Abstract

Processes for the preparation of compounds of the formula <IMAGE> in which R is the group <IMAGE> and X is -CH(NH2)- or -CO- or R is the group <IMAGE> and X is -C(NOH)- or -CH(NH2)-. Preparation takes place by cultivation of a Nocardia strain which produces nocardicin C, D, E, F and/or G, preferably Nocardia uniformis subsp. tsuyamanensis ATCC 12301, in an aqueous nutrient medium under aerobic conditions, and isolation of the compounds from the culture broth. The compounds and their salts have an antibiotic action; they can be used for the treatment of infectious diseases.

   PATENTANSPRÜCHE
1. Verfahren zur Herstellung eines der folgenden Nocardieine, nämlich Nocardicin C, Nocardicin D, Nocardicin E, Nocardicin F und Nocardicin G, wobei Nocardicin C der folgenden chemischen Struktur:

 entsprechen, dadurch gekennzeichnet, dass man einen zur Gattung Nocardia gehörenden, Nocardicin C, D, E, F undloder G erzeugenden Stamm in einem wässrigen Nährmedium unter aeroben Bedingungen züchtet und das Nocardicin C, D, E, F oder G daraus gewinnt.



   2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass der Nocardicin C, D, E, F undloder G erzeugende Stamm ein Stamm von Nocardia uniformis subsp. tsuyamanensis ist
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass der Stamm Nocardia uniformis subsp. tsuyamanensis ATCC 21806 ist.



   4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Nocardicin C aus der Kulturbrühe gewinnt.



   5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Nocardicin D aus der Kulturbrühe gewinnt.



   6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Nocardicin E aus der Kulturbrühe gewinnt
7. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Nocardicin F aus der Kulturbrühe gewinnt.



   8. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Nocardicin G aus der Kulturbrühe gewinnt.



   Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen Verbindungen, nämlich Nocardicin C, D, E, F und G, durch Züchtung eines der Gattung Nocardia angehörenden, Nocardicin C, D, E, F undloder G erzeugenden Stammes in wässrigen einem Nährmedium unter aeroben Bedingungen und Isolierung der besagten Nocardicine.



   Die erfindungsgemäss erhältlichen Nocardicin C, D, E, F und G (nachstehend als Nocardicine bezeichnet) entsprechen den folgenden Formeln:

Als erfindungsgemäss zur Anwendung gelangender Mikroorganismus ist ein Stamm vorgesehen, welcher der Gattung Nocardia angehört und befähigt ist, Nocardicin C, D, E, F und/oder G zu erzeugen.



   Unter solchen Stämmen wird der Stamm Nocardia uniformis subsp. tsuyamanensis bevorzugt, welcher am 13. Juni 1972 bei American Type Culture Collection   (ATCC), 12301    Parklawn Drive, Rockville (Maryland 20852, USA) unter der Nummer ATCC 21806 hinterlegt worden ist. Dieser Stamm Nocardia uniformis subsp. tsuyamanensis ATCC 21806 und Ein zelheiten hierüber, z. B. die mikrobiologischen Eigenschaften usw., sind ausführlich publiziert und in der Literatur veröffentlicht worden, z. B. in der amerikanischen Patentschrift Nr.



     3923977    und der deutschen Offenlegungsschrift 2   242699.   



   Dabei ist zu verstehen, dass für die Herstellung der erfindungsgemäss angestrebten Nocardicine die vorliegende Erfindung keineswegs auf die Verwendung des hier beschriebenen spezifischen Organismus eingeschränkt sein soll, da dieser Organismus lediglich zu Erläuterungszwecken angeführt ist.



  Die Erfindung soll sich auch auf die Verwendung von natürlichen Mutanten sowie von künstlich geschaffenen Mutanten beziehen, welche sich von dem hier beschriebenen Mikroorganismus ableiten lassen, z. B. durch Bestrahlung mit Röntgenstrahlen oder Ultraviolettstrahlen, durch Behandlung mit   N'-Nitro-N-nitrosoguanidin, 2-Aminopurin    oder Stickstofflost oder dergleichen.



   Das erfindungsgemässe Verfahren wird mit Vorteil so ausgeführt, dass man einen der Gattung Nocardia, z. B. Nocardia uniformis subsp. tsuyamanensis, angehörenden, Nocardicin C, D, E, F und/oder G erzeugenden Stamm in einem Nährmedium, welches assimilierbare Kohlenstoff- und Stickstoffquellen enthält, unter submersen, aeroben Bedingungen züchtet. Als Nährmedium kann man ein beliebiges verwenden, welches durch den besagten Mikroorganismus für die Herstellung der erfindungsgemäss gewünschten Nocardicine verwendet werden kann. Die bevorzugten Kohlenstoffquellen sind Kohlehydrate, wie Glucose, Saccharose, Maltose, Glycerin, Stärke usw.

  Die bevorzugten Stickstoffquellen sind organische Stickstoffquellen, wie Hefeextrakte, Pepton, Glutenmehl, Baumwollsamenmehl, Sojabohnenmehl, Maisstärke, getrocknete Hefe, Rinderextrakte, Kaseinhydrolysat, Maisquellflüssigkeit, Harnstoff und dergleichen, sowie anorganische Stickstoffquellen, wie Ammoniumsalze, z. B. Ammoniumnitrat, Ammoniumsulfat, Ammoniumphosphat usw.



   Gewünschtenfalls kann man dem Medium Mineralsalze, z. B. Calciumcarbonat, Natrium- oder Kaliumphosphat, Magnesiumchlorid oder -sulfat usw. in kleineren Mengen zugeben.



  Ferner kann man dem Medium auch eine oder mehrere organische Verbindungen, wie Tyrosin, Glycin, Serin, Homoserin, p-Hydroxyphenylglycin, a-Aminobuttersäure,   a,p-Diaminopro-    pionsäure, N-Acetyltyrosin, N-Acetyltyrosinamid, p-Hydroxy   phenylbrenztraubensäure,    p-Hydroxyphenylglycolsäure, p-Hydroxyphenylglyoxalsäure,   Shikiminsäure, 2-Amino-3-(4-       hydroxyphenyl)-propionohydroxaminsäure, 2-Acetamido-3-(4    hydroxyphenyl)-propionohydrazid und dergleichen, hinzugeben. Diese organischen Verbindungen können als eine Art Vor  läufer wirken und können für die Steigerung der Ausbeute an Nocardicinen dienlich sein.



   Für die Herstellung der erwünschten Nocardicine in grossen Mengen bedient man sich mit Vorteil des Fermentierverfahrens unter submersen aeroben Züchtungsbedingungen. Für die Herstellung von begrenzten Mengen kann man auch eine Schüttelkultur oder Oberflächenkultur in einem Kolben oder in einer Flasche anwenden. Erfolgt das Wachstum in grossen Behältern, so verwendet man vorzugsweise die vegetative Form des Organismus für die Impfung in den Produktionsbehältern, um eine Wachstumsverzögerung bei der Herstellungsmethode der Nocardicine zu verhindern. Damit ist es wünschenswert, zuerst ein vegetatives Inokulum des Organismus zu erzeugen, indem man eine relativ kleine Kulturmediummenge mit Sporen oder Mycelien des Organismus impft und hierauf die Züchtung vornimmt. Der Transfer des gezüchteten vegetativen Inokulums in die grossen Behälter erfolgt dann aseptisch.

  Das Medium, in welchem das vegetative Inokulum erzeugt wird, kann praktisch das gleiche Medium sein wie jenes, welches für die Herstellung der Nocardicine verwendet wird, oder es kann auch ein davon verschiedenes Medium sein.



   Das Rühren und Belüften des Züchtungsgemisches kann in verschiedener Weise bewirkt werden. Das Rühren kann beispielsweise mit einem Propeller oder mit einer ähnlichen, mechanischen Rührausrüstung geschehen, indem der Fermentierbehälter gedreht oder geschüttelt wird oder indem man sich verschiedenartiger Pumpsysteme bedient oder aber sterile Luft durch das Medium leitet. Die Belüftung kann durch Einleiten von steriler Luft in das Fermentiergemisch erfolgen. Im Verlaufe der Fermentierung kann man dem Medium insbesondere in jenen Fällen, in welchen das Kulturmedium in beachtenswer ter Weise schäumt, ein Entschäumungsmittel, z. B. Pflanzenöle, wie Sojabohnenöl usw., höhere Alkohole, wie Octadecanol,
Tetradecanol usw., Silikone usw., zugeben.



   Die Fermentierung erfolgt gewöhnlich bei Temperaturen im Bereiche von ungefähr 20 bis 40   "C    und vorzugsweise bei ungefähr 30   "C    während einer Zeitdauer von 50 bis 100   Stunde    den.



   Die erfindungsgemäss erhältlichen Nocardicine lassen sich in bekannter Weise aus der Kulturbrühe gewinnen. Sie sind in der Kulturbrühe in den Mycelien (intracellular) und/oder ausserhalb der Myceline (extracellular) vorhanden.



   Als erste Stufe kann die Kulturbrühe in das Filtrat (überstehende Flüssigkeit) und einen Filterkuchen entweder durch Filtrieren oder durch Zentrifugieren getrennt werden.



   Die Extraktion der Nocardicine aus dem Filterkuchen erfolgt durch Behandeln dieses Kuchens mit einem organischen Lösungsmittel, in welchem die Nocardicine löslich sind, wie z. B. Alkoholen, z. B. Methanol, usw., Ketonen, z. B. Aceton usw., wässrigen Alkoholen, Äthanol, z. B. wässrigem Methanol, wässrigem Äthanol usw.



   Aus dem so erhaltenen Filtrat und/oder Extrakt lassen sich die Nocardicine isolieren und in bekannter Weise reinigen.



   Zu diesem Zwecke kann man beispielsweise mit Adsorptionsmitteln, z. B. Aktivkohle, Kieselsäure, Kiesegel, Aluminiumoxyd usw., anionischen oder kationischen Austauschharzen oder makroporösen, nicht-ionischen Adsorptionsharzen [z. B. Amberlite XAD-2, XAD4, XAD-7 und XAD-8 (Warenzeichen der Firma Rohm  & Haas   Co.)J,    behandeln. Man kann auch mit Lösungsmitteln extrahieren, unter vermindertem Druck einengen, lyophilisieren, den pH-Wert einstellen, eine Kristallisierung vornehmen, umkristallisieren und dergleichen. Alle diese Mittel kann man vorzugsweise unabhängig voneinander oder in Kombination miteinander in beliebiger und gewünschter Reihenfolge oder wiederholt anwenden.

 

   Erhält man ein Rohmaterial, z. B. das Kulturfiltrat und Extrakte aus dem Filterkuchen, welches sämtliche Nocardicine enthält, d. h. Nocardicin A, Nocardicin B, welches ein geometrisches Isomer von Nocardicin A an der Hydroxyiminogruppe darstellt, Nocardicin C, Nocardicin D, Nocardicin E, Nocardicin F und Nocardicin G, so kann man ein jedes dieser Nocardicine beispielsweise nach dem folgenden Schema isolieren:

Trennung von Nocardicin C und G von Nocardicin A, B, D,   E und F:   
Wird das besagte Rohmaterial unter Verwendung eines makroporösen, nicht-ionischen Adsorptionsharzes, z. B. Diaion HP 20, der Säulenchromatographie unterworfen, so werden Nocardicin A, B, D, E und F auf dem Harz adsorbiert. Andererseits gehen Nocardicin C und G durch die Säule hindurch.



  Stufe b)
Trennung von Nocardicin C von Nocardicin G:
Die nach den obigen Angaben durch die Säule   hindurchge-    flossene Lösung wird hierauf fraktioniert, indem man sie einer Säulenchromatographie unterwirft, wobei man Cellulose mit  einem geeigneten Entwicklerlösungsmittel, wie mit Wasser gesättigtem n-Butanol oder einem Gemisch von n-Butanol, Essigsäure und Wasser im Mischungsverhältnis 20 : 1 : 6, verwendet, wodurch man zu der Fraktion I, welche Nocardicin C enthält, und zur Fraktion   II,    welche Nocardicin G enthält, gelangt.



   Stufe c)
Trennung von Nocardicin D von Nocardicin A, B, E und F:
Das Adsorbat aus der vorangehenden Stufe a) wird mit einem hydrophilen Lösungsmittel,   z    B. wässrigem Methanol, aus dem Harz eluiert. Das Nocardicin A, B, D, E und F enthal tende Eluat wird hierauf in drei Fraktionen, d. h. Fraktion III, welche Nocardicin E und F enthält, Fraktion IV, welche   Nocar-    dicin D enthält, und Fraktion V, welche Nocardicin A und B   enthält.    durch Säulenchromatographie über Cellulose mit einem geeigneten Entwicklerlösungsmittel, wie mit Wasser gesättigtem n-Butanol oder einer oberen Schicht einer Mischung von Äthylacetat, n-Butanol und Wasser im Mischungsverhältnis von 5 :5 :2. fraktioniert.



   Stufe d)
Trennung von Nocardicin E von Nocardicin F:
Die gemäss vorangehender Stufe c) erhaltene Fraktion III wird über Kieselsäure mit einem geeigneten   Entwicklerlö-    sungsmittel. z. B. einem   Gemisch    von Chloroform und Äthylacetat im Mischungsverhältnis von   4:1    oder einem Gemisch von Chloroform und Methanol im Mischungsverhältnis von 10 : 1 durch Säulenchromatographic einer weiteren Fraktionierung unterworfen, wobei man zur Fraktion VI, welche Nocardicin E enthält, und zur Fraktion   VII.    welche Nocardicin F enthält, gelangt.



  Stufe e)
Alternatimethode für die Trennung von Nocardicin E und Nocardicin F:
Nocardicin E und F lassen sich aus dem Rohmaterial auch durch Säulenchromatographie über Kieselsäure mit dem gleichen geeigneten Entwicklerlösungsmittel, wie es oben bei spielsweise angegeben worden ist, trennen, da Nocardicin A, B,
C, D und G durch die Kieselsäure stark adsorbiert werden und sich mit dem besagten Lösungsmittel praktisch nicht eluieren lassen.



   Jedes Nocardicin C, D, E, Fund G lässt sich aus den Fraktio nen, d. h. Fraktionen I, II, IV, VI und   VII,    in üblicher Weise reini gen.



   Die in der Kulturbrühe erzeugten Nocardicine können in freier Form isoliert werden; sie können auch in Form ihrer Alkalimetallsalze erhalten werden, indem man das die Nocardicine enthaltende Rohmaterial mit einem Alkali, z. B. Natriumoder Kaliumhydroxyd. während der Isolierung oder der Reinigung behandelt.



   Die in freier Form erhältlichen Nocardicine können auch mit einer anorganischen oder einer organischen Base, z. B.



   Kalium- oder Natriumhydroxyd, Äthanolamin, Dicyclohexyla min usw., in an sich bekannter Weise in ein Salz überführt wer den.



   Andererseits lassen sich die unter Zuhilfenahme von anorganischen oder organischen Basen erhaltenen Salze der Nocardicine auch   leicht ion    die freie Form überführen, indem man solche Salze mit einer Säure, z. B. einer Mineralsäure, wie Salzsäure usw., in an sich bekannter Weise behandelt.



   Die physikochemischen Eigenschaften der neuen Nocardicine finden sich nachstehend.

   broad s)    11,14(1    H,s) Aufgrund von weiteren Studien liessen sich die chemischen Strukturen der erfindungsgemäss erhältlichen Nocardicine, welche die obigen physikochemischen Eigenschaften aufweisen, identifizieren.



   Die erfindungsgemäss erhältlichen Nocardicine und deren Salze besitzen spezifische antibiotische Spektren, wobei sie eine Wirkung gegen pathogene Bakterien bei niedriger Toxizität besitzen. Daher sind die erfindungsgemäss erhältlichen Nocardicine und deren Salze für die Behandlung von Infektionskrankheiten, welche durch solche Bakterien bei Mensch und Tier verursacht werden, wertvoll.



   Die pharmakologischen Teste einiger der erfindungsgemäss erhältlichen Nocardicine, d. h. antimikrobielle Teste und Toxizitätsteste, finden sich nachstehend.



  Minimale Hemmkonzentration(M.I.C.)
Der M.l.C.-Test wurde durchgeführt unter Anwendung einer üblichen Agarverdünnungsmethode unter Verwendung von Herzinfusionsagar (oder Mueller Hinton-Agar), welches während 20 Stunden bei 37  C inkubiert wurde. M.I.C.-Wert wird als die minimale Konzentration von Nocardicin C, D, E oder F   (mcglml)    ausgedrückt, welche das Wachstum des Mikroorganismus verhindert. Die Resultate finden sich in der folgenden Tabelle 6.

Eine wässrige Natriumhydroxydlösung von Nocardicin C mit einem pH-Wert von 7,4 wurde intravenös 

   in jeweils fünf männlichen Mäusen vom ICR-Stamm und einem Gewicht von 18 bis 22 g (Dosis: 1 g/kg) injiziert, worauf man während 1 Woche nach der Verabreichung die Tiere beobachtete. Es ergab sich dabei, dass sämtliche getesteten Mäuse sich   wäh-    rend dieser Periode normal verhielten.



   Ferner wurden jeweils wässrige Natriumhydroxydlösungen von Nocardicin D, E, F und G mit einem pH-Wert von 7,4 jeweils fünf Mäusen vom ICR-Stamm mit einem Gewicht von 18 bis 22 g (Dosis: 500 mg/kg) subkutan injiziert, und dann nach Ablauf von einer Woche nach Verabreichung wurden die Tiere beobachtet. Es ergab sich dabei, dass sich sämtliche getesteten Tiere während der ganzen Dauer normal verhielten.



   Die neuen Nocardicine und die pharmazeutisch zulässigen Salze derselben lassen sich für die Verabreichung in beliebiger Weise formulieren, und zwar wie dies normalerweise bei bekannten Antibiotika der Fall ist, indem man ihnen einen pharmazeutischen Träger beimischt.



   Pharmazeutisch zulässige Salze der erfindungsgemäss erhältlichen Nocardicine umfassen Salze mit anorganischen oder organischen Basen, z. B. Natriumhydroxyd, Kaliumhydroxyd, Calciumhydroxyd, Ammoniak, Äthanolamin, Triäthylamin, Dicyclohexylamin und dergleichen.



   Somit lassen sich die antimikrobiellen Präparate in Form von pharmazeutischen Formen, z. B. in fester, halbfester oder flüssiger Form, verwenden, wobei die Wirksubstanz mit einem pharmazeutischen, organischen oder anorganischen Trägermittel oder Füllstoff, welcher für die äussere, enterale oder parenterale Verabreichung geeignet ist, vermischt wird. Die Wirksubstanz kann beispielsweise mit üblichen Trägermitteln, wie sie für Tabletten, Kügelchen, Kapseln, Suppositorien, Lösungen, Emulsionen, Suspensionen usw. verwendet werden, compoundiert werden. Auch andere Formen mögen sich eig nen.

  Als verwendbare Trägermittel kommen Wasser, Glucose
Lactose, Akaziengummi, Gelatine, Mannitol, Stärkepaste,
Magnesiumtrisilikat, Kalk, Maisstärke, Keratin, kolloidales Ki selgel, Kartoffelstärke, Harnstoff und andere Trägermittel, welche sich für die Herstellung solcher Präparate in fester,   halbfester    oder flüssiger Form eignen, in Frage. Solche Präparate können überdies Hilfsstoffe, Stabilisiermittel, Verdünnungsmittel und Färbemittel sowie Riechstoffe enthalten. Die antimikrobiellen Präparate können auch Schutzmittel oder bakteriostatische Mittel enthalten, um auf diese Weise die Wirksubstanz in den gewünschten Präparaten aktiv zu halten.



  Die erfindungsgemäss erhältlichen Nocardicine oder deren Salze werden in einer solchen Menge in die antimikrobiellen Präparate eingearbeitet, welche ausreicht, um die gewünschte therapeutische Wirkung auf den bakteriell infizierten Prozess oder Zustand zu gewährleisten.



   Will man die in Frage kommenden antimikrobiellen Präpa rate am Menschen anwenden, so wird man dies vorzugsweise in Form von intravenösen oder intramuskulären Injektionen tun. Dabei schwankt die Dosierung oder die therapeutisch wirksame Menge der neuen Nocardicine und deren Salze je nach Alter und Bedingung des einzelnen, zu behandelnden Patienten. Tägliche Dosierungen von ungefähr 2 bis 50 mg Wirksubstanz/kg Körpergewicht bei Mensch oder Tier werde allgemein für die Behandlung von Erkrankungen verwendet.



   Die Nocardicine können auch als Zwischenprodukte für di Herstellung der anderen   3-Acylamino-1  < a-carboxy-t-hydroxy      benzyl > 2-azetidinone    mit besserer antimikrobieller Wirkung gegen pathogene, grampositive Bakterien verwendet werden.



  Unter Bezugnahme auf einige der erfindungsgemäss erhältlichen Nocardicine sei die Herstellung von   3-Acylamino-lXa-ca      boxy-4-hydroxybenzylS2-azetidionen    aus den erfindungsgemä sen Nocardicinen mit dem folgenden Schema erläutert.

   Die folgenden Beispiele erläutern die vorliegende Erfindung.



  Beispiel 1 Herstellung von Nocardicin C
Ein wässriges Medium (100 ml), welches 2%   Saccarose, 2%      Baumwollsamenmehl, 1%    getrocknete   Hefe,2,18%    KH2PO4 und   1,43%      Na2HPO4.12H20    enthält, wurde jeweils in 20 500-ml Schüttelkolben gegossen und während 20 Minuten bei 120   "C    sterilisiert. Dann wurde jedes Medium mit einer Öse voll Schrägkultur von Nocardia uniformis subsp. tsuyamanensis ATCC 21806 geimpft und hierauf während 48 Stunden bei 30   "C    gezüchtet.



   Andererseits wurde ein anderes wässriges Medium (100 Liter), welches 2% lösliche   Stärke, 1%    Pepton,   0,4%    Hefeex   trakte, 1%      KH2PO4,1%      Na2HPO4-      12H20,0,5%      MgSO-    7H20,   0.1%    L-Tyrosin und 0,1% Glycin enthält, durch Zugabe einer wässrigen 6n-Natriumhydroxydlösung auf einen pH-Wert von 6,0 eingestellt. 20 Liter des Mediums wurden in jeweils fünf 30-Liter-Fermentierbehälter gegossen und während 20 Minuten sterilisiert.



   Jedes dieser Medien wurde mit der nach den obigen Angaben erhaltenen Samenkultur versetzt, und zwar in einer Menge von 2 Volumen-% Medium. Der Organismus wurde während 96 Stunden bei 30   "C    wachsen gelassen. Während dieser Wachstumsperiode wurde die Brühe mit 250 Umdrehungen pro Minute gerührt und durch Einblasen von steriler Luft in einer Menge von 20 Litern pro Minute behandelt.



   Die so erhaltene Kulturbrühe (80 Liter) wurde mit Hilfe von Diatomeenerde (8 kg) filtriert, wobei man einen Filterkuchen und ein Filtrat erhielt. Der Filterkuchen wurde mit 70%igem wässrigem   Äthanol (20    Liter) versetzt und das Gemisch hierauf während 1 Stunde gerührt und filtriert. Diese Extraktion wurde zweimal wiederholt. Die vereinigten Extrakte (40 Liter) wurden auf ein Volumen von 2 Litern unter vermindertem Druck eingeengt.



   Andererseits wurde das nach den obigen Angaben erhaltene Kulturfiltrat auf ein Volumen von 10 Litern eingeengt.



  Dieses Konzentrat wurde hierauf mit 50 Liter Methanol unter Rühren versetzt, worauf man das Gemisch während 1 Stunde stehen liess und anschliessend filtrierte. Das Filtrat wurde unter vermindertem Druck auf ein Volumen von 2 Litern eingeengt.



   Die vereinigten Konzentrate wurden durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 3,0 eingestellt und zweimal mit Äthylacetat (4 Liter) extrahiert. Die wässrige Schicht wurde unter vermindertem Druck auf ein Volumen von 1,8 Liter eingeengt. Das Konzentrat wurde mittels wässriger   1 n-Natriumhydroxydlösung    auf einen pH-Wert von 4,0 eingestellt und hierauf durch eine mit Diaion HP20 (4 Liter) beschickte Säule hindurchgeleitet, um auf diese Weise eine ausfliessende Lösung zu erhalten. Anschliessend wurde die Säule mit Wasser gewaschen, wobei man Waschflüssigkeiten erhielt Die durch die Säule geflossene Lösung und die Waschflüssigkeiten wurden vereinigt und die wässrige Lösung (6 Liter) durch Zugabe einer wässrigen 6n-Natriumhydroxydlösung auf einen pH-Wert von 8,0 eingestellt.

  Dann wurde diese Lösung mit 120 g Aktivkohle versetzt, worauf man das Gemisch während 10 Minuten rührte und hierauf filtrierte. Die Aktivkohle wurde mit 1 Liter Wasser ausgewaschen, worauf man 80%iges Methanol (1 Liter) hinzugab.



   Das Gemisch wurde während 10 Minuten gerührt und filtriert. Der Eluierungsvorgang wurde einmal mehr wiederholt.



  Die vereinigten Eluate (2 Liter) wurden auf ein Volumen von 10 ml eingeengt. Dieses Konzentrat wurde hierauf mit 200 ml Methanol unter Rühren versetzt, worauf die unlöslichen Bestandteile aus dem Gemisch abfiltriert wurden. Das Filtrat wurde auf ein Volumen von 10 ml eingeengt. Das Konzentrat wurde dann einer Säulenchromatographie über Cellulose (Ent   wicklerlösungsmittel:    obere Schicht einer Mischung von n-Butanol, Essigsäure und Wasser im Mischungsverhältnis von 100 :5 :30) unterworfen. Das Eluat (500 ml), welches die gewünschte Verbindung enthielt, wurde gesammelt. Das Eluat wurde hierauf mit 500 ml n-Hexan versetzt und das Gemisch gerührt und hierauf die wässrige Schicht abgetrennt. Zu dieser wässrigen Schicht gab man 100 ml Äthylacetat und rührte dann das Gemisch. Die wässrige Schicht wurde hierauf abgetrennt und auf ein Volumen von 20 ml eingeengt.

  Dieses Konzentrat wurde mit 40 ml Aceton versetzt und das Gemisch in einem Kühlschrank (5   "C)    stehen gelassen, wobei sich Kristalle bildeten. Das Gemisch wurde filtriert, und die erhaltenen Kristalle wurden getrocknet, wobei man 26 mg Nocardicin C in Form von rohen Kristallen erhielt. Diese rohen Kristalle wurden in 4 ml Wasser gelöst und die Lösung mit 8 ml Aceton versetzt.

 

  Das Gemisch wurde dann stehen gelassen, wobei sich Kristalle ausschieden. Diese wurden abgetrennt und getrocknet, wobei man Nocardicin C (15 mg) in Form von weissen Kristallen erhielt.



  Beispiel 2 Herstellung von Nocardicin D
Ein wässriges Medium (100 ml), welches 2%   Saccharose, 2%      Baumwollsamenmehl, 1%    getrocknete   Hefe, 2,18%    KH2PO4   ond      1 > 43%      Na2HPO4-    12H20 enthielt, wurde in jeweils 20 500-ml-Schüttelkolben eingetragen und während 20 Minuten  bei 120   "C    sterilisiert. Dann wurde eine Öse voll Schrägkultur von Nocardia uniformis subsp. tsuyamanensis ATCC 21806 zwecks Impfung in jedes dieser Medien eingetragen, worauf die Züchtung während 48 Stunden bei 30   "C    durchgeführt wurde.



   Andererseits wurde ein wässriges Medium (100 Liter), welches 2% lösliche   Stärke, 1%      Pepton, 0,4%      Hefeextrakte, 1%      KH2PO4,1 1% Na2HPO4 - 12H20,0,5% MgSO - 7H2O, 0,1%    L-Tyrosin und 0,1% Glycin enthielt, durch Zugabe von wässriger 6n-Natriumhydroxydlösung auf einen pH-Wert von 6,0 eingestellt, worauf 20 Liter dieses Mediums in jeweils fünf Fermentierbehälter mit einem Fassungsvermögen von jeweils 30 Litern gegossen und während 20 Minuten bei 120   "C    sterilisiert
Ein jedes dieser Medien wurde mit der Samenkultur, wie sie oben erhalten worden war, in einer Menge von 2 Volumen-% des Mediums versetzt. Der Organismus wurde während 96 Stunden bei 30    C    wachsen gelassen.

  Während der Wachstumsperiode wurde die Kulturbrühe mit 250 Umdrehungen pro Minute gerührt, wobei man gleichzeitig sterile Luft in einer Menge von 20 Litern pro Minute durchleitete.



   80 Liter der so erhaltenen Kulturbrühe wurden dann mit 8 kg Diatomeenerde versetzt und das Gemisch anschliessend filtriert, wobei man zu einem Filtrat und einem Filterkuchen gelangte. Das   Filtrat (70    Liter) wurde unter vermindertem Druck auf ein Volumen von 7 Liter eingeengt. Das Konzentrat wurde dann unter Rühren mit 90 Liter Methanol versetzt und das Gemisch anschliessend während 1 Stunde stehen gelassen und filtriert, um den Niederschlag zu entfernen. Das Filtrat (80 Liter) wurde auf ein Volumen von 5 Liter eingeengt. Dann wurde das Konzentrat mit 15 Litern Wasser und 1 kg Aktivkohle versetzt.



   Das Gemisch wurde während 10 Minuten gerührt und filtriert, um die Aktivkohle zu gewinnen. Diese Aktivkohle wurde hierauf mit 10 Litern einer 80%igen wässrigen Methanollösung versetzt, worauf man den pH-Wert des Gemisches durch Zugabe von   5%iger    wässriger Ammoniaklösung auf einen pH-Wert von 8,0 einstellte und das Gemisch während 10 Minuten rührte und hierauf filtrierte. Diese Eluierung wurde ein zweites Mal durchgeführt. Die vereinigten Filtrate wurde unter vermindertem Druck auf ein Volumen von 2 Litern eingeengt.



  Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 4,0 eingestellt und dann durch eine mit Diaion HP20 (4 Liter) beschickten Säule geleitet. Diese Säule wurde mit 4 Litern Wasser gewaschen, worauf man die gewünschte Verbindung mit   50%igem    wässrigem Äthanol eluierte. Das Eluat (1,5 Liter), welches die gewünschte Verbindung enthielt, wurde gesammelt und unter vermindertem Druck auf ein Volumen von 100 ml eingeengt. Das Konzentrat wurde dann über Cellulose (Entwicklerlösungsmittel: mit Wasser gesättigtes n-Butanol) der Säulenchromatographie unterworfen. Das die gewünschte Verbindung enthaltende Eluat (200 ml) wurde gesammelt und unter vermindertem Druck auf ein Volumen von 10 ml eingeengt.

  Das Konzentrat wurde hierauf über Cellulose (Entwicklerlösungsmittel: obere Schicht einer Mischung von Äthylacetat, n-Butanol und Wasser im    Mischungsverhältnis von 5 :5 :2) der Säulenchromatographie    unterworfen. Das Eluat (100 ml), welches die gewünschte Verbindung enthielt, wurde unter vermindertem Druck auf ein Volumen von 10 ml eingeengt, wobei ein Niederschlag gebildet wurde, welcher durch Filtrieren und anschliessendes Trocknen gewonnen wurde. Der Niederschlag wurde in 5 ml Wasser suspendiert, worauf man die Suspension durch Zugabe einer wässrigen In-Natriumhydroxydlösung auf einen pH-Wert von 8,0 einstellte, um den Niederschlag zu lösen.

  Die Lösung wurde durch Zugabe von 1 n-Salzsäure auf einen pH-Wert von 3,0 angesäuert und dann über Nacht in einem Kühlschrank (5   "C)    stehen gelassen, wobei sich Kristalle bildeten, die man hierauf durch Filtrieren isolierte. Diese Kristalle wurden hierauf getrocknet, wobei man 50 mg Nocardicin D in Form von weissen Kristallen erhielt
Der nach den obigen Angaben erhaltene, die Mycelien enthaltende Filterkuchen wurde zweimal mit 70%igem wässrigem   Äthanol (20    Liter) extrahiert. Die vereinigten Extrakte wurden unter vermindertem Druck auf ein Volumen von 2 Litern eingeengt. Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 3,0 eingestellt und dreimal mit Äthylacetat (2 Liter) extrahiert, um die Verunreinigungen zu entfernen. Die wässrige Schicht wurde abgetrennt und unter vermindertem Druck eingeengt.

  Das Konzentrat wurde mittels wässriger 6n-Natriumhydroxydlösung auf einen pH-Wert von 4,0 eingestellt und dann durch eine mit Diaion HP20 (2 Liter) beschickten Säule geleitet. Die Säule wurde mit Wasser gewaschen, worauf die gewünschte Verbindung mit 50%igem Äthanol (1,5 Liter) eluiert wurde. Das   Eluat (1,5    Liter) wurde unter vermindertem Druck auf ein Volumen von 100 ml eingeengt.



  Der so erhaltene Rückstand wurde über Cellulose (Entwicklerlösungsmittel: mit Wasser gesättigtes n-Butanol) der Säulenchromatographie unterworfen. Das die gewünschte Verbindung enthaltende Eluat wurde gesammelt und unter vermindertem Druck eingeengt. Der erhaltene Rückstand wurde erneut über Cellulose (Entwicklerlösungsmittel: obere Schicht einer Mischung von Äthylacetat, n-Butanol und Wasser im Mischungsverhältnis von 5 :5   :2) der    Säulenchromatographie unterworfen. Das die gewünschte Verbindung enthaltende Eluat wurde unter vermindertem Druck auf ein Volumen von 10 ml eingeengt, wobei man zu einem Niederschlag gelangte, welcher durch Filtrieren abgetrennt und hierauf getrocknet wurde.

  Der Niederschlag wurde in 2 ml Wasser suspendiert und diese Suspension durch Zugabe einer wässrigen   1 n-Natri-    umhydroxydlösung auf einen pH-Wert von 8,0 eingestellt, wodurch der Niederschlag gelöst wurde. Hierauf wurde die Lösung durch Zugabe von   1 n-Salzsäure    auf einen pH-Wert von 3,0 eingestellt und über Nacht in einem Kühlschrank (5   "C)    stehen gelassen, wobei sich Kristalle bildeten, welche durch Filtrieren abgetrennt und getrocknet wurden. Auf diese Weise erhielt man 10 mg Nocardicin D in Form von weissen Kristallen.



  Beispiel 3 Herstellung von Nocardicin E
Ein wässriges Medium (300 ml), welches 2% Saccharose, 2% Baumwollsamenmehl, 1% getrocknete Hefe, 2,18% KH2PO4    und 1,43% NaHPO'- zu 1 2H20 enthielt, wurde in einen 1Liter    Erlenmeyerkolben eingetragen und während 20 Minuten bei 120   "C    sterilisiert. Dann wurde mit einer Öse voll Schrägkultur von Nocardia uniformis subsp. tsuyamanensis ATCC 21806 geimpft und hierauf während 54 Stunden bei 30   "C    gezüchtet.



   Ein 500-Liter-Fermentierbehälter wurde mit dem gleichen Medium (150 Liter), wie es oben erwähnt worden ist, beschickt.



  Das Fermentiermedium wurde während 20 Minuten bei 120    C    sterilisiert und hierauf mit dem gesamten Volumen an vegetativem Inokulum, wie es nach den obigen Angaben erhalten worden ist, inokuliert und während 42 Stunden bei 30   "C    gezüchtet.

 

   Andererseits wurde ein wässriges Medium (3000 Liter), welches 2% lösliche Stärke, 1% Pepton, 0,4% Hefeextrakte, 1% KH2PO4, 1%   Na2HPO4 1 2H20,    0,5%   MgSO -    7H20, 0,1% L-Tyrosin und 0,1% Glycin enthielt, in einen 4000-Liter-Fermentierbehälter eingetragen und darin während 20 Minuten bei 120   OC    sterilisiert. Dann wurde das Medium mit dem gesamten Volumen der nach den obigen Angaben erhaltenen Kulturbrühe geimpft.



   Der Organismus wurde im Fermentiermedium während 119 Stunden bei 30   C    wachsen gelassen. Während dieser Wachstumsperiode wurde die Kulturbrühe mit 230 Umdrehungen pro Minute gerührt, wobei man gleichzeitig sterile Luft durch die Brühe in einer Menge von 3000 Litern pro Minute  hindurchleitete. Nach beendeter Züchtung wurde die Kulturbrühe mit Hilfe von Diatomeenerde (180 kg) filtriert. Zu einem Teil des Filtrats (1500 Liter) wurden 45 kg Aktivkohle zugegeben, worauf das Gemisch während 30 Minuten gerührt und filtriert wurde, um die Aktivkohle abzutrennen.

  Die Aktivkohle wurde hierauf mit einer Mischung von Aceton und Wasser im Mischungsverhältnis 7 :3 (200 Liter) versetzt, worauf das erhaltene Gemisch durch Zugabe von wässrigem Ammoniak auf einen pH-Wert von 8,0 eingestellt und während 60 Minuten gerührt wurde. 500 Liter des so erhaltenen Eluats wurden auf ein Volumen von 50 Liter eingeengt. Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 4,0 angesäuert. Anschliessend wurde das Konzentrat mit 60 Litern n-Butanol versetzt, worauf man das Gemisch während 30 Minuten rührte. Die n-Butanolschicht (50 Liter) wurde abgetrennt und unter vermindertem Druck auf ein Volumen von 10 Liter eingeengt. Das Konzentrat wurde mittels   5%igem    wässrigem Ammoniak auf einen pH-Wert von 8,0 eingestellt und während 30 Minuten gerührt.

  Dann wurde die wässrige Schicht (15 Liter) abgetrennt und unter vermindertem Druck auf ein Volumen von 1,3 Litern eingeengt. Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 4,0 eingestellt, mit 1,3 kg Diatomeenerde vermischt und hierauf getrocknet.



  Das Pulver wurde mit 3 Liter Chloroform gewaschen und die gewünschte Verbindung mit Äthylacetat eluiert. Das Eluat (5 Liter) wurde unter vermindertem Druck auf ein Volumen von 50 ml eingeengt. Dann wurde das Konzentrat mit 50 g Diatomeenerde vermischt und getrocknet. Das Pulver wurde über Kieseläure (Entwicklerlösungsmittel: Äthylacetat) der Säurenchromatographie unterworfen. Das Eluat (600 ml) wurde unter vermindertem Druck auf ein Volumen von 10 ml eingeengt. Das Konzentrat wurde mit 10 g Diatomeenerde vermischt und getrocknet. Das Pulver wurde anschliessend über Kieselsäure (Entwicklerlösungsmittel: Mischung von Chloroform und Methanol im Mischungsverhältnis von 100 :7) der Säulenchromatographie unterworfen. Das Eluat (200 ml) wurde unter vermindertem Druck auf ein Volumen von 3 ml eingeengt. Hierauf wurde das Konzentrat mit 3 g Diatomeenerde vermischt und getrocknet.

  Das Pulver wurde über Kieselsäure (Entwicklerlösungsmittel: eine Mischung von Chloroform und Äthylacetat im Mischungsverhältnis von   1    der Säulenchromatographie unterworfen. Das Eluat (50 ml) wurde unter vermindertem Druck zur Trockne eingedampft. Der so erhaltene Rückstand wurde in 1 ml Methanol gelöst. Diese
Lösung wurde anschliessend mit 5 ml Chloroform versetzt und bei 4   C    über Nacht stehen gelassen, wobei sich 230 mg Kristalle bildeten, welche man aus einer Mischung von Methanol und Chloroform   (1:5) (12    ml) umkristallisierte. Auf diese Weise erhielt man Nocardicin Ein einer Menge von 190 mg in Form von weissen Kristallen.



  Beispiel 4 Herstellung von Nocardicin F
Ein wässriges Medium (100 ml), welches 2%   Saccharose, 2%      Baumwollsamenmehl, 1%    getrocknete   Hefe, 2,18%    KH2PO4   und 1,43% Na2HPO4 - 12H20 enthielt, wurde in jeweils 24    500-ml-Sakaguchikolben gegossen und während 20 Minuten bei
120   "C    sterilisiert. Dann wurde mit einer Öse voll Schrägkultur von Nocardia uniformis subsp. tsuyamanensis ATCC 21806 geimpft und der Organismus während 48 Stunden bei 30   "C    gezüchtet.



   Andererseits wurde ein wässriges Medium (20 Liter), welches 2% lösliche   Stärke, 1%      Pepton,0,4%      Hefeextrakt, 1%       KH2PO4, 1% Na2HPO4- 12H2O, 0,5% MgSO- 7H20,0,1%   
L-Tyrosin und 0,1% Glycin enthielt in jeweils sechs 30-Liter-Fermentierbehälter gegossen und darin während 20 Minuten bei
120   "C    sterilisiert. Hierauf wird die nach den obigen Angaben erhaltene Kulturbrühe zur Impfung in jedes der Medien in einem Verhältnis von 2 Volumen-% des Mediums hinzugegeben. Der Organismus wurde dann während 96 Stunden bei 30   "C    im Medium wachsen gelassen. Während der Wachstumsperiode wurde die Brühe mit 250 Umdrehungen pro Minute gerührt und sterile Luft gleichzeitig mit einer Geschwindigkeit von 20 Liter pro Minute in die Brühe eingeleitet.

  Nach beendeter Züchtung wurde die Kulturbrühe (100 Liter) durch Diatomeenerde   kg)    filtriert. Das Filtrat (90 Liter) wurde hierauf mit 1,5 kg Aktivkohle versetzt, worauf man das Gemisch   wäh-    rend 10 Minuten rührte und hierauf filtriere. Die Aktivkohle wurde zweimal mit Wasser (10 Liter) gewaschen und die gewünschte Verbindung zweimal mit 80%igem wässrigem Methanol (30 Liter bzw. 20 Liter) eluiert. Das Eluat (50 Liter) wurde unter vermindertem Druck auf ein Volumen von 6 Liter eingeengt. Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 4,0 eingestellt und dann durch eine mit Diaion   HP20 (3    Liter) beschickte Säule geleitet. Nach dem Waschen der Säule mit 1 Liter Wasser wurde die gewünschte Verbindung mit 40%igem Äthanol (8 Liter) eluiert.



  Die Fraktionen (4 Liter), welche die gewünschte Verbindung enthielten, wurden gesammelt und unter vermindertem Druck auf ein Volumen von 100 ml eingeengt. Das Konzentrat wurde mit 300 g pulvriger Cellulose gemischt, worauf man das Gemisch unter vermindertem Druck trocknete. Das getrocknete Pulver wurde einer Säule zugeführt und darin mit Aceton (6 Liter) eluiert. Das Eluat wurde unter vermindertem Druck auf ein Volumen von 50 ml eingeengt. Hierauf wurde es mit   50 g    Diatomeenerde vermischt und das Gemisch getrocknet. Das getrocknete Gemisch wurde anschliessend in einer mit Kieselsäure beschickten Säule (Entwicklerlösungsmittel: ein Gemisch von Chloroform und Methanol im Mischungsverhältnis von 10:1) der Säulenchromatographie unterworfen. Die die gewünschte Verbindung enthaltenden Fraktionen wurden gesammelt und unter vermindertem Druck zur Trockne eingedampft.

  Der so erhaltene Rückstand wurde in 20 ml Methanol gelöst. Diese Lösung wurde mit 40 ml Wasser versetzt, worauf das Gemisch über Nacht bei 4   "C    stehen gelassen wurde, wodurch sich ein Niederschlag (560 mg) bildete, der durch Filtrieren abgetrennt und hierauf in 10 ml Methanol gelöst wurde.



  Zu dieser Lösung gab man 20 ml Wasser hinzu, worauf das Gemisch über Nacht bei 4   "C    stehen gelassen wurde. Dabei bildeten sich Kristalle, die man abtrennte und trocknete,   um,auf    diese Weise Nocardicin F (300 mg) in Form von weissen Kristallen zu erhalten.



  Beispiel 5 Herstellung von Nocardicin G
Ein wässriges Medium (100 ml), welches 2%   Saccharose, 2%      Baumwollsamenmehl, 1%    getrocknete   Hefe, 2,18%      KH2PO4    und   1,43% NaHPO4-      12H20    enthielt, wurde in jeweils acht 500-ml-Schüttelkolben eingetragen und darin während 20 Minuten bei 120   "C    sterilisiert Hierauf wurde mit einer Öse voll Schrägkultur von Nocardia uniformis subsp. tsuyamanensis ATCC 21806 jedes Medium angeimpft und der Organismus während 48 Stunden bei 30   "C    gezüchtet.



   Andererseits wurde ein anderes wässriges Medium (40 Liter), welches 2% lösliche   Stärke, 2%      Baumwollsamenmehl, 2%    getrocknete   Hefe, 2,18%      KH2PO4, 1,43%      NaHPO4- 12H2O,    0,5%   MgSO-      7H20,0,1%    L-Tyrosin und 0,1% Glycin enthielt, durch Zugabe einer wässrigen   6n-Natriumhydroxydlösung    auf einen pH-Wert von 6,0 eingestellt, worauf man 20 Liter des Mediums in zwei 30-Liter-Fermentierbehälter goss und   wäh-    rend 20 Minuten bei 120   "C    sterilisierte. Ein jedes dieser Medien wurde mit der nach den obigen Angaben erhaltenen Samenkultur in einer Menge von 2   Volumen%    des Mediums versetzt. 

  Der Organismus wurde während 96 Stunden bei 30   "C    im Medium wachsen gelassen. Während dieses Wachstumszustandes wurde die Brühe mit einer Geschwindigkeit von 300   Umdrehungen pro Minute gerührt und sterile Luft gleichzeitig mit einer Geschwindigkeit von 20 Litern pro Minute in die Brühe eingeleitet.



   Hierauf wurde die Kulturbrühe (35 Liter) durch Diatomeenerde (3,5 kg) hindurchfiltriert. Das Filtrat (30 Liter) wurde unter vermindertem Druck auf 3 Liter eingeengt. Das Konzentrat wurde hierauf unter Rühren mit 15 Liter Methanol versetzt, worauf man das Gemisch während 1 Stunde stehen liess.



  Dabei bildete sich ein Niederschlag, welcher durch Filtrieren entfernt wurde. Das Filtrat (17 Liter) wurde unter vermindertem Druck auf 1 Liter eingeengt.



   Sämtliche oben erwähnten Vorzüge wurden bei niedriger Temperatur von 4   "C    durchgeführt.



   Das Konzentrat wurde durch Zugabe von 6n-Salzsäure auf einen pH-Wert von 2,0 eingestellt. Dann wurde es mit 2 Liter Äthylacetat versetzt, worauf man das Gemisch während 10 Minuten rührte und die wässrige Schicht abtrennte. Zur wässrigen Schicht gab man 1,5 Liter n-Butanol und 250 g Natriumchlorid hinzu, worauf man das Gemisch während 10 Minuten rührte und die n-Butanolschicht abtrennte. Diese Extraktion wurde noch einmal wiederholt. Die vereinigten n-Butanolschichten (3 Liter) wurden mit 3 Liter n-Hexan versetzt, worauf man das Gemisch während 10 Minuten rührte und die wässrige Schicht (400 ml) abtrennte. Die wässrige Schicht wurde durch Zugabe einer wässrigen   1 n-Natriumhydroxydlösung    auf einen pH-Wert von 7,0 eingestellt und dann unter vermindertem Druck auf ein Volumen von 300 ml eingeengt.

  Das Konzentrat wurde anschliessend mit 5 Liter Wasser und 50 g Natriumchlorid versetzt, worauf man das Gemisch durch Zugabe von   I n-Salzsäure    auf einen pH-Wert von 4,0 ansäuerte. Das Gemisch wird dann durch eine mit Diaion HP20 (2 Liter) beschickten Säule geleitet. Anschliessend wurde die gewünschte Verbindung mit 20 Liter Wasser aus der Säule eluiert. Das die gewünschte Verbindung enthaltende Eluat wurde gesammelt. Das Eluat wurde dann mit 60 g   Na2HPO4      12H20    versetzt, worauf man das Gemisch mittels einer wässrigen   1 n-Natriumhydroxydlösung    auf einen   pH-Wf    von 9,0 einstellte. Dann wird dieses Material in einer mit 700   X    Diaion HP20 beschickten Säule der Säulenchromatographie unterworfen.

  Die Säule wurde mit   1%iger    wässriger Lösung von sek. Natriumphosphat (1 Liter) gewaschen.   Anschliessen    wurde das gewünschte Produkt mit 3 Liter Wasser eluiert.   De    Eluat (2 Liter), welches die gewünschte Verbindung enthielt, wurde gesammelt und dessen pH-Wert durch Zugabe von   ln-Salzsäure    auf 8,0 eingestellt. Hierauf wurde das ganze Mat rial durch eine mit 300 g Aktivkohle beschickte Säule geleitet Die Säule wurde anschliessend mit 1 Liter Wasser gewascher Hierauf wurde die gewünschte Verbindung mit 2 Liter 80%igem wässrigem Methanol eluiert. Das Eluat wurde unter vermindertem Druck auf ein Volumen von 50 ml eingeengt.

 

  Dann wurde das Konzentrat lyophilisiert, mit wenig Cellulose gemischt und schliesslich über Cellulose der Säulenchromato graphie unterworfen. Die Säule wurde mit mit Wasser gesätti tem n-Butanol entwickelt. 400 ml dieses Eluats, welches die gewünschte Verbindung enthielt, wurden gesammelt. Dieses Eluat wurde dann mit 400 ml n-Hexan versetzt, worauf man d Gemisch während 10 Minuten rührte und die wässrige   Schicht    (60 ml) abtrennte. Die wässrige Schicht wurde dann durch Zugabe einer wässrigen 1 n-Natriumhydroxydlösung auf eine pH-Wert von 5,0 gebracht und unter vermindertem Druck au ein Volumen von 30 ml eingeengt. Dieses Konzentrat wurde dann über Nacht stehen gelassen, wobei sich Kristalle bildete die man abtrennte, mit wenig Wasser wusch, trocknete und hierauf umkristallisierte, um auf diese Weise Nocardicin G (1 mg) in Form von weissen Kristallen zu erhalten.

A microorganism which is used according to the invention is a strain which belongs to the genus Nocardia and is capable of producing nocardicin C, D, E, F and / or G.

   It should be understood that the present invention should in no way be limited to the use of the specific organism described here for the production of the nocardicines sought according to the invention, since this organism is only mentioned for the purposes of illustration.



  The invention is also intended to relate to the use of natural mutants and of artificially created mutants which can be derived from the microorganism described here, e.g. B. by irradiation with X-rays or ultraviolet rays, by treatment with N'-nitro-N-nitrosoguanidine, 2-aminopurine or nitrogen mustard or the like.



   The inventive method is advantageously carried out so that one of the genus Nocardia, for. B. Nocardia uniformis subsp. tsuyamanensis, belonging to, Nocardicin C, D, E, F and / or G producing strain in a nutrient medium which contains assimilable carbon and nitrogen sources, grown under submerged, aerobic conditions. Any nutrient medium can be used which can be used by the said microorganism for the production of the nocardicines desired according to the invention. The preferred carbon sources are carbohydrates such as glucose, sucrose, maltose, glycerin, starch, etc.

  The medium in which the vegetative inoculum is produced can be practically the same medium as that used for the production of the nocardicins, or it can be a different medium.



   The stirring and aerating of the cultivation mixture can be effected in various ways. The stirring can be done, for example, with a propeller or with a similar mechanical stirring equipment, by rotating or shaking the fermentation tank or by using different pump systems or by passing sterile air through the medium. Aeration can be carried out by introducing sterile air into the fermentation mixture. In the course of the fermentation, the medium can be defoamed, in particular in those cases in which the culture medium foams in a remarkable manner.    The nocardicins and their salts obtainable according to the invention have specific antibiotic spectra, whereby they have an action against pathogenic bacteria with low toxicity. The nocardicins and their salts obtainable according to the invention are therefore valuable for the treatment of infectious diseases which are caused by such bacteria in humans and animals.



   The pharmacological tests of some of the nocardicins obtainable according to the invention, i. H. antimicrobial and toxicity tests can be found below.



  Minimum inhibitory concentration (M.I.C.)
The M.L.C. test was performed using a standard agar dilution method using cardiac infusion agar (or Mueller Hinton agar) which was incubated at 37 C for 20 hours. M.I.C. is expressed as the minimum concentration of Nocardicin C, D, E or F (mcglml) which prevents the growth of the microorganism. The results can be found in Table 6 below.

An aqueous sodium hydroxide solution of Nocardicin C with a pH of 7.4 became intravenous

   Furthermore, aqueous sodium hydroxide solutions of nocardicin D, E, F and G with a pH of 7.4 were each subcutaneously injected with five mice from the ICR strain weighing 18 to 22 g (dose: 500 mg / kg), and then, one week after the administration, the animals were observed. It was found that all of the animals tested behaved normally throughout the duration.



   The new nocardicins and the pharmaceutically acceptable salts thereof can be formulated for administration in any manner, as is normally the case with known antibiotics, by admixing them with a pharmaceutical carrier.



   Pharmaceutically acceptable salts of the nocardicines obtainable according to the invention include salts with inorganic or organic bases, e.g. As sodium hydroxide, potassium hydroxide, calcium hydroxide, ammonia, ethanolamine, triethylamine, dicyclohexylamine and the like.



   Thus, the antimicrobial preparations in the form of pharmaceutical forms, e.g. B. in solid, semi-solid or liquid form, the active substance being mixed with a pharmaceutical, organic or inorganic carrier or filler which is suitable for external, enteral or parenteral administration. The active substance can, for example, be compounded with conventional carriers, such as those used for tablets, beads, capsules, suppositories, solutions, emulsions, suspensions, etc. Other shapes may also be suitable.

  Water and glucose can be used as carriers
Lactose, acacia, gelatin, mannitol, starch paste,
Magnesium trisilicate, lime, maize starch, keratin, colloidal silica gel, potato starch, urea and other carriers which are suitable for the production of such preparations in solid, semi-solid or liquid form.   The nocardicins or their salts obtainable according to the invention are incorporated into the antimicrobial preparations in such an amount that is sufficient to ensure the desired therapeutic effect on the bacterially infected process or condition.



   If you want to use the antimicrobial preparations in question in humans, you will preferably do so in the form of intravenous or intramuscular injections. The dosage or the therapeutically effective amount of the new nocardicins and their salts fluctuates depending on the age and condition of the individual patient to be treated. Daily doses of approximately 2 to 50 mg of active substance / kg body weight in humans or animals are generally used for the treatment of diseases.



   The nocardicines can also be used as intermediates for the preparation of the other 3-acylamino-1 <a-carboxy-t-hydroxy benzyl> 2-azetidinones with better antimicrobial activity against pathogenic, gram-positive bacteria can be used.

   The following examples illustrate the present invention.



  Example 1 Preparation of Nocardicin C
An aqueous medium (100 ml) containing 2% sucrose, 2% cottonseed flour, 1% dried yeast, 2.18% KH2PO4 and 1.43% Na2HPO4.12H20 was each poured into 20 500 ml shake flasks and for 20 minutes sterilized at 120 ° C. Then each medium was inoculated with an eyelet full of slant culture from Nocardia uniformis subsp. tsuyamanensis ATCC 21806 and then grown at 30 ° C. for 48 hours.



   On the other hand, another aqueous medium (100 liters) which extracted 2% soluble starch, 1% peptone, 0.4% yeast extract, 1% KH2PO4.1% Na2HPO4- 12H20.0.5% MgSO- 7H20, 0.1% L- Contains tyrosine and 0.1% glycine, by adding an aqueous 6N sodium hydroxide solution to a pH of 6.0. 20 liters of the medium were poured into five 30 liter fermentation containers and sterilized for 20 minutes.



   Each of these media was mixed with the seed culture obtained as above, in an amount of 2% by volume of medium. The organism was grown for 96 hours at 30 ° C. During this growth period, the broth was stirred at 250 revolutions per minute and treated by blowing sterile air at a rate of 20 liters per minute.



   The culture broth thus obtained (80 liters) was filtered with the aid of diatomaceous earth (8 kg), whereby a filter cake and a filtrate were obtained. The filter cake was mixed with 70% aqueous ethanol (20 liters) and the mixture was then stirred for 1 hour and filtered. This extraction was repeated twice. The combined extracts (40 liters) were concentrated to a volume of 2 liters under reduced pressure.



   On the other hand, the culture filtrate obtained according to the above information was concentrated to a volume of 10 liters.



  This concentrate was then mixed with 50 liters of methanol while stirring, whereupon the mixture was left to stand for 1 hour and then filtered. The filtrate was concentrated to a volume of 2 liters under reduced pressure.



   The combined concentrates were adjusted to pH 3.0 by adding 6N hydrochloric acid and extracted twice with ethyl acetate (4 liters). The aqueous layer was concentrated under reduced pressure to a volume of 1.8 liters. The concentrate was adjusted to a pH of 4.0 by means of an aqueous 1N sodium hydroxide solution and then passed through a column charged with Diaion HP20 (4 liters) in order to obtain an outflowing solution. The column was then washed with water, whereby washing liquids were obtained. The solution which had flowed through the column and the washing liquids were combined and the aqueous solution (6 liters) was adjusted to a pH of 8.0 by adding an aqueous 6N sodium hydroxide solution.

  Then 120 g of activated carbon were added to this solution, whereupon the mixture was stirred for 10 minutes and then filtered. The activated carbon was washed out with 1 liter of water, after which 80% methanol (1 liter) was added.



   The mixture was stirred for 10 minutes and filtered. The elution process was repeated once more.



  The combined eluates (2 liters) were concentrated to a volume of 10 ml. This concentrate was then mixed with 200 ml of methanol with stirring, whereupon the insoluble constituents were filtered off from the mixture. The filtrate was concentrated to a volume of 10 ml. The concentrate was then subjected to column chromatography over cellulose (developer solvent: upper layer of a mixture of n-butanol, acetic acid and water in a mixing ratio of 100: 5:30). The eluate (500 ml) containing the desired compound was collected. 500 ml of n-hexane were then added to the eluate, and the mixture was stirred and the aqueous layer was then separated off. To this aqueous layer was added 100 ml of ethyl acetate and then the mixture was stirred. The aqueous layer was then separated and concentrated to a volume of 20 ml.

  To this concentrate was added 40 ml of acetone and the mixture was left to stand in a refrigerator (5 ° C.), whereby crystals formed. The mixture was filtered and the crystals obtained were dried, giving 26 mg of nocardicin C as crude crystals These crude crystals were dissolved in 4 ml of water and 8 ml of acetone were added to the solution.

 

  The mixture was then left to stand, whereby crystals separated out. These were separated and dried, whereby nocardicin C (15 mg) was obtained in the form of white crystals.



  Example 2 Preparation of Nocardicin D

Each of these media was added with the seed culture as obtained above in an amount of 2% by volume of the medium. The organism was grown at 30 C for 96 hours.

  During the growing period, the culture broth was stirred at 250 revolutions per minute while simultaneously passing sterile air at a rate of 20 liters per minute.



   80 liters of the culture broth thus obtained were then mixed with 8 kg of diatomaceous earth and the mixture was then filtered, a filtrate and a filter cake being obtained. The filtrate (70 liters) was concentrated under reduced pressure to a volume of 7 liters. The concentrate was then mixed with 90 liters of methanol and the mixture was then left to stand for 1 hour and filtered to remove the precipitate. The filtrate (80 liters) was concentrated to a volume of 5 liters. Then the concentrate was mixed with 15 liters of water and 1 kg of activated carbon.



   The mixture was stirred for 10 minutes and filtered to recover the activated carbon. This activated carbon was then mixed with 10 liters of an 80% strength aqueous methanol solution, whereupon the pH of the mixture was adjusted to pH 8.0 by adding 5% strength aqueous ammonia solution and the mixture was stirred for 10 minutes and then filtered. This elution was carried out a second time. The combined filtrate was concentrated under reduced pressure to a volume of 2 liters.



  The concentrate was adjusted to pH 4.0 by adding 6N hydrochloric acid and then passed through a column charged with Diaion HP20 (4 liters). This column was washed with 4 liters of water and the desired compound was eluted with 50% aqueous ethanol. The eluate (1.5 liters) containing the desired compound was collected and concentrated under reduced pressure to a volume of 100 ml. The concentrate was then subjected to column chromatography over cellulose (developing solvent: n-butanol saturated with water). The eluate containing the desired compound (200 ml) was collected and concentrated under reduced pressure to a volume of 10 ml.

  The concentrate was then subjected to column chromatography over cellulose (developer solvent: upper layer of a mixture of ethyl acetate, n-butanol and water in a mixing ratio of 5: 5: 2). The eluate (100 ml), which contained the desired compound, was concentrated under reduced pressure to a volume of 10 ml, a precipitate being formed, which was obtained by filtering and then drying. The precipitate was suspended in 5 ml of water, and the suspension was adjusted to pH 8.0 by adding an aqueous sodium hydroxide solution to dissolve the precipitate.

  The solution was acidified to pH 3.0 by the addition of 1N hydrochloric acid and then left overnight in a refrigerator (5 "C) to form crystals, which were then isolated by filtration. These crystals became then dried to give 50 mg of nocardicin D in the form of white crystals
The filter cake containing the mycelia obtained as above was extracted twice with 70% aqueous ethanol (20 liters). The combined extracts were concentrated to a volume of 2 liters under reduced pressure. The concentrate was adjusted to pH 3.0 by adding 6N hydrochloric acid and extracted three times with ethyl acetate (2 liters) to remove the impurities. The aqueous layer was separated and concentrated under reduced pressure.

  The concentrate was adjusted to pH 4.0 using 6N aqueous sodium hydroxide solution and then passed through a column charged with Diaion HP20 (2 liters). The column was washed with water and the desired compound was eluted with 50% ethanol (1.5 liters). The eluate (1.5 liters) was concentrated under reduced pressure to a volume of 100 ml.



  The residue thus obtained was subjected to column chromatography over cellulose (developer solvent: n-butanol saturated with water). The eluate containing the desired compound was collected and concentrated under reduced pressure. The residue obtained was again subjected to column chromatography over cellulose (developer solvent: upper layer of a mixture of ethyl acetate, n-butanol and water in a mixing ratio of 5: 5: 2). The eluate containing the desired compound was concentrated under reduced pressure to a volume of 10 ml, a precipitate being obtained, which was separated off by filtration and then dried.

  The precipitate was suspended in 2 ml of water and this suspension was adjusted to a pH of 8.0 by adding an aqueous 1N sodium hydroxide solution, as a result of which the precipitate was dissolved. The solution was then adjusted to a pH of 3.0 by adding 1N hydrochloric acid and left to stand overnight in a refrigerator (5 ° C.), during which crystals formed which were separated off by filtration and dried This gave 10 mg of nocardicin D in the form of white crystals.



  Example 3 Preparation of Nocardicin E.
An aqueous medium (300 ml) containing 2% sucrose, 2% cottonseed flour, 1% dried yeast, 2.18% KH2PO4 and 1.43% NaHPO 'to 1 2H20 was added to a 1 liter Erlenmeyer flask and for 20 minutes Sterilized at 120 ° C.    A 500 liter fermentation tank was charged with the same medium (150 liters) as mentioned above.



  The fermentation medium was sterilized at 120 ° C. for 20 minutes and then inoculated with the entire volume of vegetative inoculum as obtained according to the above information and grown at 30 ° C. for 42 hours.

 

   On the other hand, an aqueous medium (3000 liters) containing 2% soluble starch, 1% peptone, 0.4% yeast extracts, 1% KH2PO4, 1% Na2HPO4 1 2H20, 0.5% MgSO - 7H20, 0.1% L- Contained tyrosine and 0.1% glycine, placed in a 4000 liter fermentation container and sterilized therein at 120 OC for 20 minutes. The medium was then inoculated with the entire volume of the culture broth obtained as described above.



   The organism was grown in the fermentation medium at 30 ° C. for 119 hours. During this growth period, the culture broth was stirred at 230 rpm while simultaneously passing sterile air through the broth at a rate of 3000 liters per minute. After the cultivation had ended, the culture broth was filtered using diatomaceous earth (180 kg). To a portion of the filtrate (1500 liters) was added 45 kg of activated carbon, whereupon the mixture was stirred for 30 minutes and filtered to separate the activated carbon.

  A mixture of acetone and water in a mixing ratio of 7: 3 (200 liters) was then added to the activated carbon, whereupon the resulting mixture was adjusted to a pH of 8.0 by adding aqueous ammonia and stirred for 60 minutes. 500 liters of the eluate thus obtained were concentrated to a volume of 50 liters. The concentrate was acidified to pH 4.0 by adding 6N hydrochloric acid. The concentrate was then mixed with 60 liters of n-butanol, whereupon the mixture was stirred for 30 minutes. The n-butanol layer (50 liters) was separated and concentrated to a volume of 10 liters under reduced pressure. The concentrate was adjusted to pH 8.0 using 5% aqueous ammonia and stirred for 30 minutes.

  Then the aqueous layer (15 liters) was separated and concentrated under reduced pressure to a volume of 1.3 liters. The concentrate was adjusted to pH 4.0 by adding 6N hydrochloric acid, mixed with 1.3 kg of diatomaceous earth and then dried.



  The powder was washed with 3 liters of chloroform and the desired compound eluted with ethyl acetate. The eluate (5 liters) was concentrated under reduced pressure to a volume of 50 ml. Then the concentrate was mixed with 50 g of diatomaceous earth and dried. The powder was subjected to acid chromatography over silica (developing solvent: ethyl acetate). The eluate (600 ml) was concentrated under reduced pressure to a volume of 10 ml. The concentrate was mixed with 10 g of diatomaceous earth and dried. The powder was then subjected to column chromatography over silica (developer solvent: mixture of chloroform and methanol in a mixing ratio of 100: 7). The eluate (200 ml) was concentrated to a volume of 3 ml under reduced pressure. The concentrate was then mixed with 3 g of diatomaceous earth and dried.

  The powder was subjected to column chromatography over silica (developing solvent: a mixture of chloroform and ethyl acetate in a mixing ratio of 1). The eluate (50 ml) was evaporated to dryness under reduced pressure. The residue thus obtained was dissolved in 1 ml of methanol. This
The solution was then mixed with 5 ml of chloroform and left to stand at 4 C overnight, 230 mg of crystals being formed, which were recrystallized from a mixture of methanol and chloroform (1: 5) (12 ml). In this way, nocardicin in an amount of 190 mg was obtained in the form of white crystals.



  Example 4 Preparation of Nocardicin F

The organism was then grown in the medium for 96 hours at 30 ° C.   After the cultivation had ended, the culture broth (100 liters) was filtered through diatomaceous earth (kg). The filtrate (90 liters) was then mixed with 1.5 kg of activated carbon, whereupon the mixture was stirred for 10 minutes and then filtered. The activated carbon was washed twice with water (10 liters) and the desired compound eluted twice with 80% aqueous methanol (30 liters or 20 liters). The eluate (50 liters) was concentrated under reduced pressure to a volume of 6 liters. The concentrate was adjusted to pH 4.0 by adding 6N hydrochloric acid and then passed through a column charged with Diaion HP20 (3 liters). After washing the column with 1 liter of water, the desired compound was eluted with 40% ethanol (8 liters).



  The fractions (4 liters) containing the desired compound were collected and concentrated under reduced pressure to a volume of 100 ml. The concentrate was mixed with 300 g of powdery cellulose, and the mixture was dried under reduced pressure. The dried powder was fed to a column and eluted therein with acetone (6 liters). The eluate was concentrated under reduced pressure to a volume of 50 ml. Then it was mixed with 50 g of diatomaceous earth and the mixture was dried. The dried mixture was then subjected to column chromatography in a column loaded with silica (developer solvent: a mixture of chloroform and methanol in a mixing ratio of 10: 1). The fractions containing the desired compound were collected and evaporated to dryness under reduced pressure.

  The residue thus obtained was dissolved in 20 ml of methanol. 40 ml of water were added to this solution and the mixture was left to stand at 4 ° C. overnight, forming a precipitate (560 mg) which was separated by filtration and then dissolved in 10 ml of methanol.



  20 ml of water were added to this solution, and the mixture was left to stand overnight at 4 ° C. Crystals which were separated off and dried formed in this way, Nocardicin F (300 mg) in the form of white crystals to obtain.



  Example 5 Preparation of Nocardicin G
An aqueous medium (100 ml) containing 2% sucrose, 2% cottonseed flour, 1% dried yeast, 2.18% KH2PO4 and 1.43% NaHPO4-12H20 was placed in each of eight 500 ml shake flasks and held therein for Sterilized for 20 minutes at 120 ° C.    On the other hand, another aqueous medium (40 liters) containing 2% soluble starch, 2% cottonseed flour, 2% dried yeast, 2.18% KH2PO4, 1.43% NaHPO4-12H2O, 0.5% MgSO- 7H20.0, Contained 1% L-tyrosine and 0.1% glycine by adding an aqueous 6N sodium hydroxide solution to a pH value of 6.0, whereupon 20 liters of the medium were poured into two 30-liter fermentation vessels and during 20 Sterilized for minutes at 120 ° C. The seed culture obtained as described above was added to each of these media in an amount of 2% by volume of the medium.

  The organism was grown in the medium for 96 hours at 30 ° C. During this growth state, the broth was stirred at a rate of 300 revolutions per minute and sterile air was simultaneously introduced into the broth at a rate of 20 liters per minute.



   The culture broth (35 liters) was then filtered through diatomaceous earth (3.5 kg). The filtrate (30 liters) was concentrated to 3 liters under reduced pressure. 15 liters of methanol were then added to the concentrate while stirring, and the mixture was allowed to stand for 1 hour.



  A precipitate formed which was removed by filtration. The filtrate (17 liters) was concentrated to 1 liter under reduced pressure.



   All of the above mentioned advantages were carried out at a low temperature of 4 "C.



   The concentrate was adjusted to a pH of 2.0 by adding 6N hydrochloric acid. Then 2 liters of ethyl acetate were added, and the mixture was stirred for 10 minutes and the aqueous layer was separated. 1.5 liters of n-butanol and 250 g of sodium chloride were added to the aqueous layer, and the mixture was stirred for 10 minutes and the n-butanol layer was separated. This extraction was repeated again. The combined n-butanol layers (3 liters) were mixed with 3 liters of n-hexane, whereupon the mixture was stirred for 10 minutes and the aqueous layer (400 ml) separated. The aqueous layer was adjusted to pH 7.0 by adding an aqueous 1N sodium hydroxide solution and then concentrated to a volume of 300 ml under reduced pressure.

  The concentrate was then mixed with 5 liters of water and 50 g of sodium chloride, whereupon the mixture was acidified to pH 4.0 by the addition of 1N hydrochloric acid. The mixture is then passed through a column charged with Diaion HP20 (2 liters). The desired compound was then eluted from the column with 20 liters of water. The eluate containing the desired compound was collected. 60 g of Na2HPO4 12H20 were then added to the eluate, whereupon the mixture was adjusted to a pH Wf of 9.0 by means of an aqueous 1N sodium hydroxide solution. This material is then subjected to column chromatography in a column loaded with 700 X Diaion HP20.

  The column was washed with 1% aqueous solution of sec. Washed sodium phosphate (1 liter). The desired product was then eluted with 3 liters of water. The eluate (2 liters), which contained the desired compound, was collected and its pH was adjusted to 8.0 by the addition of 1N hydrochloric acid. The entire material was then passed through a column charged with 300 g of activated carbon. The column was then washed with 1 liter of water. The desired compound was then eluted with 2 liters of 80% aqueous methanol. The eluate was concentrated under reduced pressure to a volume of 50 ml.

 

  Then the concentrate was lyophilized, mixed with a little cellulose and finally subjected to column chromatography on cellulose. The column was developed with n-butanol saturated with water. 400 ml of this eluate, which contained the desired compound, was collected. This eluate was then mixed with 400 ml of n-hexane, whereupon the mixture was stirred for 10 minutes and the aqueous layer (60 ml) was separated. The aqueous layer was then brought to pH 5.0 by adding an aqueous 1N sodium hydroxide solution and concentrated to a volume of 30 ml under reduced pressure. This concentrate was then left to stand overnight to form crystals, which were separated, washed with a little water, dried and then recrystallized, to thereby obtain nocardicin G (1 mg) as white crystals.

PATENTANSPRÜCHE 1. Verfahren zur Herstellung eines der folgenden Nocardieine, nämlich Nocardicin C, Nocardicin D, Nocardicin E, Nocardicin F und Nocardicin G, wobei Nocardicin C der folgenden chemischen Struktur: EMI1.1 Nocardicin D der folgenden chemischen Struktur: EMI1.2 Nocardicin E der folgenden chemischen Struktur: EMI1.3 Nocardicin F der folgenden chemischen Struktur: EMI1.4 und Nocardicin G der folgenden chemischen Struktur: EMI1.5 entsprechen, dadurch gekennzeichnet, dass man einen zur Gattung Nocardia gehörenden, Nocardicin C, D, E, F undloder G erzeugenden Stamm in einem wässrigen Nährmedium unter aeroben Bedingungen züchtet und das Nocardicin C, D, E, F oder G daraus gewinnt.  PATENT CLAIMS 1.
