Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 34Iydroxy-4-oxo-19-nor-A- -homo-steroid-1(10),2,4a-trienen der Formel

worin R einen gesättigten oder ungesättigten niederen aliphatischen Kohlenwasserstoffrest bedeutet.
Die neuen Verbindungen der Formel I sind durch eine spermicide Wirkung ausgezeichnet und können somit zur Steuerung der Fertilität verwendet werden. Besonders wertvoll sind die Verbindungen obiger Formel, worin R eine Methyl- oder Äthinylgruppe bedeutet. So zeigt z.B.
das 3,17ss-Dihydroxy-4-oxo-17α-äthinyl-A-homo- 19-nor -androsta-51(l0)22ss4a-trien an der Ratte bereits bei 2 X 10 mg s.c. eine starke Wirkung auf die Beschleunigung der Eipassage. Sie erzeugt auch mit Dosen von 0,3-3 mg subcutan, weiblichen Ratten gegeben, eine Gebärmutterflüssigkeit, welche die Spermenmotilität in vitro in hohem Masse beeinflusst.
Die neuen Verbindungen sind andererseits auch wertvolle Zwischenprodukte, die nach an sich bekannten chemischen und/oder mikrobiologischen Methoden in andere A-Homo-steroide übergeführt werden können.
Als niedere gesättigte oder ungesättigte aliphatische Reste kommen z.B. Niederalkyl-, wie Methyl-, Äthyl-, Propyl- oder Isopropylreste, Niederalkenyl-, wie Vinyl-, Allyl- oder Methallylreste, oder INiederalkinyl-, wie Äthinyl- oder Propinylreste in Frage. Die niederen Kohlen wasserstofireste beziehen sich im allgemeinen auf Reste mit höchstens 5 Kettenkohlenstoffatomen.
Die neuen 3-Hydroxysteroid-tropone der Formel I werden erhalten, wenn man erfindungsgemäss in einer Verbindung der Formel

die 17-Oxogruppe unter Einführung eines niederaliphatischen Kohlenwasserstoffrestes in 17,x-Stellung in einer metallorganischen Reaktion in die 17p-Hydroxylgruppe überführt.
Die verwendeten Ausgangsstoffe sind neu und können erhalten werden, wenn man ein 4-Oxo-l9-nor-4-homo- -androsta- 1,5(10) -dien mit einem Oxydationsmittel, das sich zur Spaltung von os-Glykolen eignet, z.B. Perjodsäure oder einem Alkali- oder Erdalkalimetallsalz davon, dehydriert, die erhaltenen AHomo-androsta- 1(1 0),2,5- -triene mit Hydrazin und dann mit starken Basen, wie Natrium- oder Kaliumhydroxid umsetzt.
Die metallorganische Reaktion wird zweckmässig in an sich bekannter Weise durchgeführt, z.B. mit einem Metaliderivat, insbesondere einer Grignardverbindung oder einem Alkalimetall- wie Lithium- oder Natriumderivat einer niederaliphatischen Verbindung.
Es können pharmazeutische Präparate zur Anwendung gelangen in der Human- oder Veterinärmedizin, welche die neuen, oben beschriebenen pharmakologisch wirksamen Stoffe der Formel I als aktive Substanzen zusammen mit einem pharmazeutischen Trägermaterial enthalten. Als Träger verwendet man organische oder anorganische Stoffe, die für die enterale, z.B. orale, parenterale oder topicale Applikation geeignet sind. Die pharmazeutischen Präparate können in fester oder in flüssiger oder halbflüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Die erfindungsgemäss erhältlichen Verbindungen können auch als Ausgangsprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
In 175 ml Tetrahydrofuran leitet man unter Rühren bei Zimmertemperatur während 30 Minuten Acetylen ein. Dann wird innert 12 Minuten mit 220 ml einer 0,36 m Lösung von Methylenmagnesiumchlorid in Tetrahydrofuran versetzt. Nach weiteren 30 Minuten wird der Acetylenstrom durch Stickstoff ersetzt, worauf man 2,2 g 3 -Hydroxy-4,17-dioxovA-homo-Al'l0)2224a-l9-nor-androsta- trien unter Nachspülen mit 11 ml Tetrahydrofuran zugibt. 2 Stunden später wird unter Kühlung mit einer Eis Methanolmischung langsam mit 225 ml gesättigter Seig- nettesalzlösung versetzt. Nach Verdünnen mit Wasser wird mehrmals mit Methylenchlorid extrahiert und anschliessend mit verdünnter Seignettesalzlösung gewaschen, getrocknet und im Vakuum eingedampft.
Durch Chromatographie des Rückstandes an 110 g Kieselgel erhält man aus den mit ToluolEssigester-(4:1)-Gemisch eluierten Fraktionen das 3,17;E-Dihydroxy-4-oxo-170c- -äthinyl-A- homo-#Ú(10),2,4a 19-nor-androstatrien. Es schmilzt nach Kristallisation aus Aceton bei 228-232,50.
Beispiel 2
Zu einer Lösung von 4,32 g 3-Hydroxy-4,17-dioxo-A -homo-#1(10),2,4a-19-nor-androstatrien in 90 ml absolutem Tetrahydrofuran gibt man unter Eiskühlung und Rühren 58 ml einer 3 m Lösung von Methylmagnesiumchlorid in Tetrahydrofuran. Eine Stunde später wird unter Kühlung mit gesättigter Seignettesalzlösung versetzt und mit Methylenchlorid extrahiert. Nach Waschen mit gesättigter Seignettesalzlösung wird getrocknet und im Vakuum eingedampft. Da das so erhaltene Rohprodukt nach iR.- Spektrum noch Ausgangsmaterial enthält, wird es erneut wie oben beschrieben mit Methylrnagnesiumchlorid behandelt. Darauf chromatographiert man an 215 g Kieselgel.
In den mit Toluol-Essigester-(4: 1)-Gemisch eluierten Fraktionen erhält man das 3,178 -Dihydroxy-4-oxo-l 7x- -methyl-A-homo-A (10),2,4a-19-nor-androstatrien von Smp.

Das in diesen Beispielen verwendete Ausgangsmaterial lässt sich wie folgt erhalten:
Eine Mischung von 23 g 4-Oxo-17-äthylendioxy-A -homo-1 (10),3,4a 1 9-nor-androstatrien, 230 ml Äthanol und 23 ml 80%iges Hydrazinhydrat wird während 30 Minuten im Stickstoffstrom unter Rückfluss gekocht. Darauf versetzt man die abgekühlte Reaktionslösung mit Wasser und extrahiert mit Methylenchlorid. Nach Waschen mit verdünnter Kochsalzlösung, Trocknen und Eindampfen wird der Rückstand an 1,1 kg Kieselgel chromatographiert. Das 3-Amino-4-oxo-17-äthylendioxy-A-ho- mo-#1(10),2,4a-19-nor-androstatrien wird mit Toluol-Essig ester-(l:l)JGemisch eluiert. Man erhält nach Umlösen aus einem Methylenchlorid-Äther-Gemisch 18,6 g. Die Verbindung zeigt nach erneutem Umkristallisieren im UV.
Absorptionsmaxima bei 249, 339 und 395 mm (F = 33800, 14800 und 9800).
18,6 g des oben erhaltenen Aminotropons werden mit 600 ml 90%iger Essigsäure während einer Stunde im Stickstoffstrom in ein Bad von 800 gestellt. Nach Eindampfen im Vakuum wird in Methylenchlorid gelöst, mit Wasser, gesättigter Natriumhydrogencarbonatlösung und Wasser gewaschen, getrocknet und erneut im Vakuum eingedampft. Durch Kristallisation des Rückstandes aus einem Methylenchlorid AtherWGemisch erhält man 13,84 g 3-imino-4, 17-dioxo-A-homol-L\1'10' ,3,4a - 19- nor-androsta- trien, das nach erneutem Umlösen bei 244-2450 schmilzt.
Eine Mischung von 4 g 3-Amino-4-oxo-17-äthylendi oxy-A-homo-#1(10),2,4a-19-nor-androstatrien, 500 ml Alkohol und 500 ml 2n Natronlauge lässt man während 14 Stunden im Stickstoffstrom unter Rückfluss kochen. Nach Zugabe einer Mischung von 75 ml Eisessig und 175 ml Wasser wird auf Wasser geleert und mehrmals mit Me thyienchlorid extrahiert. Den'Rückstand, der mitverdünnter Natriumbicarbonatlösung u. Wasser gewaschenen, getrockneten u. im Vakuum eingedampften organischen Lösungen chromatographiert man an 200 g Kieselgel. Die mit Toluol-Essigester-(9:1 )-Gemisch eluierten Fraktionen enthalten das 3-Hydroxy-4-oxo- 17-äthylendioxy-A-homo- (10),2,4a-19-nor-androstatrien, das nach Umlösen aus Methylenchlorid-Äther bei 143 - 1440 schmilzt.
Durch 15minutiges Erhitzen mit 90-proz. Essigsäure im Stickstoffstrom erhält man daraus das 3-Hydroxy -4,17-dioxo-A-homo-#1(10),2,4a-19- nor-androstatrien. Es schmilzt nach Umkristallisieren aus Methylenchlorid Äther bei 184-185,5 .
