<B>Verfahren zur</B> Herstellung <B>von</B> Chloraminen <B>der Steroidreihe.</B> Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Chlor aminen der Steroidreihe durch Behandeln von Aminen der Steroidreihe mit einem Chlorie- rungsmittel, welches dadurch gekennzeichnet ist, dass man als Chlorierungsmittel \-Chlor- succinimid in Gegenwart. organischer Lösungs mittel verwendet.
In den deutschen Patentsehriften N r. 693351 ( = A. P. 2286892, E. P. 502666 und 465960, F. P. 842026, Schw. P. 211731, 216339 bis 216342) und Nr. 713193 (= A. P. 2108646, E. P. 356960 und 470936, F. P. 797960, Schw. P. 222386) sowie in Medizin und Chemie , Bd.IV, S. 339 ist bereits ein Verfahren zur Herstellung von Chloraminen der Steroidreihe beschrieben.
Man geht dabei so vor, dass man unterchlorige Säure, hergestellt nach 'Vohl- Goldsehmidt (Berichte der Deutschen Chemi schen Gesellschaft, Bd. 46, S. 2731 [1913] ), auf in Äther gelöste Steroidamine einwirken lässt.
Die Reaktion verläuft z. B. nach folgendem Schema

Das bei der Reaktion entstehende Wasser wird durch wasserfreies Natriumsulfat ge bunden.
Diese Arbeitsweise liefert zwar im kleinen Massstabe befriedigende Ausbeuten, jedoch ist beim Übergang zu grösseren Ansätzen die Her stellung der wasserfreien unterchlorigen Säure in Äther umständlich und zeitraubend. Hinzu kommt, dass die ätherisehen Lösungen der un- terchlorigen Säure sich zersetzen, so dass die Lösung, wenn sie einige Stunden gestanden hat, erst titriert werden muss, um ihren Ge halt an unterchloriger Säure zu bestimmen.
Es wurde nun gefunden, dass die technische Herstellung der Chloramine der Steroidreihe wesentlich vereinfacht werden kann, wenn man N-Chlorsuccinimid an Stelle von wasserfreier unterchloriger Säure verwendet. Dieser Be fund ist ausserordentlich überraschend, da es nach Ziegler (Liebigs Annalen der Chemie, Bd.551, S.80) bekannt ist, dass das Chlor- succinimid gegenüber dem Bromsuccinimid ausserordentlich reaktionsträge ist.
N-Chlorsuecinimid reagiert beispielsweise bereits bei Zimmertemperatur mit 3-Acetoxy - ternorcholenyl- (5 )-amin (20) momentan nach folgendem Schema:

Die Reaktion wird zweckmässig im indiffe renten organischen Lösungsmittel durchge führt. Als indifferente organische Lösungs mittel kommen beispielsweise in Betracht: Benzol, Toluol, Methylenchlorid, Chloroform, Dioxan, Tetrahydrofuran. Besonders eignet sieh Äther als indifferentes organisches Lö sungsmittel.
Bei schwer löslichen Aminen kann man auch in Suspension arbeiten, da die Um- setzungsprodukte wie auch das Succinimicl löslich sind. Die Reaktion wird zweckmässigerweise bei Zimmertemperatur durchgeführt, wobei die Komponenten meist augenblicklich reagieren. Sie lässt sieh jedoch auch bei tieferen Tem peraturen, auch unterhalb 0 , durchführen, jedoch ist die Reaktionsdauer dann entspre chend länger.
Als Ausgangsmaterial kommen in Frage: sowohl Steroidamine, die die Aminogruppe als Substituenten im Steroidskelett enthalten, insbesondere mit der Aminogruppe in 17-Stel- lung, als auch solche Steroidamine, die in 17- Stellung ein durch eine Aminogruppe sub- stituierte Seitenkette tragen, z.
B.: 3-Acet.oxy- ternorcholenyl-(5)-amin-(20), 3-Oxy-andro- sten-(5)-amin-(17), 3-Acetoxy-bisnoreholanyl- amin- (22), 3-Oxo-17-amino-androsten-(4), Cholesterylamin, Cholesterylmethylamin, 3- Amino-androsten- (5) -o1- (17) u. a.
Auch Amin- salze können als Ausgangsmaterial verwendet werden, jedoch ist es zweckmässig, aus den Sal zen mit der entsprechenden Menge alkoholi scher Lauge das Amin in Freiheit zu setzen.
Gegenüber dem bekannten Verfahren mit unterchloriger Säure zeichnet sich das Verfah ren gemäss der vorliegenden Erfindung durch grosse Einfachheit aus. Das N-Chlorsuceinimid ist eine stabile Verbindung, die beliebig lange haltbar ist. Bei der Reaktion entsteht Sueein- imid, das zurückgewonnen werden kann. Au sserdem kommt die Anwendung des wasser freien Natriumsulfats in Fortfall, da. bei der Reaktion der Amine der Steroidreihe mit N- Chlorsuecinimid kein Wasser entsteht.
Die nach dem Verfahren der vorliegenden Erfindung gewonnenen Chloramine der Ste- roidreihe dienen als Zwisehenprodukte.
Sie können z. B. durch Behandeln mit Metallalkoholaten und anschliessender Hydro lyse der erhaltenen Produkte in Oxoverbin- dangen übergeführt werden. So erhält man z. B. Pregnen-(5)-ol-(3)-on-(20) aus 3-Acet- oxy-ternoreholenyl-(5)-amin- (20), Dehydro- androsteron aus 3 - Oxy- androsten - (5) - amin- (17), Oxybisnorcholanaldehyd aus 3-Acetoxy- bisnorcholanylamin-(22),
Androstendion aus 3-Oxo-17-amino-androsten-(4) .
<I>Beispiel 1:</I> In eine getrocknete ätherische Lösung (41), die 60 g 3-Acetoxy-ternorcholenyl-(5)-amin- (20) enthält, trägt man bei Zimmertemperatur 22,6 g fein gepulvertes und gesiebtes N-Chlor- suecinimid (99o/oig) unter Rühren ein. Das N-Chlorsuccinimid geht in Lösung, die Lösung selbst. reagiert danach neutral. Man filtriert von einigen Flocken ab und dampft die Lö sung auf dem Wasserbad auf 1,2-1,4l ein, wobei Succinimid auskristallisiert. Man saugt ab und engt, die Mutterlauge restlos im Vakuum ein.
Das 3-Acetoxy-ternorcholenyl- (5)-chloramin-(20) bleibt als weisse Kristall- masse zurück. Es kann durch Umsetzung mit lIetallalkoholaten und anschliessender Hydro lyse in Pregnen-(5)-ol-(3)-on-(20) (Medizin und Chemie, Band 4, Seite 339) übergeführt. werden.
<I>Beispiel</I> N: Zu einer Lösung von 480 g 3-Acetoxy-ter- noreholenyl-(5)-amin-(20) in 281 absolutem Äther werden unter Rühren bei Zimmertem peratur 185,8 g N-Chlorsuccinimid (96o/oig) zu gefügt. Die Lösung reagiert nach kurzer Zeit. neutral. Es wird noch 30 Minuten nachge rührt, die Reaktionslösung von einigen Flok- ken abfiltriert -und dann auf dem Wasserbad bis zum Eintritt einer Kristallisation einge engt.
Man lässt abkühlen, saugt das Succinimid ab und engt die klare Lösung im Vakuum bis zur Troekne ein, wobei das 3-Acetoxy-ternor- cholenyl-(5)-chloramin-(20) als weisse Kri- stallmasse in quantitativer Ausbeute zurück bleibt. Die Weiterverarbeitung kann, wie im Beispiel 1 angegeben ist, erfolgen.
<I>Beispiel 3:</I> 2 g 3-Oxy-androsten-(5)-amin-(17) werden in 150,em3 trockenem Äther suspendiert, 2 g fein gepulvertes N-Chlorsuccinimid (87 o/oig) zugefügt und die Mischung 11/2 Stunde bei Zimmertemperatur gerührt. Das gebildete Chloramin geht hierbei in Lösung, während das entstandene Succinimid grossenteils un gelöst bleibt. Es wird abfiltriert, die Äther lösung zur Hälfte abdestilliert und abgekühlt.
Es kristallisiert weiteres Succinimid aus, das ebenfalls abfiltriert wird. Die Ätherlösung wird bis zur Trockne eingeengt. Es hinter bleibt das3-Oxy-androsten (5)-ehlaramin-(17) als weisse Kristallmasse in quantitativer Aus beute. Es kann aus wässrigem Aceton oder aus Äther umkristallisiert werden und schmilzt dann bei 290 unter Zersetzung. Es ist stabil und kann über lange Zeit aufbewahrt werden.
Die Weiterverarbeitung zum Dehydroandro- steron kann Behandlung mit Metallalkohola- ten und anschliessende Hydrolyse nach Medi zin und Chemie, Band 4, Seite 341 oder nach der Patentschrift Nr. 693351 erfolgen.
Beispiel <I>4:</I> Zu einer Löstuig von 36,1 g 3-Aeetoxy- ternorcholanyl-amin-(20) in 3 1 Äther abs. werden unter Rühren bei Zimmertemperatur 13,4g N-Chlorsuccinimid (99o/oig) zugesetzt. Die Lösung reagiert nach kurzer Zeit neutral.
Es wird noch 1/2 Stunde nachgerührt, die Re- aktionslösung filtriert und dann auf dem Wasserbad bis zur beginnenden Kristallisation eingeengt. Nach dein Abkühlen wird das aus geschiedene Succinimid abgesaugt und das Filtrat im Vakuum bis zur Trockne einge dampft, wobei das 3-Acetoxy-ternorcholan5=1- ehloramin-(20) als weisse Kristallmasse zu rückbleibt.
Es kann nach dein in der deutschen Pa tentschrift Nr. 693351 beschriebenen Verfah ren in Pregn.anolon übergeführt werden.
<I>Beispiel 5:</I> 3,75 g 3-Acet.oxy-bisnorcholanyl-ainin-(22) werden in absolutem Äther gelöst und die Lö sung unter Eiskiihlung und Rühren mit 1,34 N-Chlorsuccinimid (99o/oig) versetzt. Die Mischung wird noch 1/2 Stunde nachgerührt. Die Ätherlösung wird nun eingeengt und ab gekühlt.
Das auskristallisierte Succinimid wird abfiltriert und die Ätherlösung bis zur Trockne eingedampft, wobei die Chloryiver- bindung des 3-Acetoxy-bisnorchola.nyl-amins- (22) als amorphe Substanz in quantitativer Ausbeute erhalten wird.
<I>Beispiel 6:</I> Zu einer Lösung von 2,87 g 3-Oxo-17- aniino-androsten-(4) in 11 absolutem Äther werden bei -5 1,34 g N-Chlorsuccinimid un ter kräftigem Rühren gegeben. Die Lösung re- amiert nach kurzer Zeit neutral und wird ein geengt.. Beim Abkühlen scheidet sieh das ge bildete Succinimid ab. Nach dem Abfiltrieren wird die Ätherlösung zur Trockne einge dampft. -Man erhält. in praktisch quantitativer Ausbeute das 3-Oxo-17-chlor-amino-androsten- (4).
Es kann nach dem in der deutschen Pa- tentsehrift 1\7r.693351 beschriebenen Verfah ren in Androstendion übergeführt werden.
<I>Beispiel</I> 3,85 g Cholesterylamin werden in Äther gelöst, und unter Rühren bei Zimmertempera tur mit 1,34g Chlorsueeinimid versetzt. Man lässt noch einige Zeit. nachrühren, filtriert dann und engt die Lösung ein. Nach dem Ab kühlen saugt man von dem ausgeschiedenen Succ.inimid ab und dampft, das Filtrat zur Trockne ein. plan erhält in quantitativer Aus beute das Cholesterylehloramin.
Durch Erhitzen mit. Nat.riumalkoliolat- Lösung und. Stehenlassen des Reaktionsge misches nach dem Ansäuern mit Schwefel säure in der durch Patentsehrift Nr.693351 angegebenen Weise kann man aus dem Chole- sterylehloramin Cholestenon gewinnen.
Beispiel <I>8:</I> 2,87g 3-Amino-androsten-(5)-ol-(17) wer den in absolutem Äther gelöst. und die Lösung unter Rühren mit. 1,34 - N-Clilor-succininiid versetzt. Die Lösung wird eingeengt, abge kühlt und filtriert. Das Filtrat. wird anschlie ssend zur Trockne eingedampft. Man erhält das 3-Chloramino-androst,en-(5)-ol-(17), das durch Behandeln mit Met.allalkoholaten und ansehliessender Hydrolyse gemäss dem in Me dizin und Chemie, Band 4, Seite 341 ange gebenen Verfahren in Testosteron übergeführt. werden kann.
An Stelle von Äther kann man auch Chloroform oder Methylenchlorid als Lösungs mittel verwenden. Beispiel <I>9:</I> 4,0 g Cholesterylmet.hzlamin werden in ab solutem Äther gelöst. und unter lebhaftem Rühren mit 1,35 g N-Chlorsueeininiid versetzt. Die Ätherlösung wird eingeengt, nach dem Abkühlen das ausgeschiedene Suceinimid ab gesaugt. und das Filtrat zur Trockne einge dampft. Man erhält in quantitativer Ausbeute die Chlorylverbindung des Cholesterylmethyl- amins in farblosen Kristallen.
Durch Weiterverarbeitung nach dem Ver fahren der deutschen Patentschrift Nr. 693351 kann man die erhaltene Verbindung in Cholestenon überführen.
An Stelle von Äther kann man auch Dioxan oder Tetrahydrofuran als Lösun-smit- tel verwenden.

Although this procedure gives satisfactory yields on a small scale, the manufacture of anhydrous hypochlorous acid in ether is cumbersome and time-consuming when moving to larger batches. It has now been found that the industrial production of the chloramines of the steroid series can be considerably simplified if N-chlorosuccinimide is used instead of anhydrous hypochlorous acid. Compared to the known method with hypochlorous acid, the method according to the present invention is characterized by great simplicity. N-chlorosuceinimide is a stable compound that can be kept indefinitely. The reaction produces sueeinimide which can be recovered. The chloramines of the steroid series obtained by the process of the present invention serve as intermediate products.

<I> Example 6: </I> 1.34 g of N-chlorosuccinimide are added to a solution of 2.87 g of 3-oxo-17-aniino-androsten- (4) in 11 absolute ether with vigorous stirring given. The solution reacts neutrally after a short time and is concentrated. On cooling, the succinimide formed separates out. After filtering off the ether solution is evaporated to dryness. -You get. 3-oxo-17-chloro-amino-androstene- (4) in practically quantitative yield.
It can be converted into androstenedione according to the procedure described in the German Patent Register 1 \ 7r.693351.
