Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen Benzodiazepinderivaten der folgenden Formel:

worin der Rest R1 ein Wasserstoffatom, ein Halogenatom, eine Nitrogruppe oder einen Trifluormethylrest, jeder der Reste R2 und R3 ein Wasserstoffatom, ein Chloratom oder ein Bromatom, einen Alkylrest mit 1 bis 4 Kohlenstoffatomen oder einen Trifluormethylrest, der Rest R4 einen Alkylrest mit 1 bis 4 Kohlenstoffatomen und der Buchstabe n eine ganze Zahl von 1 bis 3 bedeuten, sowie von Säureadditionssalzen davon.
Bei den Verbindungen der obigen Formel (I) sind Beispiele von Halogenatomen Chlor-, Brom-, Jod- und Fluoratome, während als Beispiele von 1 bis 4 Kohlenstoffatome enthaltenden Alkylresten Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n Butyl-, Isobutyl- und tert.-Butylreste in Frage kommen. Die -C,Hin-Gruppe kann eine geradkettige oder eine verzweigte Alkylengruppe mit 1 bis 3 Kohlenstoffatomen sein, wie z. B.
Methylen-, Athylen-, 1-Methyläthylen-, 2-Methyläthylenund Trimethylenreste.
Überraschenderweise wurde festgestellt, dass die Verbindungen der Formel (I) und deren pharmazeutisch annehmbaren Salze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften haben und insbesondere hervorragende Tranquillizer, Sedativa, Muskelrelaxantien und Spasmolytika sind und hexobarbitalpotenzierende Wirkungen aufweisen.
Demzufolge bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung von neuen und wertvollen Benzodiazepinderivaten und Salzen davon, welche hervorragende pharmakologische Eigenschaften aufweisen.
Die neuen Benzodiazepinderivate der allgemeinen Formel (I) lassen sich dadurch herstellen, dass man 2-Aminomethylindolderivate der Formel:

worin die Reste R1, R2 R3, R4 und der Buchstabe n die obigen Bedeutungen haben, oder ein Säureadditionssalz davon mit einem Oxydationsmittel behandelt. Beispiele von Säureadditionssalzen sind Chlorhydrate, Bromhydrate, Sulfate oder Phosphate. Das Oxydationsmittel für diesen Zweck kann beispielsweise Ozon, Wasserstoffperoxyd, eine Persäure, z. B.
Perameisensäure, Peressigsäure oder Perbenzoesäure, Chromsäure und Kaliumpermanganat sein, doch kann man auch andere Oxydationsmittel verwenden. Im allgemeinen erfolgt die Reaktion leicht bei Zimmertemperatur, doch kann man auch höhere oder niedrigere Temperaturen verwenden. So kann man beispielsweise bei Temperaturen im Bereiche von 0 bis ungefähr 100oC oder beim Siedepunkt des Lösungsmittels arbeiten. Vorzugsweise verwendet man Temperaturen von 10 bis 60ob, wobei man die Temperatur so auswählt, dass man die Reaktionsbedingungen lenken kann. Die Reaktionstemperatur variiert mit dem Oxydationsmittel. Das bevorzugte Oxydationsmittel ist Chromsäure oder Ozon. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines Lösungsmittels. Die Auswahl des Lösungsmittels hängt vom verwendeten Oxydationsmittel ab.
Es besteht im allgemeinen vorzugsweise aus Wasser, Aceton, Tetrachlorkohlenstoff, Ameisensäure, Essigsäure, Schwefelsäure usw. Das Oxydationsmittel kann in stöchiometrischer Menge oder in einer grösseren Menge verwendet werden.
Erfolgt die Oxydation unter Verwendung von Chromsäure in Gegenwart von Essigsäure, so wird man vorzugsweise die 2bis 3-fache äquimolare Menge Chromsäure verwenden und die Reaktion bei Zimmertemperatur durchführen. Ein 2-Aminomethylindolderivat wird im Lösungsmittel gelöst oder suspendiert und das Oxydationsmittel in die Lösung oder in die Suspension eingerührt.
Verwendet man Ozon als Oxydationsmittel, so kann man die Umsetzung bei Zimmertemperatur mit Vorteil durchführen. Dabei wird ein 2-Aminomethylindolderivat in einem Lösungsmittel, wie z. B. Ameisensäure, Essigsäure, Tetrachlorkohlenstoff oder dergleichen, gelöst oder suspendiert und ozonisierter Sauerstoff in die Lösung bzw. Suspension eingerührt.
Das erwünschte Benzodiazepinderivat lässt sich aus dem Reaktionsgemisch in roher Form durch Extraktion mit vorheriger oder ohne vorherige Neutralisierung und durch Eindampfen zur Trockne erhalten. Das Produkt wird gewünschtenfalls durch Umkristallisieren aus einem geeigneten Lösungsmittel, wie z. B. Äthanol, Isopropanol oder dergleichen, in an sich bekannter Weise weiter gereinigt.
Erfindungsgemäss lassen sich die folgenden Benzodiazepinderivate und die Säureadditionssalze davon herstellen: 1-(ss-Acetoxyäthyl)-5-phenyl-1,3-dihydro-2H-1 ,4benzodiazepin-2-on 1-(ss-Acetoxyäthyl)-5-phenyl-7-chlor- 1,3 -dihydro-2H- 1,4- benzodiazepin-2-on 1-(ss-Acetoxyäthyl)-5-phenyl-7-brom-1,3-dihydro-2H-1,4- benzodiazepin-2-on 1 -( -Acetoxyäthyl)-5-phenyl-7-nitro- 1,3 -dihydro-2H- 1,4- benzodiazepin-2-on 1-(ss-Acetoxyäthyl)-5-phenyl-7-trifluormethyl-1,3-dihydro- 2H-1 ,4-benzodiazepin-2-on 1-(ss-Acetoxyäthyl)-5-phenyl-6- oder -8-chlor-1,3-dihydro2H-1 ,4-benzodiazepin-2-on 1-ss-Acetoxyäthyl(-5-(o-chlorphenyl)-7-chlor- 1,3 dihydro-2H- 1 ,4-benzodiazepin-2-on <RTI

Die so erhaltenen Benzodiazepinderivate der Formel (I) bilden pharmazeutisch annehmbare Säureadditionssalze mit pharmazeutisch annehmbaren anorganischen und organischen Säuren, wie z. B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Salpetersäure, Essigsäure, Maleinsäure, Fumarsäure, Weinsäure, Bernsteinsäure, Zitronensäure, Camphersulfonsäure, Äthansulfonsäure, Ascorbinsäure, Milchsäure usw.
Die Benzodiazepinderivate der Formel (I) oder deren pharmazeutisch annehmbare Säureadditionssalze sind wertvolle Sedativa, Muskelrelaxantien, Hypnotika und krampflösende Mittel.
So sind beispielsweise 1-(ss -Acetoxyäthyl)-5-phenyl-7-chlor- 1,3 -dihydro- 2H-1,4-benzodiazepin-2-on und 1 -(ss-Acetoxyäthyl)-5-(o-chlorphenyl)-7-chlor-1,3- dihydro-2H-1,4-benzodiazepin-2-on hervorragende krampflösende Mittel, Muskelrelaxantien und Mittel, welche eine hexobarbitalpotenzierende Wirkung bei minimaler Toxizität aufweisen.
Benzodiazepine und Salze davon, wie sie erfindungsgemäss erhalten werden, lassen sich parenteral oder oral in therapeutischen Dosierungsformen je nach Bedarf anwenden. Sie können in Form von festen Substanzen oder flüssigen Präparaten verwendet werden, wie z. B. in Form von Tabletten, Dragees, Kapseln, Suspensionen, Lösungen, Elixieren und dergleichen.
Die Erfindung wird durch das folgende Beispiel erläutert.
Beispiel
Eine Lösung von 1 g Chromsäureanhydrid in 1 cm3 Wasser wird einer Mischung von 1 g 1-(ss-Acetoxyäthyl)-2-aminome- thyl-3-phenyl-5-chlorindol-hydrochlorid und 20 cm3 Essigsäure zugegeben. Das Gemisch wird über Nacht bei Zimmertemperatur gerührt. Das Reaktionsgemisch wird mit Wasser verdünnt, mit Ammoniakwasser alkalisch gestellt und mit Äther extrahiert. Die ätherischen Extrakte werden vereinigt und über Natriumsulfat getrocknet. Dann wird das Lösungsmittel entfernt. Der Rückstand wird auf Kieselgel unter Verwendung von Chloroform als Eluiermittel chromatographiert, wobei man 1-(ss-Acetoxyäthyl)-5-phenyl-7-chlor-1,3-dihydro- 2H-1,4-benzodiazepin-2-on erhält.
Das in üblicher Weise unter Verwendung von ätherischem Chlorwasserstoff erhältliche Chlorhydrat wird aus einer Mischung von Methanol und Aceton umkristallisiert, wobei man farblose Prismen vom Schmelzpunkt 193 bis 1950C unter Zersetzung) erhält.

The present invention relates to a process for the preparation of new benzodiazepine derivatives of the following formula:

In the compounds of the above formula (I), examples of halogen atoms are chlorine, bromine, iodine and fluorine atoms, while examples of alkyl radicals containing 1 to 4 carbon atoms are methyl, ethyl, n-propyl, isopropyl, n-butyl -, isobutyl and tert-butyl radicals are possible. The -C, Hin group can be a straight-chain or a branched alkylene group having 1 to 3 carbon atoms, such as. B.
Methylene, ethylene, 1-methylethylene, 2-methylethylene and trimethylene radicals.
It has surprisingly been found that the compounds of the formula (I) and their pharmaceutically acceptable salts with inorganic and organic acids have valuable pharmacological properties and in particular are excellent tranquillizers, sedatives, muscle relaxants and spasmolytics and have hexobarbital potentiating effects.
Accordingly, the present invention relates to a process for the preparation of new and useful benzodiazepine derivatives and salts thereof which have excellent pharmacological properties.
The new benzodiazepine derivatives of the general formula (I) can be prepared by adding 2-aminomethylindole derivatives of the formula:

Examples of acid addition salts are chlorohydrates, bromohydrates, sulfates or phosphates. The oxidizing agent for this purpose may, for example, be ozone, hydrogen peroxide, a peracid e.g. B.
Performic acid, peracetic acid or perbenzoic acid, chromic acid and potassium permanganate, but other oxidizing agents can also be used. In general, the reaction will readily occur at room temperature, but higher or lower temperatures can be used. For example, you can work at temperatures in the range from 0 to about 100 ° C or at the boiling point of the solvent. The desired benzodiazepine derivative can be obtained from the reaction mixture in crude form by extraction with or without prior neutralization and by evaporation to dryness. The benzodiazepine derivatives of formula (I) thus obtained form pharmaceutically acceptable acid addition salts with pharmaceutically acceptable inorganic and organic acids, such as e.g. B. hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, nitric acid, acetic acid, maleic acid, fumaric acid, tartaric acid, succinic acid, citric acid, camphorsulfonic acid, ethanesulfonic acid, ascorbic acid, lactic acid, etc.
The benzodiazepine derivatives of formula (I) or their pharmaceutically acceptable acid addition salts are valuable sedatives, muscle relaxants, hypnotics and anticonvulsants.
