Verfahren zur Herstellung von Pyrazokpyriinidinen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Alkyl-4, 6-dioxo-4, 5, 6, 7-tetrahydro- pyrazolo-[3-,4-d]-pyrimidinen, das dadurch gekennzeichnet ist, dass man eine 3-Aminopyrazol-4-carbon- säure oder ein Säurederivat derselben mit Alkylisocyanaten kondensiert und ringschliesst.
Als Alkylisocyanate verwendet man vorzugsweise niedere Alkylisocyanate, z. B. Methylisocyanat oder Äthylisocyanat. Reaktionsfähige oxogruppenhaltige Säurederivate sind vor allem Carbonsäureester, z. B. niedere Alkylester, wie der Methyl- oder Äthylester oder Amide.
Die als Ausgangsstoffe verwendeten 3-Aminopyrazol-4-carbonsäuren können am Ring beliebig substituiert sein. Insbesondere tragen sie jedoch an einem der beiden Ringstickstoffatome eine Alkylgruppe, z. B. eine Methyl- oder Isopropylgruppe.
Die Reaktion, die zweckmässig in Gegenwart von Verdünnungs- und/oder Kondensationsmitteln durchgeführt wird, verläuft besonders leicht und gut bei Verwendung von 3 -Aminopyrazol-4-carbonsäure-niederalkylestern, wie dem 3 ; Aminopyrazol-4-carbon- säure-äthylester. Als Kondensationsmittel können z. B. tertiäre Amine, wie Trimethyl- oder Triäthylamin dienen.
Bei der erfindungsgemässen Reaktion können zunächst die ringoffenen 3 -Carb amylamino-pyrazol-4- carbonsäuren oder ihre Derivate entstehen, oder besonders bei Verwendung eines Uberschusses an Alkylisocyanat, auch die entsprechenden 3-([N-Alkylcarb amyl-alkylcarbamyll'- amino) -Verbindungen. Diese Harnstoff- bzw. Biuretderivate lassen sich leicht zu den Pyrazolo-[3 ,4-d]-pyrimidinen weiter kondensieren, z. B. im Falle von Harnstoff- oder Biuretderivaten der 3-Amino-4-carbalkoxypyrazole durch alkalische Behandlung, wobei Alkohol und, bei Verwendung von Biureten, Kohlensäure und primäres Amin abgespalten wird.
Die nach dem neuen Verfahren erhaltenen 4-R-5 Allcyl -6-oxo-4,5 ,6,7-tetrahydro-pyrazolo-[3-,4-d]-pyri- midine sind zum Teil neu. So sind z. B. die 7-uii & ub- stituierten 1 -Alkyl - 4-R-5-alkyl-6-oxo 4,5,6,7-tetra- hydro-pyrimidine bisher noch nicht beschrieben worden. Die verfahrensgemäss erhaltenen Verbindungen sind wertvolle Verbindungen mit koffeinartiger Wirkung und können somit als Stimulantia oder Diuretica verwendet werden.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie sauren Charakter, so können Me tallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Charakter bilden Salze mit anorganischen oder organischen Säuren.
Als salzbildende Säuren kommen beispielsweise in Frage: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphor säuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Apfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxyma eine oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyloder p-Aminosalicylsäure; Methansulfon-, Äthansulfon-, Oxyäthansulfon-, Athylensulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Die als Ausgangsstoffe verwendeten l-Alkyl-3amino-pyrazol-4-carbonsäuren und ihre funktionellen Derivate sind neu. Ihre Herstellung ist in unserem Patent Nr. 396 011 beschrieben. Sie erfolgt durch Umsetzung von a-Cyan-ss-oxo-propionsäuren oder ihren funktionellen Säure- und/oder Oxoderivaten, z. B. Äthoxymethylencyanessigester, mit N-Alllyl- hydrazinen, deren N'-Stickstoffatom durch einen hydrolytisch abspaltbaren Rest geschützt ist, und nachfolgende Hydrolyse und Ringschluss. Die übrigen Ausgangs stoffe sind bekannt oder werden in analoger Weise gewonnen.
Die oben beschriebenen Pyrazolopyrimidine, ihre Salze, oder entsprechende Gemische können z. B. in Form pharmazeutischer Präparate Verwendung finden.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1 19,7 g 2-Isopropyl-3 -amino-4-carbäthoxy-pyrazol, gelöst in 150 cm3 Benzol, werden mit 28,4 g Äthylisocyanat und 3 cm3 Triäthylamin 10 Stunden im geschlossenen Rohr auf 1000 erhitzt. Nach Eindampfen der Reaktionslösung erhält man rohen, kristallinen N- [2-Isopropyl-4-carbäthoxy-pyrazolyl-(3)]-N'-äthyl- N'-äthylcarbamyl-harnstoff der Formel

Das Rohprodukt, das nach Umkristallisieren in Äther bei 129-1300 schmilzt, kann direkt für die nachfolgend beschriebene Cyclisierung verwendet werden:
16 g roher N-[2-Isopropyl-4-carbäthoxy-pyrazolyl-(3)]-N'-äthyl-N'-äthylcarbamyl-harnstoff werden mit 160 cm3 2n Natronlauge versetzt und 8 Minuten in einem Ölbad von 1500 am Rückfluss gekocht. Setzt man im obigen Beispiel das 2-Isopropyl-3 amino-4-carbäthoxy-pyrazol nur mit einem Mol Äthylisocyanat um, so erhält man ein Zwischenprodukt, aus dem sich nach Abdestillation des Ausgangsproduktes bei 1270/0,99 mm Hg, der N-[2-Isopro pyl -4- carbäthoxy - pyrazoly1 -(3)]-N'-äthyl-harnstoff vom F. 129-1300 (aus Äther-Petroläther kristallisiert) isolieren lässt. Er lässt sich wie oben angegeben cyclisieren.

13,8 g 2-Isopropyl-3-amino-4-carbäthoxy-pyrazol werden in 105 cm3 Benzol mit 16 g Methylisocyanat und 2,1 cm3 Triäthylamin im geschlossenen Rohr 10 Stunden auf 1000 erhitzt. Nach Eindampfen der Reaktionslösung erhält man rohen kristallinen N-[2-Isopropyl-4-carbäthoxy-pyrazolyl-(3)]-N'-methyl-N'-me thylcarbamyl-harnstoff der Formel

Die im obigen Beispiel und in Beispiel 4 verwendeten l-Alkyl-pyrazole werden wie folgt erhalten:
8 g N1-Isopropyl-N2-benzyliden-hydrazin und 8 g Äthoxymethylen-cyanessigester werden in 50 cm3 Benzol während 10 Stunden auf 800 erwärmt. Nachdem man das Lösungsmittel im Vakuum entfernt hat, wird der Rückstand aus Äthanol umkristallisiert. Man erhält den ss-(N2-benzyliden-N1-isopropyl-hydrazino) ocyan-acrylsäureäthylester in gelben Prismen vom F. 118-1200.
4 g ss-(N2-benzyliden-N1-isopropyl-hydrazino)-α- cyan-acrylsäureäthylester kocht man 2 Stunden mit lOn alkoholischer Salzsäure und entfernt hierauf den Alkohol durch Destillation im Vakuum. Der Rückstand wird in 200 cm3 2n Salzsäure aufgenommen und die Lösung mit Äther extrahiert. Nach dem Abtrennen der wässerigen Lösung wird diese durch Zu gabe von 2n Natronlauge alkalisch gemacht. Die ausgeschiedene Base extrahiert man mit Äther. Nach dem Trocknen und Abdampfen des ethers bleibt 1 Isopropyl-3-amino-4-carbäthoxy-pyrazol der Formel

Process for the preparation of pyrazokpyriinidines
The invention relates to a process for the preparation of alkyl-4, 6-dioxo-4, 5, 6, 7-tetrahydropyrazolo- [3-, 4-d] -pyrimidines, which is characterized in that a 3- Aminopyrazole-4-carboxylic acid or an acid derivative thereof condenses with alkyl isocyanates and closes the ring.
The alkyl isocyanates used are preferably lower alkyl isocyanates, e.g. B. methyl isocyanate or ethyl isocyanate. Reactive acid derivatives containing oxo groups are mainly carboxylic acid esters, e.g. B. lower alkyl esters, such as methyl or ethyl esters or amides.
The 3-aminopyrazole-4-carboxylic acids used as starting materials can be substituted as desired on the ring. In particular, however, they carry an alkyl group on one of the two ring nitrogen atoms, e.g. B. a methyl or isopropyl group.
The reaction, which is expediently carried out in the presence of diluents and / or condensing agents, proceeds particularly easily and well when using 3-aminopyrazole-4-carboxylic acid lower alkyl esters, such as the 3; Aminopyrazole-4-carboxylic acid ethyl ester. As a condensing agent, for. B. tertiary amines, such as trimethylamine or triethylamine.
In the reaction according to the invention, the ring-open 3-carbamylamino-pyrazole-4-carboxylic acids or their derivatives can be formed first, or especially when using an excess of alkyl isocyanate, the corresponding 3 - ([N-alkylcarbamyl-alkylcarbamyl'-amino) Links. Some of the 4-R-5 alkyl-6-oxo-4,5,6,7-tetrahydropyrazolo- [3-, 4-d] -pyrimidines obtained by the new process are new. So are z. B. the 7-uii & ub- substituted 1-alkyl-4-R-5-alkyl-6-oxo 4,5,6,7-tetrahydro-pyrimidines have not yet been described. The compounds obtained according to the method are valuable compounds with a caffeine-like effect and can thus be used as stimulants or diuretics.
Depending on the substituents present in the process products, various salts can be prepared. If they have an acidic character, metal salts can be obtained, e.g. B. by dissolving in alkaline solutions. Compounds of a basic character form salts with inorganic or organic acids.
Examples of suitable salt-forming acids are: hydrohalic acids, sulfuric acids, phosphoric acids, nitric acid, perchloric acid; aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids, such as formic, acetic, propionic, oxalic, amber, glycol, milk, apple, wine, lemon, ascorbic, oxymalein, dioxyma one or pyruvic acid; Phenylacetic, benzoic, p-aminobenzoic, anthranil, p-oxybenzoic, salicylic or p-aminosalicylic acid; Methanesulphonic, ethanesulphonic, oxyethanesulphonic, ethylene sulphonic acid; Toluenesulfonic, naphthalenesulfonic or sulfanilic acid; Methionine, tryptophan, lysine or arginine.
The 1-alkyl-3amino-pyrazole-4-carboxylic acids used as starting materials and their functional derivatives are new. Their production is described in our patent no. 396 011. It is carried out by reacting a-cyano-ss-oxo-propionic acids or their functional acid and / or oxo derivatives, e.g. B. ethoxymethylene cyanetyl acetate, with N-alllyl hydrazines, the N'-nitrogen atom of which is protected by a hydrolytically cleavable radical, and subsequent hydrolysis and ring closure. The other starting materials are known or are obtained in an analogous manner.
The pyrazolopyrimidines described above, their salts, or mixtures thereof can, for. B. find use in the form of pharmaceutical preparations.
