Verfahren zur Herstellung von Buttersäureamiden
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von basisch substituierten Buttersäure- amiden der Formel

worin Rt Wasserstoff bzw. einen Alkylrest mit höch- stens 4 Kohlenstoffatomen und R2 einen gegebenenfalls substituierten Phenylrest oder Rt und R2 gemeinsam mit dem verbundenen Stickstoffatom Glieder eines heterocyclischen Ringsystems bedeuten.
Beispielsweise können nach dem Verfahren gemäss der Erfindung Verbindungen der obigen Formel, worin R2 einen in p-Stel-lung, vorzugsweise durch einen niedrigmolekularen Alkoxyrest substituierten Phenylrest bedeutet, hergestellt werden.
Die nach dem Verfahren gemäss der Erfindung erhaltenen, basisch substituierten Buttersäureamide zeichnen sich durch gute analgetische und sedative Eigenschaften bei überraschend germer Toxizität aus. Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dal3 man die Aminogruppe in a Amino-} S-oxy-buttersäureamiden der Formel

mit Formaldehyd in Gegenwart von Ameisensäure oder mit Formaldehyd in Gegenwart von Wasserstoff und einem Katalysator umsetzt.
Als a-Amino-}-oxy-buttersäureamide kommen beispielsweise in Frage : a-Amino-jB-oxy-buttersäure-anilid, a-Amino-8S- oxy-buttersäure-phenetidid, a-Amino-S-oxy-butter- säure-p-anisidid, a-Amino-, B-oxy-buttersäure-N-me- thyl-p-phenetidid, a-Amino-p-oxy-buttersäure-N-äthyl- p-phenetidid, a-Amino-, B-oxybuttersäure-m-anisidid, a-Amino-S-oxy-buttersäure-N-isopropyl-anilid, a- Amino-ss-oxy-buttersäure-N-n-butyl-p-anisidid, a Amino-p-oxy-buttersäure-N-isobutyl-anilid, a-Amino- p-oxy-buttersäure-o-anisidid, a-Amino-p-oxy-butter- säure-p-toluidid, a-Amino-3-oxy-buttersäure-xylidide, a-Amino-ss-oxy-buttersäure-N-propyl-p-phenetidid,
a Amino-ss-oxy-buttersäure- (p-carbäthoxy)-anilid, a- Amino--oxy-buttersäure-1, 2,3,4-tetrahydro-chinolid und a-Amino-ss-oxy-buttersäure-1, 2,3,4-tetrahydro-6methoxy-chinolid.
Diese Ausgangsstoffe können beispielsweise in der Weise gewonnen werden, dass man entsprechend substituierte Acetessigsäureanilide mit salpetriger Säure behandelt und in den erhaltenen Nitrosierungs- produkten sowohl die Isonitrosogruppe als auch die Ketogruppe in üblicher Weise reduziert. Beispiels- weise kann man die nach dem Verfahren gemäss Patent Nr. 342945 erhältlichen Verbindungen in Form der erythro-oder threo-Verbindungen als Ausgangsstoffe einsetzen.
Formaldehyd wird zweckmässig in wässriger Lösung eingesetzt. Man kann jedoch auch in Gegenwart indifferenter Lösungsmittel, beispielsweise Benzol, Toluol oder Xylol, arbeiten. Weiterhin sind auch aliphatische Alkohole und Cyclohexan als Lösungsmittel geeignet. Auch Paraformaldehyd in Gegenwart indifferenter Lösungsmittel kann eingesetzt werden.
Im Falle der Verwendung von Ameisensäure kann es zweckmässig sein, die Umsetzung durch Er- wärmen auf mässig erhöhte Temperaturen, vorteilhaft auf die Siedetemperatur des verwendeten Lösungsmittels, durchzuführen und das Reaktionsgemisch einige Zeit auf dem Dampfbad zu erhitzen.
Man erhält nach dem Verfahren gemäss der Erfindung kristalline Verbindungen, die je nach den enthaltenen Substituenten mehr oder weniger was serlöslich sind. Sie stellen wertvolle Heilmittel dar und weisen insbesondere eine gute analgetische und sedative Wirkung bei auffallend geringer Toxizität auf. Infolge der damit verbundenen grossen therapeu- tischen Breite sind sie bekannten Verbindungen überlegen.
Beispielsweise entspricht die analgetische Wirkung des a-Dimethylamino-ss-oxy-buttersäure-p- phenetidids, von dem eine 1'Voigc wässrige Lösung hergestellt werden kann, der des bekannten, praktisch wasserunlöslichen N-Acetyl-p-phenetidins. Ausserdem besitzt das a-Dimethylamino-ss-oxy-buttersäure-p- phenetidid eine sedative Komponente und zeigt weiterhin eine antiphlogistische Wirkung, die derjenigen der obigen bekannten Verbindung entspricht.
Von entscheidender Bedeutung bei der Verwen- dung als Heilmittel ist jedoch die gegenüber der obigen bekannten Verbindung erheblich geringere Toxizität des a-Dimethylamino-ss-oxy-buttersäure-p- phenetidids. So beträgt bei oraler Applikation an der Maus die Dos. let. min. 3 g/kg, während diejenige der bekannten Verbindung bei 1,5 g/kg liegt. Bei der Ratte betragen die gleichen Daten 9 glkg und 5 glkg.
Bei Untersuchungen über die chronische Toxizität bei Katzen stellte sich heraus, dass 3 von 4 Tieren 27 Fütterungen mit je 0,25 g a-Dimethylamino-ss- oxy-buttersäure-p-phenetidid pro kg ohne weiteres vertragen ; lediglich ein Tier ging nach 14 Fütterungen ein. Die Applikation der gleichen Dosis der bekannten Verbindung führte bei fast allen Tieren nach 3 bis 4 Fütterungen zum Tode.
Bei Berücksichtigung der mit der besseren Lös- lichkeit verbundenen verbesserten Resorbierbarkeit des erfindungsgemäss erhaltenen a-Dimethylamino-- oxy-buttersäure-p-phenetidids im Vergleich zu der oben genannten bekannten Verbindung werden die Vorteile, welche die erfindungsgemäss erhaltene Verbindung im Hinblick auf die Toxizität bietet, noch deutlicher.
Beispiel 1 a) 23,8 g a-Amino-ss-oxy-buttersäure-pJphenetidid 13,5 g Ameisensäure und 16 g 40 /Oige Formaldehyd- lösung werden eine Stunde auf dem Dampfbad erhitzt, wobei lebhafte Kohlendioxyd-Entwicklung eintritt. Die mit überschüssiger Natronlauge versetzte Lösung wird ausgeäthert. Nach Abdestillieren des Äthers wird a-Dimethylamino-ss-oxy-buttersäure-p- phenetidid als Ö1 erhalten, das nach mehrfachem Auskochen mit Wasser zur Kristallisation gebracht wird.
Der Schmelzpunkt des aus einem Gemisch aus Toluol und Petroläther umkristallisierten Produktes beträgt 99 . b) 720 g a-Amino-B-oxy-buttersäufe-p-phenetidid 340 g 100 /oige Ameisensäure, 540 cm3 40"/oigne wässrige Formaldehydlösung und 3 1 Wasser werden gemeinsam fünf Stunden auf dem Dampfbad erhitzt, wobei lebhafte Kohlendioxyd-Entwicklung auftritt.
Anschliessend wird das Reaktionsgemisch durch Zugabe von verdünnter Salzsäure gerade kongosauer gestellt. Nach Filtrieren und Klären mit Tierkohle wird das Filtrat mit verdünnter Natronlauge eben alkalisch gemacht. Man erhält unmittelbar 648 g (80 /o der Theorie) a-Dimethylamino-ss-oxy-butter- säure-p-phenetidid vom Schmelzpunkt 98 . c) 147 g a-Amino--oxy-buttersäure-p-phenetidid, 68 g 100 /oige Ameisensäure, 108 g wässrige 40 /oige Formaldehydlösung und 300 cm3 Benzol werden gemeinsam etwa 3 Stunden auf dem Dampfbad unter Rückfluss erhitzt. Nach dem Abkühlen werden dem Reaktionsgemisch eine Mischung von 50 cm3 konzentrierter Salzsäure und 400 cm3 Wasser zugesetzt.
Aus dem 2-Phasen-System wird die wässrige Schicht abgetrennt und mit verdünnter Natronlauge alkalisch gestellt. Dabei fällt das a-Dimethylamino-ss-oxy-but- tersäure-p-phenetidid mit guter Ausbeute aus. Der Schmelzpunkt liegt nach Umkristallisieren aus Essigsäureäthylester bei 98 . d) Die gleiche Verbindung kann aus a-Amino-ss- oxy-buttersäure-p-phenetidid auch durch katalytische Hydrierung in Gegenwart von Formaldehyd erhalten werden.
Beispiel 2 26, 6 g a-Amino-ss-oxy-buttersäure-p-carbäthoxy- anilid werden mit 13,5 g Ameisensäure und 16 g 40 /o ige Formaldehydlösung eine Stunde auf dem Dampfbad erwärmt. Nach Abkühlen und Versetzen mit überschüssiger Natronlauge scheidet sich das a-Dimethylamino-ss-oxy-buttersäure- (p-carbäthoxy- anilid) in öliger Form ab, welches in Ather aufgenommen wird und nach Trocknen und Abdestillieren des Lösungsmittels kristallisiert. Schmelzpunkt 85 nach Umkristallisieren aus einer Mischung von Essigester und Petroläther.
Die Herstellung des als Ausgangsstoff verwendeten a-Amino-ss-oxy-buttersäure-p-carbäthoxy-anilids wurde auf folgendem Wege vorgenommen :
Durch Umsetzung von Diketen mit p-Aminobenzoesäureäthylester wurde Acetessigsäure- (p-carb- äthoxy-anilid) erhalten, welches über das a-Isonitroso acetessigsäure- (p-carbäthoxy-anilid) durch Hydrieren der Isonitroso-Verbindung in das a-Amino-ss-oxy- buttersäure- (p-carbäthoxy-anilid) überführt wurde.
Beispiel 3
26,4 g a-Amino-ss-oxy-buttersäure-6-methoxy- 1,2,3,4-tetrahydrochinolid werden mit 13,5 g Ameisensäure und 16 g 40 /oiger Formaldehydlösung entsprechend der in Beispiel 1 angegebenen Vorschrift umgesetzt. Es werden 20,5 g a-Dimlethylamino-ss-oxy buttersäure-(6-methoxy-1, 2,3,4-tetrahydrochinolid) er halten. Schmelzpunkt 97,5 nach Umkristallisieren aus einer Mischung von Essigester und Petroläther.
Die Herstellung des als Ausgangsstoff verwendeten a-Amino-ss-oxy-buttersäure-6-methoxy-1, 2,3,4 tetrahydrochinolids wurde auf folgendem Weg vor genommen :
Durch Umsetzung von 6-Methoxy-1, 2,3,4-tetra hydrochinolin mit Diketen wird Acetessigsäure- 1, 2,3,4-tetrahydro-6-methoxychinolid erhalten, dieses durch Nitrosierung und Reduktion in die Isonitroso- Verbindung und letztere durch Hydrierung in a-Amino ss-oxy-buttersäure-6-methoxy-1, 2,3,4-tetrahydrochinolid übergeführt.
Beispiel 4
26,5 g a-Amino-ss-oxy-buttersäure-(N-methyl-p- phenetidid) werden mit 26,5 cm3 40 /oiger Formal- dehydlösung versetzt. Die klare Lösung wird mit
13 cm3 85 /oiger Ameisensäure 45 Minuten auf dem Dampfbad erhitzt. Nach Abkühlen und Versetzen mit überschüssiger Kaliumcarbonatlösung fällt a-Di methylamino-ss-oxy-buttersäure- (N-methyl-p-phenetidid) in öliger Form aus und wird in Äther aufgenommen. Nach Trocknen und Verdampfen des Lö sungsmittels tritt nach einigem Stehen Kristallisation ein. Ausbeute 18 g ; Schmelzpunkt 82 .
Die Herstellung des a-Amino-ss-oxy-buttersäure- (N-methyl-p-phenetidids) erfolgte auf nachstehendem Wege :
N-Methyl-p-phenetidin wird mit Diketen in Acet essigsäure-N-methyl-p-phenetidid und dieses nach Nitrosierung in üblicher Weise in die Isonitroso-Verbindung des Acetessigsäure-N-methyl-p-phenetidids übergeführt, aus der durch Hydrierung das a-Amino ss-oxy-buttersäure- (N-methyl-p-phenetidid) entsteht.
Beispiel 5
21,5 g a-Amino-ss-oxy-buttersäure-(N-n-butyl-p- anisidid) werden mit 21,5 cm3 40 /o iger Formaldehyd lösung und 21,5 cm3 Wasser unter Schütteln kurz erwärmt. Es bildet sich ein 61, das nach einigen Minuten wieder erstarrt. Nach Zugabe von 11 cm3 85"/oiger Ameisensäure wird noch 30 Minuten auf dem Dampfbad erhitzt. Nach Versetzen mit Wasser und Filtration wird mit Kaliumcarbonatlösung alkalisch gemacht. Das ausgefallene O1 (Rohausbeute 22,5 g) erstarrt sofort. Nach Umkristallisieren aus Cyclohexan wird das a-Dimethylamino--oxy-butter- säure- (N-n-butyl-p-anisidid) vom Schmelzpunkt 101 erhalten.
Die Herstellung des a-Amino-ss-oxy-buttersäure- (N-n-butyl-p-anisidids) erfolgte auf folgendem Wege : N-n-Butyl-acetessigsäure-p-anisidid, erhalten aus
N-n-Butyl-p-anisidin und Diketen, ergibt nach Nitro sierung das Isonitroso-acetessigsäure-N-n-butyl-p- anisidid, welches durch Hydrierung, in das a-Amino ss-oxy-buttersäure- (N-n-butylanisidid) übergeführt wird.
Beispiel 6
35 g a-Amino-ss-oxy-buttersäure-p-chloranilid werden mit 21 g Ameisensäure, 35 cm3 Wasser und 25,4 g 40 /Oiger wässriger Formaldehydlösung eine Stunde am Rückfluss gekocht. Die zunächst getrübte Lösung klärt sich nach Abkühlen und wird mit ver dünnter Natronlauge alkalisch gemacht. Das abgeschiedene 01 erstarrt bald und wird abgesaugt. Nach Umkristallisieren aus einer Mischung von Essigsäure- äthylester und Petroläther wird das a-Dimethylamino- ss-oxy-buttersäure-p-chloranilid vom Schmelzpunkt 90-91 erhalten.
Die Herstellung der Ausgangsverbindung erfolgte auf nachstehend beschriebenem Wege :
Acetessigsäure-p-chloranilid, erhalten aus p-Chloranilin und Diketen, ergibt nach Nitrosierung das a-Isonitroso-acetessigsäure-p-chloranilid vom Schmelzpunkt 177-178 , welches durch Hydrierung in das a-Amino-ss-oxy-buttersäure-p-chloranilid vom Schmelzpunkt 126-127 übergeführt wird.
Beispiel 7
109 g a-Amino-ss-oxy-buttersäure-3-chlor-4-me- thyl-anilid werden mit 450 cm3 Wasser, 51,5 g 100 /o iger Ameisensäure und 82 g 40 böiger wäss- riger Formaldehydlösung vier bis fünf Stunden auf dem Dampfbad erhitzt. Nach Abkühlen wird das Reaktionsgemisch mit 37 cm3 konzentrierter Salzsäure versetzt, wobei weitgehend Lösung eintritt.
Nach Verdünnen mit 1,5 Liter Wasser wird filtriert, mit Natronlauge alkalisch gemacht, worauf nach Stehen in Eis 88 g a-Dimethylamino-yS-oxy-butter- säure-3-chlor-4-methyl-anilid erhalten wird. Schmelzpunkt 101-102 nach Umkristallisieren aus einer Mischung von Essigester und Petroläther.

These starting materials can be obtained, for example, by treating correspondingly substituted acetoacetic anilides with nitrous acid and reducing both the isonitroso group and the keto group in the nitrosation products obtained in the customary manner. Formaldehyde is best used in an aqueous solution. However, it is also possible to work in the presence of inert solvents, for example benzene, toluene or xylene. If formic acid is used, it can be advantageous to carry out the reaction by heating to moderately elevated temperatures, advantageously to the boiling point of the solvent used, and to heat the reaction mixture on the steam bath for some time.
The process according to the invention gives crystalline compounds which, depending on the substituents present, are more or less soluble in water. They are valuable remedies and, in particular, have a good analgesic and sedative effect with remarkably low toxicity. As a result of the large therapeutic breadth associated with this, they are superior to known compounds.
For example, the analgesic effect of α-dimethylamino-ss-oxy-butyric acid-p-phenetidide, from which a 1'Voigc aqueous solution can be prepared, corresponds to that of the known, practically water-insoluble N-acetyl-p-phenetidine. In addition, the a-dimethylamino-ss-oxy-butyric acid-p-phenetidide has a sedative component and furthermore shows an anti-inflammatory effect which corresponds to that of the above known compound.
When used as a medicinal product, however, the significantly lower toxicity of α-dimethylamino-ß-oxy-butyric acid-p-phenetidide is of decisive importance compared to the above known compound. For oral administration to the mouse, the Dos is. let. min. 3 g / kg, while that of the known compound is 1.5 g / kg. In the rat the same data are 9 glkg and 5 glkg.
Studies of the chronic toxicity in cats showed that 3 out of 4 animals tolerated 27 feedings with 0.25 g of a-dimethylamino-ssoxy-butyric acid p-phenetidide per kg without any problems; only one animal died after 14 feedings. Application of the same dose of the known compound resulted in death in almost all animals after 3 to 4 feedings.
Taking into account the improved absorbability associated with the better solubility of the a-dimethylamino-oxy-butyric acid-p-phenetidide obtained according to the invention compared to the above-mentioned known compound, the advantages offered by the compound obtained according to the invention with regard to toxicity , even clearer.
Example 1 a) 23.8 g of α-amino-β-oxy-butyric acid-pJphenetidid 13.5 g of formic acid and 16 g of 40% formaldehyde solution are heated for one hour on the steam bath, during which vigorous evolution of carbon dioxide occurs. After recrystallization from ethyl acetate, the melting point is 98. d) The same compound can also be obtained from a-amino-ss- oxy-butyric acid-p-phenetidide by catalytic hydrogenation in the presence of formaldehyde.
Example 2 26.6 g of α-amino-β-oxy-butyric acid-p-carbethoxyanilide are heated for one hour on the steam bath with 13.5 g of formic acid and 16 g of 40% formaldehyde solution. After cooling and adding excess sodium hydroxide solution, the a-dimethylamino-ss-oxy-butyric acid (p-carbethoxyanilide) separates out in oily form, which is taken up in ether and crystallizes after drying and distilling off the solvent. The a-amino-ss-oxy-butyric acid-p-carbethoxy-anilide used as starting material was prepared in the following way:
By reacting diketene with ethyl p-aminobenzoate, acetoacetic acid (p-carbethoxyanilide) was obtained, which via the α-isonitroso acetacetic acid (p-carbethoxyanilide) by hydrogenation of the isonitroso compound into the α-amino-ss -oxy-butyric acid- (p-carbethoxy-anilide) was transferred.

26.4 g of α-amino-β-oxy-butyric acid-6-methoxy-1,2,3,4-tetrahydroquinolide are reacted with 13.5 g of formic acid and 16 g of 40% formaldehyde solution in accordance with the instructions given in Example 1. N-methyl-p-phenetidine is converted into acetoacetic acid-N-methyl-p-phenetidide with diketene and this after nitrosation in the usual way into the isonitroso compound of acetoacetic acid-N-methyl-p-phenetidide, from which by hydrogenation the a-Amino ss-oxy-butyric acid (N-methyl-p-phenetidide) is formed.
Example 5
21.5 g of a-amino-ss-oxy-butyric acid (N-n-butyl-p-anisidide) are briefly heated with 21.5 cm3 of 40% formaldehyde solution and 21.5 cm3 of water while shaking. A 61 forms, which solidifies again after a few minutes. After adding 11 cm3 of 85% formic acid, the mixture is heated on the steam bath for a further 30 minutes. After adding water and filtration, it is made alkaline with potassium carbonate solution. The precipitated O1 (crude yield 22.5 g) solidifies immediately. After recrystallization from cyclohexane, a-Dimethylamino-oxy-butyric acid- (Nn-butyl-p-anisidide) with a melting point of 101 obtained.
The a-amino-ss-oxy-butyric acid (N-n-butyl-p-anisidide) was prepared in the following way: N-n-butyl-acetacetic acid-p-anisidide, obtained from
N-n-butyl-p-anisidine and diketene, after nitro-sation, gives the isonitroso-acetacetic acid-N-n-butyl-p-anisidide, which is converted by hydrogenation into the a-amino-ss-oxy-butyric acid (N-n-butyl anisidide).
Example 6
35 g of a-amino-ß-oxy-butyric acid-p-chloroanilide are refluxed for one hour with 21 g of formic acid, 35 cm3 of water and 25.4 g of 40% aqueous formaldehyde solution. The initially cloudy solution clears after cooling and is made alkaline with dilute sodium hydroxide solution. The separated oil soon solidifies and is sucked off. After recrystallization from a mixture of ethyl acetate and petroleum ether, the α-dimethylamino-s-oxy-butyric acid-p-chloroanilide with a melting point of 90-91 is obtained.
The starting compound was prepared as follows:
Acetoacetic acid-p-chloroanilide, obtained from p-chloroaniline and diketene, gives after nitrosation the a-isonitroso-acetic acid-p-chloroanilide with a melting point of 177-178, which by hydrogenation into the a-amino-ss-oxy-butyric acid-p- chloranilide with a melting point of 126-127.
Example 7
109 g of a-amino-ss-oxy-butyric acid-3-chloro-4-methyl anilide are mixed with 450 cm3 of water, 51.5 g of 100% formic acid and 82 g of 40 g of aqueous formaldehyde solution Heated for hours on the steam bath. After cooling, 37 cm3 of concentrated hydrochloric acid are added to the reaction mixture, largely dissolving.
After dilution with 1.5 liters of water, the mixture is filtered and made alkaline with sodium hydroxide solution, whereupon 88 g of α-dimethylamino-γS-oxy-butyric acid-3-chloro-4-methyl-anilide are obtained. Melting point 101-102 after recrystallization from a mixture of ethyl acetate and petroleum ether.
