worin Het, X, Y, RÚ und Rê obige Bedeutung besitzen und Y und -CRÚRê-CH2OH an nicht benachbarte Koh lenstoffatome des heterocyclischen Ringes Het gebunden sind, mit einem den Rest R3 einführenden Acylierungs- mittel behandelt.
Als Acylierungsmittel kann man beispielsweise ein Säureanhydrid oder Säurehalogenid verwenden, und die Acylierung kann in einem Überschuss des Acylierungs- mittels und/oder in Gegenwart eines inerten Verdün nungsmittels oder Lösungsmittels, z.B. Pyridin, ausge führt werden. Die Umsetzung mit einem Säureanhydrid oder Säurehalogenid kann unter dem Einfluss von Wär me ausgeführt werden. Die Acylierung kann aber auch unter Verwendung der entsprechenden Carbonsäure und von Dicyclohexylcarbodiimid in einem Verdünnungs mittel oder Lösungsmittel, z.B. Tetrahydrofuran, erfol gen.
Falls R3 den N-Phenylcarbamylrest darstellt, kann die Acylierung unter Verwendung von Phenylisocyanat als Acylierungsmittel ausgeführt werden, gegebenenfalls in Gegenwart eines inerten Verdünnungsmittels oder Lösungsmittels, z.B. Benzol.
Es ist selbstverständlich, dass die Verbindungen, wenn RÚ und Rê verschieden sind, racemisch sind und in bekannter Weise in die entsprechenden d- und @- Iso- mere aufgespalten werden können. Die Herstellung von Verbindungen, in welchen Y einen oder zwei Substituen- ten der oben beschriebenen Art enthält, stellt eine be vorzugte Ausführungsform der Erfindung dar, weil sie allgemein wirksamer sind als die entsprechenden unsub- stituierten Derivate.
Eine geeignete Bedeutung von X ist z.B. Wasser stoff oder der Methyl- oder Methoxyrest. Der bzw. die Halogensubstituenten in Y können z.B. Fluor-, Chlor- und/oder Bromatome sein.
Eine geeignete Bedeutung von RÚ oder Rê ist z.B. Wasserstoff oder der Methylrest. Eine geeignete Be deutung von R3 ist beispielsweise der Acetyl-, Propio- nyl-, Benzoyl-, o-Carboxybenzoyl-, o-Acetoxybenzoyl-, a - (2 - Isopropyl - 5 - methylcyclohexyloxy) - acetyl-, 2 - (4- -Chlorphenyl)-thiazolyl-(4)-acetyl- oder N-Phenylcarb- amylrest.
Als geeignete Säureadditionssalze seien nicht toxi sche pharmazeutisch verwendbare Säureadditionssalze, z.B. Hydrochloride oder Tartrate, erwähnt.
Die als Ausgangsprodukte verwendeten Alkohole (II) können durch Reduktion der entsprechenden Ester mittels Lithiumaluminiumhydrid oder Natriumborhydrid hergestellt werden. Die verfahrensgemäss eingesetzten Acylierungsmittel können durch bekannte Methoden er halten werden.
<I>Beispiel 1</I> Ein Gemisch aus 2-(4-Chlorphenyl)-4-(2-hydroxy- äthyl)-thiazol (0,48 g) und Essigsäureanhydrid (0,41 g) wurde während 18 Stunden auf 100 C erhitzt. Das Ge misch wurde in Chloroform (5 cm ) gelöst und die Lö sung nacheinander mit Wasser, gesättigter Natriumbicar- bonatlösung und Wasser gewaschen. Die Chloroform schicht wurde mit wasserfreiem Magnesiumsulfat ge trocknet und das Chloroform bei vermindertem Druck abdestilliert, wobei ein weisser fester Rückstand zurück blieb, der aus Hexan kristallisiert wurde. Auf diese Weise wurde 4-(2-Acetoxyäthyl)-2-(4-chlorphenyl)-thia- zol vom Schmelzpunkt 50 bis 52 C erhalten.
Das Ausgangsprodukt kann folgendermassen herge stellt werden.
2-(4-Chlorphenyl)-thiazolyl-(4)-essigsäuremethylester (16 g, Schmelzpunkt 72 C) wurde in warmem trocke nem Äthanol (125 cm ) gelöst und unter Rühren mit Natriumborhydrid (8 g) versetzt. Das Gemisch wurde 1 Stunde zum Rückfluss erhitzt und dann unter gutem Rühren die Lösung tropfenweise mit Wasser (125 cm ) versetzt. Der zuerst gebildete Niederschlag löste sich wieder auf, wobei ein klebriger Rückstand zurückblieb, der getrennt in heissem Wasser (25 cms ) gelöst wurde. Diese Lösung wurde zu der klaren überstehenden Flüs sigkeit gegeben. Das ganze klare Gemisch wurde auf 5 bis 10 C abgekühlt und unter kräftigem Rühren trop fenweise mit Wasser (225 cm ) versetzt.
Die Emulsion, die sich bildete, musste gewöhnlich mit einer kleinen Menge des Produktes angeimpft werden, ehe der Nie derschlag schnell erstarrte. Das Gemisch wurde während 1 Stunde bei 5 C gerührt und dann filtriert. Die zurück bleibende feste Substanz wurde auf dem Filter trocken gepresst und dann so weit wie möglich in warmem Äthanol (125 cm ) wieder aufgelöst. Die klare Lösung wurde von dem klebrigen unlöslichen Rückstand abde kantiert. Nach dem Abkühlen der klaren Lösung auf 5 bis 10 C wurde unter kräftigem Rühren tropfenweise Wasser (375 cm ) zugesetzt, um das Produkt auszufällen (wiederum war gewöhnlich ein Animpfen der zuerst ge- bildeten Emulsion erforderlich, ehe der Niederschlag erstarrte).
Das Produkt wurde nach dem Rühren des Gemisches während 1 Stunde bei 5 C abfiltriert und im Vakuum bei 35 C getrocknet. Auf diese Weise wurde 2-(4-Chlorphenyl)-4-(2-hydroxyäthyl)-thiazol vom Smp. 66 bis 66,5 C erhalten.
<I>Beispiel 2</I> Ein Gemisch aus 2-(4-Chlorphenyl)-4-(2-hydroxy- äthyl)-thiazol (4,8 g) und Propionsäureanhydrid (5,2 g) wurde während 18 Stunden auf 100 C erhitzt. Das Reak tionsgemisch wurde in Chloroform (50 cm') gelöst und die Lösung nacheinander mit Wasser, gesättigter Na- triumbicarbonatlösung und Wasser gewaschen. Die Chlo roformschicht wurde mit wasserfreiem Magnesiumsulfat getrocknet und das Chloroform bei vermindertem Druck abdestilliert, wobei ein weisser fester Rückstand zurück blieb, der aus Hexan kristallisiert wurde.
Auf diese Weise wurde 2-(4-Chlorphenyl)-4-(2-propionyloxyäthyl)- -thiazol vom Schmelzpunkt70 bis 72 C erhalten.
<I>Beispiel 3</I> Ein Gemisch aus Dicyclohexylcarbodiimid (4,12g) und 2-(4-Chlorphenyl)-thiazolyl-(4)-essigsäure (5,07 g) in wasserfreiem Tetrahydrofuran (10 cm ) wurde während 30 Minuten bei O C gerührt. 2-(4-Chlorphenyl)-4-(2-hy- droxyäthyl)-thiazol (4,79 g) wurde in wasserfreiem Te- trahydrofuran (10 cm ) gelöst und die Lösung bei 0 C unter Rühren tropfenweise zu dem obigen Gemisch gegeben. Das resultierende Gemisch wurde während weiteren 2 Stunden bei 0 C und dann während 3 Tagen bei Umgebungstemperatur gerührt.
Das Gemisch wurde darauf filtriert und das Filtrat zur Trockene eingedampft, wobei ein weisser fester Rückstand zurückblieb. Die feste Substanz wurde so weit wie möglich in warmem Cyclohexan (l50 cm ) gelöst, das Gemisch noch warm filtriert und das Filtrat auf Umgebungstemperatur ab kühlen gelassen. Das resultierende Gemisch wurde fil triert und ergab als festen Rückstand 2-(4-Chlorphenyl)- -4 - f 2 - [2 - (4 - chlo rphenyl)-thiazolyl- (4) -acetoxy]-äthyl}- -thiazol vom Schmelzpunkt 118 bis 119 C.
<I>Beispiel 4</I> Eine Lösung von 2-(4-Chlorphenyl)-4-(2-hydroxy- äthyl)-thiazol (3 g) in trockenem Benzol (25 cm ) wurde bei Umgebungstemperatur unter Rühren tropfenweise zu einer Lösung von Phenylisocyanat (1,48g) in trocke nem Benzol (5 cm ) gegeben. Das Gemisch wurde bei Umgebungstemperatur während 18 Stunden gerührt und dann filtriert. Der feste Rückstand bestand aus 2-(4- -Chlorphenyl) -4- (2-N-phenylcarbamyloxyäthyl)-thiazol vom Schmelzpunkt 122 bis 125 C.
<I>Beispiel 5</I> Acetylsalicylylchlorid (7,15 g), gelöst in trockenem Benzol (25 cm3), wurde unter Rühren und Kühlen auf eine Temperatur unter 20 C zu einer Lösung von 2-(4 -Chlorphenyl)-4-(2-hydroxyäthyl)-thiazol (6,3 g) und Tri- äthylamin (7 cm3) in trockenem Benzol (100 cm3) gege ben.
Das Gemisch wurde über Nacht gerührt und zum Rückfluss erhitzt. mit Chloroform verdünnt und die organische Phase mit gesättigter Natriumbicarbonatlö- sung gewaschen und mit wasserfreiem Magnesiumsulfat getrocknet. Das organische Lösungsmittel wurde unter vermindertem Druck abdestilliert, wobei eine rote feste Substanz zurückblieb, die aus einem kleinen Volumen Äthanol kristallisiert wurde. Auf diese Weise wurde 4- -(2-Acetylsalicylyloxyäthyl) - 2 - (4-chlorphenyl)-thiazol v. Schmelzpunkt 99 bis 100 C erhalten.
In ähnlicher Weise wurde die folgende Verbindung aus den entsprechenden Ausgangsmaterialien erhalten: 4-(4-Bromphenyl) - 2 - f 2 - [c- (2 - isopropyl-5-methylcyclo- hexyloxy)-acetoxy]-isopropoxy@-thiazol, starke =C=O- Absorption bei 1750 cm-Ú und keine -O-H-Absorption bei 3400 cm-.
<I>Beispiel 6</I> 4 - (4 - Bromphenyl) - 2 - (2 - hydroxyisopropyl) - thiazol (11,92 g) wurde in trockenem Pyridin (64 cm ) gelöst und die Lösung mit gepulvertem Phthalsäureanhydrid (6,2 g) versetzt. Das Gemisch wurde über Nacht bei Raumtemperatur und darauf während 1 Stunde auf dem Wasserdampfbad gerührt. Das Reaktionsgemisch wurde in ein Gemisch aus konzentrierter Salzsäure (82,6 cm ) und Eis gegossen und das resultierende Gemisch mit Chloroform (dreimal 25 cm ) extrahiert. Die vereinigten Chloroformextrakte wurden mit Wasser gewaschen und dann zur Trockene eingedampft. Der Rückstand wurde in überschüssigem warmem verdünnten Ammoniumhy droxyd gelöst und die Lösung auf Raumtemperatur ab gekühlt. Das Gemisch wurde filtriert und das Filtrat mit konzentrierter Salzsäure auf pH = 4 angesäuert.
Das ausgeschiedene öl wurde mit Chloroform (dreimal 25 cm ) extrahiert und die vereinigten Chloroformextrakte über wasserfreiem Magnesiumsulfat getrocknet. Das Lö sungsmittel wurde unter vermindertem Druck verdampft, und auf diese Weise wurde der Phthalsäurehalbester des 4-(4-Bromphenyl)-2-(2-hydroxyisopropyl)-thiazols erhalten; Infrarotabsorption vom Carboxylgruppentyp zwischen 2600 und 3400 cm-Ú und bei 1680 cm-Ú, keine alkoholische Hydroxylgruppenabsorption bei 3350 -1.

The solvent was evaporated under reduced pressure, and thus the phthalic acid half-ester of 4- (4-bromophenyl) -2- (2-hydroxyisopropyl) thiazole was obtained; Carboxyl group type infrared absorption between 2600 and 3400 cm-Ú and at 1680 cm-Ú, no alcoholic hydroxyl group absorption at 3350 -1.
The starting product can be obtained in the same way as in. Described in the appendix to example 1. 2- (4 - bromophenyl) - 4 - (2-hydroxyisopropyl) -thiazole shows an O-H absorption at 3400 cm-Ú, no = CO-absorption at 1730 cm-Ú.
