s-Werte), d.h.
mit im Vergleich zu Clofibrat höheren therapeutischen Indexwerten, wären wünschbar.
Aufgabe der Erfindung sind daher Verbindungen, die sich zur therapeutischen Verwendung für die Verminderung überhöhter Cholesterin- oder/und Lipidwerte des menschlichen Blutes bzw. Blutserums eignen und vergleichsweise wesentlich höhere therapeutische Indexwerte als Clofibrat haben.
Es wurde gefunden, dass die von der Anmelderin z.B. in der DE-OS 2461 069 beschriebenen Verbindungen der Formel (10)

in welcher R' Wasserstoff, Halogen, Niederalkyl oder Niederalkoxy und X Alkyl bedeuten, ihre im Vergleich zu Clofibrat besseren Wirkungen mindestens teilweise der
asymmetrischen Struktur der in Formel (10) als A' bezeichneten Gruppe verdanken; wenn R" und R"' jeweils unterschiedliche Niederalkylgruppen bedeuten, werden besonders vorteilhafte Wirkungen erzielt.
Es wurde überraschenderweise gefunden, dass auch dann, wenn der Ring B von Formel (10) kein Benzolring, sondern ein heterocyclischer und gegebenenfalls alkyl- oder halogensubstituierter heteroaromatischer Ring ist, die Gruppe A' asymmetrisch ist (R" und R"' sind unterschiedliche Niederalkylgruppen) und die Methylenbrücke zwischen den Ringen B, C in para-Stellung oder aber in ortho-Stellung zur Sauer stoffbrücke zwischen dem Ring C und der asymmetrischen Gruppe A' steht, im Vergleich zum Clofibrat geringere Toxizitäten und höhere Wirksamkeiten, d.h. erheblich höhere therapeutischen Indexwerte erzielt werden.

Beispiele von Kationen von zur Salzbildung mit der Carboxylgruppe geeigneten, pharmakologisch zulässigen Basen sind Alkali- oder Erdalkalimetall-lonen wie das Calciumoder Magnesiumion, das Aluminiumion oder organische Amine wie Diethylaminoethanol, Piperazin.
Beispiele für erfindungsgemäss geeignet Gruppen Q sind Thienylreste (z.B. Thiophen-2-yl oder -3-yl-), die gegebenenfalls einfach oder mehrfach durch Niederalkylgruppen (CI-C4) oder/und Halogenatome (Cl, Br, J) einfach oder mehrfach substituiert sind; Furylreste (z.B. Furan-2-yl- oder -3-yl-), die gegebenenfalls in analoger Weise wie die Thienylreste substituiert sind; und Pyridylreste (z.B. Pyridin-2-yl oder -3-yl oder -4-yl), die ebenfalls durch ein oder mehrere Niederalkylgruppen oder/und Halogenatome der genannten Art substituiert sein können.
Der Rest Q kann auch mehrere gleiche oder unterschiedliche Heteroatome enthalten.
Gemäss einer bevorzugten Ausführungsform bedeutet Rl in der Formel (1) den Methylrest und R2 den Ethyl-, Propyloder Butylrest, wobei der Ethylrest für R2 besonders bevorzugt wird.
Wegen der Unterschiedlichkeit von R' und R2 sind die Verbindungen (I) optisch aktiv und die Erfindung umfasst sowohl die D- bzw. L-Isomeren der Formel (1) als auch racemische Mischungen.
Vorzugsweise ist R3 eine Niederalkylgruppe mit 1 bis 4 C-Atomen, insbesondere die Ethylgruppe. R3 kann auch eine Methylpyridingruppe

Wenn der Rest Q in Formel (1) basisch ist und/oder R3 eine Methylpyridingruppe darstellt, können erfindungsgemässe Verbindungen als Säureadditionssalze vorliegen. Für die therapeutische Verwendung sind dies die Additionssalze mit pharmakologisch zulässigen anorganischen Säuren, wie Halogenwasserstoff, insbesondere HCI, Salpetersäure Schwefelsäure, Phosphorsäure, usw., oder mit pharmakologisch zulässigen organischen Säuren, wie Milchsäure, Zitronensäure, Apfelsäure, Weinsäure, usw. Die Wahl geeigneter Säuren liegt im Rahmen des fachmännischen Wissens.
Erfindungsgemässe Verbindungen können nach verschiedenen, an sich bekannten Methoden hergestellt werden, z.B.

(3) haben die für Formel (1) oben angegebene Bedeutung; R4 kann gleich wie R3 in Formel (1) sein oder das Wasserstoffatom bzw. eine andere Alkylgruppe als R3 sein, in welchem Fall R4 durch Veresterung bzw. Umesterung in R3 umgewandelt wird. Die Umwandlung basischer Verbindungen der Formel (1) in die entsprechenden Säureadditionssalze kann in an sich bekannter Weise erfolgen.
Zur therapeutischen Verwendung können die erfindungsgemässen Verbindungen in an sich üblicher Weise, etwa wie in der US-PS 3'262'850 beschrieben, dosiert und verwendet werden, z.B. als solche, in Mischung miteinander und insbesondere in Mischung mit üblichen flüssigen, halbfesten oder festen pharmakologischen Trägerstoffen bzw. Konfektionierungsmitteln, gegebenenfalls zusammen mit anderen pharmakologisch aktiven Substanzen.
Erfindungsgemässe Verbindungen sowie ihre Herstellung und Eigenschaften werden anhand der folgenden, nichtbeschränkenden, Beispiele erläutert.

Herstellung der Verbindung der Formel (11)

In einen 500 ml-Dreihalskolben mit Rührer, Rückflusskühler und Stickstoffzuleitung wurden 100 ml abs. Ethanol und 4,83 g (0,21 Mol) frisch geschnittenes metallisches Natrium eingegeben. Nach Auflösung des Natriums wurde die Reaktionsmischung unter vermindertem Druck auf einem Büchi Verdampfer eingedampft.
Zu so gebildeten Natriumethoxid wurde eine Lösung von 40 g (0,20 Mol) 2-(4-Hydroxybenzyl)-thiophen in 200 ml trok kenem Xylol gegeben und die Mischung 90 min auf dem Ölbad unter Rühren auf Rückfluss gehalten. Dann wurde der Rückfluss unterbrochen und die Reaktionsmischung innerhalb von 10 Min mit 43,9 g (0,21 Mol) Ethyl-d,l-2-brom-2methyl-butyrat durch Zutropfen versetzt. Die Umsetzung wurde weitere 20 Std. laufen gelassen und dann die Lösungsmittelanteile unter Vakuum abgedampft. Das zurückbleibende braune Öl wurde mit 300 ml Diethyläther behandelt und die Mischung zweimal mitje 150 ml wässriger 2n Natriumhydroxidlösung extrahiert. Die organische Phase wurde mit Wasser bis zur Alkalifreiheit gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und unter vermindertem Druck eingedampft.
Als Rückstand wurde das rohe Zielprodukt in Form von gelbem Öl in einer Menge von 56,0 g erhalten. Das Rohprodukt wurde unter vermindertem Druck zur Gewinnung des reinen Zielproduktes der Formel (11), Ethyl-d, 1 -2-methyl-2-[4-(thienyl-2-methyl)-phenoxyj- butyrat, Kp 207-209"C/ 1 mm Hg, in einer Menge von 32,0 g destilliert.

Eine Lösung von 33,0 g (0,19 Mol) 2-(4-Hydroxybenzyl)furan in 160 ml trockenem Xylol wurde zu 13,0 g (0,19 Mol) frisch hergestelltem Natriumethoxid gegeben und die Mischung 1 Std. unter Rückfluss gehalten. Dann wurde innerhalb von 15 min eine Lösung von 39,71 g (0,19 Mol) Ethyl-d, l-2-brom-2-methylbutyrat in 200 ml trockenem Xylol vorsichtig zugegeben. Nach 23 Std.
Umsetzungsdauer wurde die Reaktionsmischung wie in Beispiel 1 aufgearbeitet und ergab 49,0 g öliges Rohprodukt, das nach Destillieren 32,0 g Zielprodukt der Formel (12), Ethyl-d,1-2-methyl-2-[4- (2-furanmethyl)-phenoxy]-butyrat, Kp 134-135"C/ 0,02 mm Hg, ergab.

Eine Lösung von 37,04 g (0,20 Mol) 2-(4-Hydroxybenzyl)pyridin in 200 ml trockenem Xylol wurde zu 13,60 g (0,20 Mol) frisch hergestelltem Natriumethoxid gegeben. Dann wurde eine Lösung von 41,8 g (0,20 Mol) Ethyl-d, l -2-brom- 2-methylbutyrat in trockenem Xylol innerhalb von 10 Min zugetropft. Nach 20 Std. Umsetzung und Aufarbeitung wie in Beispiel 1 wurden 51,0 g öliges Rohprodukt erhalten, das durch Destillation unter vermindertem Druck 33,5 g farbloses Zielprodukt gemäss Formel (13), Ethyl-d,1-2-methyl-2- [4-(2-pyridinmethyl)-phenoxy]-butyrat, Kp 147-1 490C/ 0,03 mm Hg, lieferte.

Eine Lösung von 35,1 g (0,16 Mol) 5-Chlor-2-(4-hydroxybenzyl)-pyridin in 190 ml trockenem Xylol wurde zu 10,88 g (0,16 Mol) frisch hergestelltem Natriumethoxid gegeben und die Mischung 2 Std. auf Rückfluss erhitzt. Dann wurde innerhalb von 15 Min eine Lösung von 33,5 g (0,16 Mol) Ethyl dl -2-brom-2-methylbutyrat in 100 ml trockenem Xylol zugegeben. Nach 22 Std.
Umsetzung und Aufarbeitung wie in Beispiel 1 wurden 20 g öliges Rohprodukt erhalten, das nach dem Destillieren unter vermindertem Druck 12 g reines Zielprodukt der Formel (14), Ethyl-d, 1 -2-methyl-2-[4-(5-chlor 2 pyridinmethyl)-phenoxy]-butyrat, als farbloses Öl, Kp 170 C/0,005 mm Hg, ergab.

Eine Lösung von 8,0 g (0,036 Mol) 6-Chlor-2-(4-hydroxybenzyl)-pyridin in 25 ml trockenem Xylol wurde zu 2,44 g (0,036 Mol) Natriumethoxid gegeben und die Mischung 2 Std. auf Rückfluss erhitzt. Dann wurde eine Lösung von 7,6 g (0,036 Mol) Ethyl-d,l-2-brom-2-methylbutyrat in 50 ml trockenem Xylol zur Mischung gegeben. Nach 22 Std.

Herstellung der Verbindung der Formel (16)

Eine Lösung von 10,0 g (0,036 Mol) 2-(2-Oxybenzyl)-thiophen in 25 ml wasserfreiem Xylol wurde zu 3,4 g (0,05 Mol) Natriumethoxid gegeben. Die Mischung wurde 11/2 Stunden auf Rückfluss erhitzt. Dann wurde eine Lösung von 11 g (0,05 Mol) Ethyl-dl-2-methyl-2-brom-2-methyl-butyrat in 5 ml Xylol tropfenweise zugesetzt und weitere 22 Stunden auf Rückflusstemperatur gehalten. Nach Abdampfen des Lösungsmittels erhielt man als Rückstand 5 g Zielprodukt der Formel (16), Ethyl-dl-2-methyl-2-[2-(thienyl-2-methyl)phenoxy]-butyrat, als farbloses Öl mit Kp 168"C/ 0,004 mm Hg.

Von den basischen Verbindungen der Formel (1) gemäss der Erfindung, z.B. den Verbindungen der Formeln (13), (14) und (15), können die entsprechenden erfindungsgemässen
Säureadditionssalze in an sich üblicher Weise durch Umset zung stöchiometrischer Mengen der jeweiligen basischen
Verbindung der Formel (1) und einer anorganischen oder organischen Säure, z.B. HCI, HNO3, Milchsäure, Zitronen säure oder dergleichen, vorzugsweise mit pharmakologisch zulässigen Säuren der genannten Art und in entsprechenden
Lösungen erhalten werden.
Die Toxizitätswerte (LDso) in mg Substanz pro kg Körper gewicht an Mäusen bei oraler Verabreichung während 10
Tagen und üblicher Bestimmung gemäss J. Pharmacol. 95,
1949, S. 99 ff, der getesteten Verbindungen der Formel (1) sind erheblich günstiger als die der Vergleichssubstanz Clo fibrat , wie sich aus folgender Tabelle I ergibt.

Die Werte der therapeutischen Wirksamkeit in bezug auf
Cholesterinsenkung (ED2s; in mg/kg) erfindungsgemässer
Verbindungen wurden in an sich bekannter Weise bestimmt und bezeichnen jeweils die Mengen (mg) an aktiver Substanz, die pro kg Körpergewicht bei männlichen Wistar-Ratten (von 100-200 g Körpergewicht und 8-10 Versuchstieren pro
Dosierungswert) gegenüber der Kontrollgruppe eine 25%ige
Verminderung des mittleren Cholesteringehaltes im Blut serum der Versuchstiere bewirken.
Die Testsubstanzen wurden oral in analoger Weise wie bei
Bestimmung der LD50-Werte als Suspensionen in wässriger Arabischgummilösung verabreicht. Jedes Versuchstier erhielt täglich während insgesamt 10 Tagen 10 ml Suspension bzw.
10 ml Arabischgummilösung (Kontrollgruppe) pro kg Körpergewicht. Die Tagesdosis wurde zunächst während fünf aufeinanderfolgenden Tagen und dann nach zweitägiger Unterbrechung nochmals während fünf aufeinanderfolgenden Tagen verabreicht. Nach einer Gesamt-Testdauer von zwei Wochen wurden die Versuchstiere einschliesslich derjenigen der Kontrollgruppe getötet und das Blutserum zur Cholesterinanalyse gewonnen. Der Cholesteringehalt des
Blutserums wurde in üblicher Weise (siehe z.B. M. Richterich, Klinische Chemie , Basel 1965, S. 232) bestimmt und die so erhaltenen Daten anhand von entsprechenden Cholesterin- bzw. Triglyceridkonzentration/Dosierungskurven ausgewertet.
In der folgenden Tabelle sind die so ermittelten ED2s Werte der Cholesterinsenkung erfindungsgemässer Verbindungen der Formel (1) mit den entsprechenden therapeutischen Indexwerten (LD5o/ED25) zusammengestellt und mit den Werten der Vergleichssubstanz verglichen.
