Verfahren zur Herstellung von p-Amino-methyl-benzoesäure oder ihren Estern Die Erfindtmg bet-rifft ein Verfahren zur IIerstellung von p-Amino-methyl-benzoesÏure oder ihren Estern.
Es ist bekannt, p-Amino-methyl-benzoe- sÏure dureh Erhitzen von N- (p-carboxy-ben- zvl)-phthalamidsäure auf 200 mit konzentrier ter Salzsäure im Druekrohr herzustellen. Man erhält dann aus der gebildeten SÏure durch Veresterung mit z. B. Methylalkohol den p Amino-methyl-benzoesÏuremethylester. Dieses Verfahren, das unter Druck arbeitet, ergibt wenig gute Ausbeuten und ist für die technische Herstellung von grosseren Mengen sehr umständlich.
Es wurde gefunden, dass man die p-Amino- methyf-benzoesäuren oder ihre Ester auf einfachem Wege und in guter Ausbeute herstellen kann, wenn man eine p-Halogen-methyl-ben- zoesäure oder ihre Ester unter Beachtung be sonderer Massnahmen mit flüssigem Ammoniak umsetzt. Man verwendet hierzu ein be sonders sorgfältig gereinigtes, flüssiges Ammoniak im ¯berschu¯ und lässt dies in einem Druckgefϯ bei Temperaturen von 0-30¯, vorzugsweise bei 10-15 , wÏhrend einer Reaktionszeit von l-2 Stunden auf das Aus gangsprodukt, z. B. p-Halogen-methyl-benzoesÏureester, einwirken.
Es stellt, sich bei Ver wendung des flüssigen Ammoniaks und bei Einhaltung oben genannter Temperatur im Reaktionsgefϯ ein durchschnittlicher Druck von 8-10 at ein. Es ist auch möglich, zunächst unter den gleichen Bedingungen Ammoniak auf p-Chlor-methyl-benzoesäure einwirken zu lassen, um dann anschliessend erst die p-Amino-methyl-benzoesäure mit z. B. Methylaikohol zu verestern. -w rde man dagegen in beiden FÏllen bei hoheren Temperaturen als angegeben arbeiten, so würde man das ent spreeliende p-Amino-methyl-benzoesäureamid (Fp etwa 118/120 ) erhalten. Anderseits ist es auch möglich, aus dem jeweils gebildeten Ester durch vorsichtiges Verseifen mit konzentrierter Salzsäure in der Kälte wiederum die freie SÏure selbst zu erhalten.
Das Ausgangsprodukt, z. B. p-Chlor-me- thyl-benzoesäure-methylester, ist leicht zugÏnglich. Man. kann es aus p-Xylylendiehlorid herstellen, wenn man dieses Dichlorid mit z. B.
50"/oiger Salpetersäure bei Temperaturen um 100 halbseitig oxydiert, das halbseitig oxy- dierte Beaktions. prod. ukt abtrennt und die p-Chlor-methyl-benzoesäure mit Methylalkohol verestert.
Sowohl der gebildete Ester als auch die SÏure können für die Herstellung von plasti schen Massen verwendet werden. Es können naeh diesem Verfahren Ester aus vorwiegend einwertigen Alkoholen hergestellt werden, wie z. B. p-Amino-methyl-benzoesÏuremethylester, -Ïthylester, -buitylester oder-amylester. Es ist aber auch möglich, diprimäre Alkohole, wie z. B. Glycol, zur Veresterung zu verwenden.
Beispiel 1
Das handelsübliehe fliissige Ammoniak wird zunÏchst filtriert, um die darin enthaltenen Spuren von z. B. Eisen oder Kupfer zu beseitigen. Danach lässt man 50 g p-Chlor- methyl-benzoesÏuremethylester mit etwa 250 g fl ssigem, gereinigtem Ammoniak 1 1/2 Stunden bei einer Temperatur von 20¯ in einem Autoklaven, in dem sich ein Druck von 8 bis 10 atü eingestellt hat, reagieren. Das bersch ssige Ammoniak wird nach Beendigung der Reaktion abgedampft und das Umsetzungsprodukt mit Methanol und anschliessend mit Wasser ausgewaschen, um insbesondere das während der Reaktion gebildete Ammonium- ehlorid zu beseitigen.
Das getrocknete Reak- tionsprodukt p-Amino-methyl-benzoesÏureme thylester fällt in einer Menge von 38 g in farblosen Kristallen vom Schmelzpunkt 176 an. Die Ausbeute beträgt 86, 3 %.
B eispiel 2
50 g p-Chlor-methyl-benzoesäure werden mit etwa 200 g flüssigem Ammoniak etwa 2 Stunden in einem Autoklaven bei Raumtempe- ratur entspreehend Beispiel 1 zur Reaktion gebracht. Nach dem Verdmusten des überschüssigen Ammoniaks und Auswasehen mit zunächst wässrigem Ammoniak, dann mit Me thanol, erhält man die p-Amino-methyl-benzoesaure, die in farblosen Kristallen vom Schmelz- punkt 234 anfällt, welche anschlie¯end mit Methylalkohol zu p-Amino-methyl-benzoesäuremethylester umgesetzt wird. Schmelzpunkt 176 .
Beispiel 3
50 g p-Brom-methyl-benzoesäure werden mit etwa 300 g flüssigem Ammoniak entspre- chend Beispiel 2 behandelt. Es resultiert p Amino-methyl-benzoesÏure, die einen Schmelzpunkt von. 234¯ aufweist. Die Umsetzung dieser SÏure mit Äthylalkohol ergibt den p Amino-methyl-benzoesÏureÏthylester.
Beispiel 4
50 g Chlor-methyl-benzoesäureätlyester lässt man mit etwa 200 g fl ssigem, sorgfÏltig gereinigtem Ammoniak entsprechend Beispiel 1 reagieren. Es restlltiert der p-Amino-metivl- benzoesäureäthylester, der einen Schmelzpunkt von 267-268 aufweist.

It has been found that the p-aminomethylbenzoic acids or their esters can be produced in a simple way and in good yield if a p-halomethylbenzoic acid or its esters are used with liquid ammonia, taking special measures implements.
