Verfahren zur Darstellung von N, N'- (4, 4'-Dicarboxy-3, 3'-dioxy)-diphenyl-harnstoff und dessen Salzen
Die vorliegendle Erfin-dung beziehtsiehauf ein Verfahren zur herstellung von N, N'-(4,4' Dicarboxy-3,3'-dioxy)-diphenyl-harnstoff und dessen Salzen, das dadruch gekennzichnet, ist, dass 4-Amino-salicylisaure mit einem reaktions- fägigen Derivat der Kohelnsäure behandelt wird.
Trotz der starken tuberkulostatischen Wirksamkeit der p-Amino-salicylsäure in vitro benötigt man infolge ihrer sehnellen Ausschei- dung aus dem Organismus zur Therapie eine hohe Dosierung,um im Blut eine wirksame Konzentration zu erzielen,
Es wurde nun gefunden, dass durch Verknüpfung von 2 Molekülen p-Amino-sa. licyl- saure durch eine-CO-Brüeke unter Bildung von N,N'-(4,4'-Dicarobys-3,3'-dioxy)-diphenylharnstoff mit der Struktur

eine Substanz erhalten wird, die langsamer ausgesehieden wird und eine p-Amino-salicylsäure-Depotwirkung entfaltet.
Als readktionsfähinges Kohelsäurederivart verwendet man Vorzugsweise Phosgen. Die Reaktion kann ohne oder bei Gegenwart eines organischen Losumgsmittets durchgeführt werden. Der erwähnte Harnstoff wird auch bei der Einwirkung von Phosgen auf eine wässrige Lösug von 4-Amino-salicylsärue oder deren Salze, wie das Natrium-oder Calcium- salz gebildet. Die Phosgenierung wird zweckmässig bei Gegenwart eines Überschusses eines säurebindenfen. Mittels, wie Natriumhydroxyd, Soda, Pottasche, Calciumdrbonat, einer tertiären Base oder dergleichen, durchgeführt.
Beispiel 1
31 g p-Amino-salicylsäure werden in einem Rührkolben in 200 ml Aceton gelöst, und die Losung wird'mit 80 g Pottasche versetzt. Nun werden unter Rühren langsam 150 ml einer 20%igen Lösung von Phosgen in Toluol zu getropft. Die Temperattur steigt langsam a. uf 40-50 C an, und es entweicht Kohlensäure.
Nach dem Zutropfen der Tolullösung wird noeh 1 Stunde weitergerührt, und dann werden die Lösungsmittel im Vakuum abgedampft. Der Rüchstand wird in 400 ml Wasser gelöst, mit Tierkohle filtriert, das Filtrat du ! rch Zusatz von Ameisenssäure angesäuert und einige Zeit erwärmt. Dadurch wird die anfangs sehleimige Fällumg kristalilinisch. Nun wi, rd abgesaugt und mit Wasser gewasehen.
Man erhält, N,N'-(4,4'-Dicarboxy-3,3'-dioxy)diphenyl-harnstoff als fast farbolses Pulver vom F. 265-270 C (Zers). Die Substanz ist leicht löslich in Soda und Ammoniak, schwer lösich in Aceton und : Alkohol. Durch Neutra- lisation in-wässriger Losung mit Natronlauge und Eindampfen zur Trochkne erhält man das Dinatriumsal als in Wasser leicht lösliches Pulver. Durch Versetzen einer wässrigen Losung des Natriumsal'zes mit einer Lösung eines Caiciumsalzes wird das Caleiumsalz der Dicarbonsäure erhalte. Dieses ist ein weisses es Pulver, schwerlöslich in kaltem, leicht loslich in heissem Wasser.
Beispiel 2
35 g p-amino-salicylsrues Natrim werden in 300 i Wasser gelost, und die Lösung wird mit 50 g Soda oder Pottasche versetzt.
Nun wird unter Rühren Phosgen eingeleitet, bis die Diazo-Reaktion auf freie aromatische Aminogruppen negativ geworden ist. Sollte das ps der Lösung vor dem negaitbverden der Reaktion sauer werden, so wird nochmals etwas Soda zugefügt und weiter Phosgen eingeltiet. Nun wird die Lösung angesäuert und der abgeschiedene N, N'- (4, 4'-Dicarobxy-3,3'dioxy)-diphenyl-harnstoff, gegebenenfalls nach Erwärmen der Lösung, abgesaugt und mit Wasser gewaschen.
Beispiel 3
50 g pa-mino0-salicylsaures Caleimu werden in 400 ml Wasser gelöst. Die Lösung wird in einem Rührkolben mit 100 g Caleiumcarbonat versetzt, worauf Phosgen eingeleitet wird, bis die Diazoreaktion auf freie Aminogruppen negativ geworden ist. Das abgeschiedene Caleiumsalz des N, N'- (4, 4'-Diearboxy-3, 3'-dioxy)- diphenyl-harnstoffes wirdl dureh Erwärmen in Losung gebracht, die Lösung heiss filtriert und abgekühlt, wobei das Calciumsalz der Säure auskristallisiert.
Der N, N'-(4, 4'-Diearboxy-3, 3'-dioxy)-di- phenyl-harnstoff und seine Salze stellen neue, bisher unbekannte Verbindungen dar. Sie haben dite ilberrasehendb Eigenschaft, dass man mit ihrer Hilfe stabile Lösungen herzustellen vermag.
