Procédé de dédoublement de la DL-sérin
La présente invention est relative à un procédé de dédoublement de la DL-sérine, compose correspondant à la formule (I). La notation DL désigne la serine racémique, mélange en proportions égales de L-sérine et de son énantiomorphe, la D-sérine.

La serine est un acide a-aminé p-hydroxylé qui intervient sous sa forme lévogyre (L-sérine) dans le métabolisme animal. Elle participe, en outre, sous ses deux formes D et L, à la structure de divers polypeptides : la forme D, par exemple, dans certaines polymyxines ; la forme L dans la viomycine, I'hypertensine,
I'azasérine et de nombreux polypeptides d'origines diverses.
Les méthodes de synthèse de la serine conduisant au racémique, il est nécessaire de pouvoir disposer d'un procédé de dédoublement facile et peu onéreux conduisant rapidement aux D-et L-sérines optiquement pures, susceptibles d'être utilisées, par exemple, dans des synthèses de polypeptides contenant ces amino-acides.
La seule méthode de dédoublement de la DL-sérine actuellement connue fait intervenir, comme intermédiaires, le sel de quinine de la N-p-nitrobenzoyl-D-sérine et le sel de brucine de la N-p-nitrobenzoyl-L-sérine (cf. E. Fischer et Jacobs, Ber. dtsch. chem. Ges., 1906, 39, 2942). Ce procédé de dédoublement est onéreux.
Dans ses brevets suisses Nos 319787, 327724 et 330126, la société titulaire a décrit l'utilisation des D (-)- et L (-)-threo-l-p- nitrophényl-2-amino-propanediols- (1, 3) dans des procédés de dédoublement du DL-tryptophane, de la DL-proline et de la DL-hydroxyproline. Or, il a été trouvé que la DL-sérine peut être aussi dédoublée, sous forme de son dérivé N-3, 5-dinitrobenzoylé (II), en utilisant ces réactifs.
Le procédé suivant la présente invention est caractérisé en ce qu'on fait réagir une quantité sensiblement hémi-moléculaire de
D (-)- ou L (-)-threo-l-p-n, itrophenyl-2- amino-propanediol- (1, 3) sur la N-3, 5-dinitro benzoyl-DL-sérine au sein d'un solvant, l'un des deux énantiomorphes donnant un sel peu soluble avec le réactif de dédoublement, on sépare ce sel, on en libère le dérivé N-3, 5dinitrobenzoylé de la D-ou L-serine par traitement alcalin suivi d'acidification, et on soumet le dérivé N-3, 5-dinitrobenzoylé de la Dou L-sérine à une hydrolyse acide.
Les sels des N-3, 5-dinitrobenzoyl-D- et -L-sérines avec les D (-)- et L (+)-threo-l-p- dinitrophényl-2-amino-propanediols- (1, 3) sont peu solubles et précipitent à l'état cristallin.
On peut encore améliorer le rendement de leur séparation en ajoutant au mélange réactionnel une quantité variable d'un solvant non miscible à 1'eau tel que l'acétate d'éthyle ou l'éther.
Dans chaque cas, le traitement alcalin du précipité cristallin suivi d'acidification conduit, après récupération presque quantitative du réactif de dédoublement, à la forme optiquement active recherchée de la N-3, 5-dinitro benzoyl-sérine qui fournit la serine correspondante par hydrolyse acide et sans racémisation notable. A partir des liqueurs d'où a cristallisé le sel, on sépare de la même manière l'énan- tiomorphe correspondant qui est, en général, souillé par de faibles quantités de racémique.
Cet énantiomorphe peut être, soit purifié par recristallisation dans un solvant approprié ou par précipitation par le thréo-l-p-nitrophényl- 2-amino-propanediol- (1, 3) correspondant, soit racémisé directement par un procédé connu, ou par traitement alcalin de la serine libre correspondante selon Crawhall et Elliott (Bio- chem. J. 1951, 48, 237) et adjoint à une nouvelle venue de racémate à dédoubler. Par le jeu des dédoublements et des racémisations successives, on transforme ainsi la totalité du racémate primitivement présent en énantio- morphe désiré.
Les D (-)- et L (+)-threo-l-p-nitrophényl- 2-amino-propanediols- (1, 3), intermédiaires de la synthèse du chloramphénicol, peuvent être obtenus selon le procédé décrit par Velluz,
Amiard et Joly (Bull. Soc. Chim. 1953, p. 342).
Ils présentent la structure (III). Leurs carac téris, tiques sont les suivantes : F. 162-163 C ; [a] 2E0, de la base D =280 20 (c = 2 /0, acide chlorhydrique 0, 1 N ; [a] 2D de la base
L = + 28¯ ¯ 2¯ (mÛmes conditions).

Les exemples qui suivent illustrent l'inven- tion. On peut notamment effectuer la N-3, 5dinitrobenzoylation par toutes les méthodes connues ou encore faire réagir le chlorure de 3, 5-dinitrobenzoyle sur un ester, un éther ou un autre dérive de la serine d'où l'on peut ensuite régénérer facilement l'aminoacide Nacylé. On peut également, lors de la préparation des sels de D (-)- ou de L (+)-thréo-1-p- nitrophényl-2-aminopropanediol- (1, 3), faire varier la nature du solvant, les températures de dissolution et de cristallisation et le temps de refroidissement.
Exemple 1 :
Préparation de la N-3, S-dinitrobenzoyl-DL-sérine
On dissout 6, 3 g de DL-sérine dans 60 cm3 de soude N, refroidit et ajoute 6, 9 g de chlorure de 3, 5-dinitrobenzoyle. On introduit encore 10 g de chlorure de 3, 5-dinitrobenzoyle en alcalinisant le milieu par addition de soude N (75 cm3). On ajoute 15 cm3 d'acide chlorhydrique, refroidit, essore, lave à l'éther pour éliminer l'acide dinitrobenzoïque et sèche.
On obtient ainsi 16, 7 g (93 /o) de N-3, 5 dinitrobenzoyl-D L-serine. Le monohydrate fond à 100-101 C (sur bloc).
Dedoublement et preparation des D-et L-serines a) Formation du sel de la N-3, 5-dinitro-
benzoyl-L- sÚrine et du L (+)-thréo-l-p- nitrophényl-2-amino-propanedíol-(1, 3).
Première forme d'exécution
On dissout, à 600 C, 10 g de N-3, 5-dinitrobenzoyl-DL-sérine dans 30 cm3 de mé thanol et ajoute 4, 2 g de L (+)-thréo-1-p- nitrophényl-2-amino-propanediol- (1, 3). Par refroidissement à + 10 C, on obtient 6, 8 g (80 /o) de sel optiquement pur de la N-3, 5 dinitrobenzoyl-L-sérine et du L (+)-thrÚo-1-pnitrophényl-2-amino-propanediol- (1, 3), F. 160 161¯C, [α]20 D = + 33¯ ¯ 1¯ (c = 1 /o, eau).
Ce produit est nouveau.
Deuxième forme d'exÚcution
On dissout, à 600 C, 10 g de N-3, 5-dinitrobenzoyl-DL-sérine dans 5 cm3 de méthanol et ajoute 4, 2 g de L (-)-threo-l-p-nitrophényl- 2-amino-propanediol- (1, 3). On introduit alors dans le mélange réactionnel 55 cm'd'acétate d'éthyle. On refroidit et essore le sel formé qu'on lave et sèche. On obtient ainsi 8, 2 g (96, 5 /o) de sel optiquement pur de la N-3, 5 dinitrobenzoyl-L-sérine et du L (+)-thrÚo-1-Pnitrophényl-2-amino-propanediol- (1, 3). b) Obtention de la N-3, 5-dinitrobenzoyl-L-
serine
On dissout 8, 2 g du sel ci-dessus dans 16, 5 cm3 de soude N. Le L (+)-thréo-1-p- nitrophÚnyl-2-amino-propanediol-(1, 3) qui cristallise est essoré et lavé.
On récupère ainsi 3, 1 g (74 %) du rÚactif de dédoublement.
Après avoir acidifié le filtrat par 1, 7 cm3 d'acide chlorhydrique concentré, la N-3, 5-di nitrobenzoyl-L-serine cristallise. On essore, lave et sèche. On obtient ainsi 4, 5 g (90 /0 par rapport à la N-3, 5- dinitrobenzoyl-DL-sÚrine mise en oeuvre) de produit cristallisé. Monohydrate : F. 110-112¯ C, [α]20 D = + 23,5¯ ¯ 1¯ (c = 1 %, Úthanol Ó 50 %). c) Séparation de la L-sérine
On chauffe pendant une heure au reflux une solution de 4, 3 g de N-3, 5-dinitrobenzoyl L-sérine dans 60 cm3 d'acide chlorhydrique 5 N. Après filtration, on évapore à sec sous vide et cristallise dans l'acétone le chlorhydrate de L-sérine obtenu.
On reprend le produit par 1 cm3 d'eau, ajoute 1, 4 cm3 d'ani- line puis 10 cm3 d'alcool absolu. On obtient après refroidissement 1, 3 g (90 /o) de L-sérine cristallisée optiquement pure, F. 228¯ C, [a] 20 = + 150 + lo (c = 4 /o, acide chlorhydri- que N). d) Séparation de la D-sérine
On extrait la solution d'acétate d'éthyleméthanol d'où a cristallisé le sel de N-3, 5-di nitrobenzoyl-L-sérine et du L (+)-thréo-1-p- nitrophényl-2-amino-propanediol- (1, 3) (exemple 2, deuxième forme d'exécution) par 10 cm3 d'acide chlorhydrique 0, 5 N, puis par 5 cm3 d'eau.
Les solution aqueuses sont réunies puis alcalinisées par de la soude N. On récupère ainsi 0, 4 g (10 /o) du réactif de dédoublement.
La phase organique est alors concentrée et additionnée d'éther de pétrole. On sépare ainsi 4, 85 g (97 O/o par rapport à la N-3, 5-dinitro benzoyl-DL-sérine mise en oeuvre) de N-3, 5 dinitrobenzoyl-D-sérine qui peut être facilement purifiée par recristallisation. Le produit brut fond à 109-111¯ C, [a] 20 D = - 22¯ lo (c = 1 %, Úthanol Ó 50 %). On obtient la D-sérine correspondante par hydrolyse chlorhydrique comme il est indiqué en c) pour la L-sérine.
Exemple 2 :
On traite la N-3, 5-dinitrobenzoyl-DL-sÚrine par le D(-)-thrÚo-1-p-nitrphÚnyl-2-amino propanediol- (1, 3) comme il est indiqué en a) de 1'exemple 1. On obtient ainsi le sel de la
N-3, 5-dinitrobenzoyl-D-sÚrine et du D (-)- threo-1-p-nitrophényl-2-amino-propanediol- (1, 3), F. 160-161¯C, [α]20 D= - 33¯ ¯ 1¯ (c = 1 /o, eau). Ce produit est nouveau.
Il conduit, comme dans l'exemple 1 b), Ó la
N-3, 5 - dinitro - benzoyl - D - sÚrine optiquement pure, monohydrate : F. 110-112 C, [α]20 D= -23, 50 lo (c = 1 /o, éthanol 50 /o), puis, suivant l'exemple 1 c), à la D-sérine, [a] 2D0 = -150 lo (c = 4 /o, acide chlorhydrique N).
La N-3, 5-dinitrobenzoyl-D-sérine n'a pas encore été décrite.
DL-serin resolution process
The present invention relates to a process for resolving DL-serine, a compound corresponding to formula (I). The notation DL denotes racemic serine, a mixture of equal proportions of L-serine and its enantiomorph, D-serine.

Serine is a p-hydroxylated α-amino acid which is involved in its levorotatory form (L-serine) in animal metabolism. It also participates, in its two forms D and L, in the structure of various polypeptides: the D form, for example, in certain polymyxins; the L form in viomycin, hypertensin,
Azaserine and many polypeptides of various origins.
The methods of synthesis of serine leading to the racemic, it is necessary to be able to have an easy and inexpensive resolution process leading rapidly to optically pure D- and L-serines, capable of being used, for example, in syntheses of polypeptides containing these amino acids.
The only currently known method for resolving DL-serine involves, as intermediates, the quinine salt of Np-nitrobenzoyl-D-serine and the brucine salt of Np-nitrobenzoyl-L-serine (cf. E. Fischer and Jacobs, Ber. Dtsch. Chem. Ges., 1906, 39, 2942). This duplication process is expensive.
In its Swiss Patents Nos. 319787, 327724 and 330126, the proprietor described the use of D (-) - and L (-) - threo-lp-nitrophenyl-2-amino-propanediols- (1, 3) in processes for resolving DL-tryptophan, DL-proline and DL-hydroxyproline. However, it has been found that DL-serine can also be resolved, in the form of its N-3, 5-dinitrobenzoyl derivative (II), using these reagents.
The process according to the present invention is characterized in that a substantially half-molecular amount of
D (-) - or L (-) - threo-lpn, itrophenyl-2-amino-propanediol- (1, 3) on N-3, 5-dinitro benzoyl-DL-serine in a solvent, l 'one of the two enantiomorphs giving a sparingly soluble salt with the resolving reagent, this salt is separated, the N-3, 5dinitrobenzoyl derivative of D- or L-serine is released from it by alkali treatment followed by acidification, and subjected the N-3, 5-dinitrobenzoyl derivative of Dou L-serine to acid hydrolysis.
The salts of N-3, 5-dinitrobenzoyl-D- and -L-serines with D (-) - and L (+) - threo-lp- dinitrophenyl-2-amino-propanediols- (1, 3) are little soluble and precipitate in the crystalline state.
The yield of their separation can be further improved by adding to the reaction mixture a varying amount of a water-immiscible solvent such as ethyl acetate or ether.
In each case, the alkaline treatment of the crystalline precipitate followed by acidification leads, after almost quantitative recovery of the resolution reagent, to the desired optically active form of N-3, 5-dinitro benzoyl-serine which provides the corresponding serine by hydrolysis. acidic and without noticeable racemization. From the liquors from which the salt has crystallized, the corresponding enantiomorph is separated in the same way, which is generally contaminated with small amounts of racemic.
This enantiomorph can be either purified by recrystallization from an appropriate solvent or by precipitation with the corresponding threo-lp-nitrophenyl-2-amino-propanediol- (1, 3), or racemized directly by a known method, or by alkali treatment of the corresponding free serine according to Crawhall and Elliott (Biochem. J. 1951, 48, 237) and added to a new occurrence of racemate to be split. By the interplay of successive duplications and racemizations, the totality of the racemate originally present is thus transformed into the desired enantiomorph.
D (-) - and L (+) - threo-l-p-nitrophenyl- 2-amino-propanediols- (1, 3), intermediates in the synthesis of chloramphenicol, can be obtained according to the process described by Velluz,
Amiard and Joly (Bull. Soc. Chim. 1953, p. 342).

A solution of 4.3 g of N-3, 5-dinitrobenzoyl L-serine in 60 cm3 of 5 N hydrochloric acid is heated for one hour at reflux. After filtration, evaporated to dryness in vacuo and crystallized from acetone. the L-serine hydrochloride obtained.
The product is taken up in 1 cm3 of water, 1.4 cm3 of aniline and then 10 cm3 of absolute alcohol are added. After cooling, 1.3 g (90 / o) of optically pure crystallized L-serine, mp 228¯ C, [a] 20 = + 150 + lo (c = 4 / o, N hydrochloric acid) are obtained. d) Separation of D-serine
The ethylmethanol acetate solution is extracted from which crystallized the salt of N-3, 5-di nitrobenzoyl-L-serine and L (+) - threo-1-p-nitrophenyl-2-amino-propanediol. - (1, 3) (example 2, second embodiment) with 10 cm3 of hydrochloric acid 0, 5 N, then with 5 cm3 of water.
The aqueous solutions are combined and then basified with N sodium hydroxide. 0.4 g (10 / o) of the resolution reagent is thus recovered.
The organic phase is then concentrated and added with petroleum ether. 4.85 g (97 O / o relative to the N-3, 5-dinitro benzoyl-DL-serine used) are thus separated from N-3, 5 dinitrobenzoyl-D-serine which can be easily purified by recrystallization. . The crude product melts at 109-111¯ C, [a] 20 D = - 22¯ lo (c = 1%, Úthanol Ó 50%). The corresponding D-serine is obtained by hydrochloric hydrolysis as indicated in c) for L-serine.
Example 2:
N-3, 5-dinitrobenzoyl-DL-sÚrine is treated with D (-) - thrÚo-1-p-nitrphÚnyl-2-amino propanediol- (1, 3) as indicated in a) of example 1. We thus obtain the salt of
N-3, 5-dinitrobenzoyl-D-sÚrin and D (-) - threo-1-p-nitrophenyl-2-amino-propanediol- (1, 3), F. 160-161¯C, [α] 20 D = - 33¯ ¯ 1¯ (c = 1 / o, water). This product is new.
It leads, as in example 1 b), to the
N-3, 5 - dinitro - benzoyl - D - optically pure sÚrin, monohydrate: F. 110-112 C, [α] 20 D = -23, 50 lo (c = 1 / o, ethanol 50 / o), then, according to Example 1 c), with D-serine, [a] 2D0 = -150 lo (c = 4 / o, N hydrochloric acid).
N-3, 5-dinitrobenzoyl-D-serine has not yet been described.
