5. Verfahren zur Herstellung eines Esters eines D-Homosteroids derJ.Il'ormel I, dadurch gekennzeichnet, dass man nach dem Verfahren von Anspruch 1 ein D-Homosteroid der FormeL \ herstellt und dieses acyliert.
6. Verfahren zur Herstellung eines nieder-Alkyl-, Cyclo alkyl-niederalkyl-, Benzyl-, Furfuryl- oder Tetrahydropyranyläthers ei;nes D-Homosteroids der Formel I, dadurch gekennzeichriet, dass man nach dem Verfahren von Anspruch 1 ein D-Homosteroid der Formel I herstellt und dieses ver äthert.

Der Ausdruck Acyl soll insbesondere organische Säurereste bezeichnen, z. B. Niedere Alkylreste können bis zu 7 C-Atomen enthalten und geradkettig oder verzweigt sein. Beispiele hierfür sind Methyl, Äthyl, Propyl, Isopropyl, Butyl und Isomere davon. Bevorzugte niedere Alkylreste sind Methyl und Äthyl.
Eine bevorzugte Verbindungsgruppe innerhalb der Formel I sind die Verbindungen, in denen R3 Oxo darstellt und der Ring C eine Doppelbindung enthält. Weiterhin sind solche Verbindungen der Formel I bevorzugt, in denen Ri7aa Methyl oder Äthinyl darstellt. Von besonderem Interesse sind das 17a-Hydroxy-D-homo-19-nor-17aa-pregna-4,9,11,16- tetraen-20-in-3-on und das 17ass-Hydroxy-17a-methyl-D-ho- mo-östra-4,9,11,16-tetraen-3-on. Die Reaktion der R17 > -Ketogruppe einer Verbindung der Formel II mit einer metallorganischen Verbindung gemäss obigem Verfahren kann in an sich bekannter Weise durchgeführt werden. Die metallorganische Verbindung kann eine Grignard-Verbindung (z. B. Äthinylmagnesiumbromid, Methylmagnesiumbromid, Vinylmagnesiumbromid) oder eine alkalimetallorganische Verbindung wie Natrium-, Kalium- oder Lithiumacetylid, oder Vinyllithium sein. Eine gleichzeitig anwesende 3-Ketogruppe kann intermidiär z. B.
als Ketal, Enoläther oder Enamin geschützt werden.
Eine 3- und 17a-ständige freie Hydroxygruppe in einem erhaltenen D-Homosteroid-der Formel I kann durch Behandlung mit einem reaktiven Säurederivat z. B. einem Säurehalogenid oder Säureanhydrid in Gegenwart einer Base, wie Pyridin, Collidin oder 4-Diäthylaminopyridin acyliert werden.
Erhaltene D-Homosteroide der Formel I können auch zu nieder-Alkyl-, Cycloalkyl-nieder-alkyl-, Benzyl-, Furfuryloder Tetrahydropyranyläther veräthert werden.
Die Ausgangsmaterialien für die erfindungsgemässe Herstellung der Verbindung der Formel I können, soweit sie nicht bekannt sind oder ihre Herstellung hier beschrieben ist, nach an sich bekannten Methoden bzw. Analogie zu den nachstehend beschriebenen Methoden hergestellt werden.
Die Verbindungen der Formel I sind hormonal wirksam.
Insbesondere zeigen die Verbindungen der Formel I, in denen R17a ungesättigte Reste darstellen, progestative Wirkung. Beispielsweise bewirkt das 17a-Hydroxy-D-homo-l9 nor-17aa-pregna-4,9,11,16-tetraen-20-in-3-on im Clauberg Test bei 50 mg/kg p. o. einen McPhail-Wert von 3,5; bei 20 mg/kg s. c. einen McPhail-Wert von 2,8. Solche Verbindungen sollen z. B. als Ovulationshemmer Verwendung finden. Verbindungen der Formel I in denen Rtvaa nieder-Alkyl bedeuten, können als androgen/anabol wirksame Mittel verwendet werden.
Die Verfahrensprodukte können als Heilmittel z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline, usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees, Suppositorien, Kapseln; oder in flüssiger Form, z. B.
als Lösungen, Suspensionen oder Emulsionen, vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netzoder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
3 g 18-Methyl-D-homoöstra-4,9(10)-16-trien-3,17a-dion wurden in 60 ml Benzol und 12 ml Äthylenglykol in Gegenwart von 20 mg p-Toluolsulfonsäure 7 Stunden am Wasser abscheider erh ltzt. Nach dem Abkühlen wurde das Reaktionsgemisch in etwa 100 ml gesättigte Natriumhydrogencarbonatlösung gegossen und mit Äther extrahiert. Die ätherischen Extrakte wurden über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wurde über 80-g Aluminiumoxyd (neutral; Aktivität III) filtriert (Beniin/Essigester 0-10 /o). Man erhielt 3,05 g eines hellgelben. Öls, [am25-99,40 (in Benzol; c = 0,515).
136 ml Tetrahydrofuran werden durch 30 Minuten langes Einleiten unter Eiskühlung mit Acetylen gesättigt. Danach wurden 34 ml einer 15 0/obigen Lösung von N-Butyllithium in Hexan und 35 mol Tetrahydrofuran bei 0 dazugetropft und weitere 45 Minuten lang Acetylen eingeleitet. Danach wurden 2,7 g 3-Äthylendioxy-18-methyl-D-homoöstra-5(10), 9(11),16-trien-17a-on in 108 ml Tetrahydrofuran tropfenweise zugegeben und das Ganze wurde 2 Stunden unter Eiskühlung gerührt. Zur Aufarbeitung wurde das Reaktionsgemisch tropfenweise mit 40 ml Wasser versetzt und danach in 400 ml Wasser gegossen.
Nach Extraktion mit Äthylacetat und Trocknen der Extrakte über Natriumsulfat und Eindampfen wurden 2,65 g 17aa-Äthinyl-3-äthylendioxy-18- methyl-D-homoöstra-5(10),9(11),16-trien-17ap-ol, Schmelzpunkt 163-1650 (Diisopropyläther/Methylenchlorid) erhalten.
Eine Lösung von 1 g 17aa-9ithinyl-3-äthylendioxy-18- methyl-D-homo-5(10),9(11),16-östratrien-17aa-ol in 50 ml Methanol und 5 ml 2N HC1 wurde 4 Stunden bei 500 unter Argon gerührt. Nach dem Abkühlen wurde das Reaktionsgemisch in 300 ml Wasser gegossen, mit Äthylacetat extrahiert und aufgearbeitet. Man erhielt 890 mg 17aa-Äthinyl- 17aa-hydroxy-18-methyl-D-homoöstra-4,9(10),16-trien-3-on, Schmelzpunkt 194-1960 (Diisopropyläther/Methylenchlorid), [a]r25 - 502,90 (Chloroform; c = 0,505).
Beispiel 2
Zu einer Lösung von 3,0 g 3,3-Diäthoxy-D-homoöstra4,9,11,16-tetraen-17a-on in 100 ml frisch destilliertem Tetrahydrofuran gab man 2,5 g Lithiumacetylid-9ithylendiamin- Komplex und leitete während 1 Stunde Acetylen durch die Lösung. Die Reaktion wurde dann vorsichtig mit Wasser versetzt und mit Ather extrahiert. Das aus dem Äther-Extrakt isolierte Produkt wurde in 60 ml Methanol gelöst, mit einer Lösung von 700 ml Oxalsäure in 20 ml Wasser versetzt und 20 Minuten bei Raumtemperatur gerührt. Die Mischung wurde mit NaHCO3-Lösung neutralisiert, mit Wasser versetzt und mit Äther extrahiert. Nach Verdampfen des Lösungsmittels erhielt man 3,2 g Rohprodukt, das auf 300 g Silicagel chromatographiert wurde.
Mit Essigester-Hexan 1:4 konnte 1,6 g reines 17aa-Äthinyl-17a,B-hydroxy-D-homo- oestra-4,9,11,16-tetraen-3-on eluiert werden, Smp. 186- 1870.
