Dans la formule (I) ci-dessus, les substituants du radical phényl dé- signés par Ar peuvent être placés dans l'une quelconque des 5 positions disponi- bles du noyau phényl et, lorsqu'il en existe plus d'un, ils peuvent être identi- ques ou différents et être situés dans n'importe laquelle de plusieurs combinai- sons de positions les uns par rapport aux autres.'Les substituants halogénés com- prennent des dérivés ohloro, 'bromo, iodo et fluoro.
Les substituants alkoxy inférieur 'et alkyl inférieur comprennent de préférence de 1 à 6 atomes de carbone et sont par exemple des substituants méthoxy, éthoxy, n-propoxy, isopropoxy, n-butoxy, isobutoxy, n-pentoxy, n-hexoxy, etc... pour le substituant alkoxy inférieur, et des substituants méthyl, éthyl, n-propyl, iso- propyl, n-butyl-isobutyl, n-pentyl, n-hexyl, etc... pour le substituant alkyl inf éri eur .
Les radicaux alkyl inférieur répondant à la formule (I) ci-dessus, désignés par R, comportent de préférence de 1 à 6 atomes de carbone et compren- nent les radicaux méthyl, éthyl, n-propyl, isopropyl, n-butyl, isobutyl, n-pentyl, n-hexyl, etc..., parmi lesquels on préfère les radicaux méthyl et éthyl.
Les atomes d'halogène des radicaux dihalogène acéthl des composés conformes à l'invention, désignés par -COCH (halogène) dans la formule (I) peu- vent être du chlore, du brome, de l'iode ou du fluor, ces radicaux dihalogéno- acétyl comprenant des radicaux dichloraoétyl, dibromoaoétyl, diiodoacétyl, difluo- roacétyl, bromochloracétyl, bromoiodoacétyl, chloroiodoacétyl, et des radicaux analogues.

N-(2,-dialkoxy-éthyl)-(halogéno-phényl)alkylamines, outre qu'elles constituent des produits intermédiaires pour la préparation des composés de la formule I, possèdent une utilité supplémentaire et inhabituelle, que ne possèdent pas tous les produits intermédiaires du groupe, par le fait qu'elles ont une activité anti- helminthique.
Les N-(2,2-dialkoxyéthyl)-(halogéno-phényl) alkylamines de la présente invention sont utiles sous la forme de leur base libre ou sous la forme de leurs sels d'addition avec un acide et, sous ces deux formes, elles entrent dans le cadre de la présente invention. Les acides qu'on peut utiliser pour préparer les sels d'addition avec des acides sont de préférence ceux qui produisent, quand ils- sont combinés avec la base libre, des sels acceptables au point de vue pharmacody- namique, c'est-à-dire des sels dont les anions sont relativement inoffensifs vis- à-vis des organismes animaux lorsque les sels sont pris en doses pharmacodynami- ques, de sorte que les propriétés physiologiques intéressantes inhérentes à la base libre ne sont pas amoindries par des effets secondaires pouvant être attribués aux anions,
en d'autres termes, ces derniers n'affectent sensiblement pas les pro- priétés pharmacodynamiques inhérentes aux cations. Lors de la mise en oeuvre de la présente invention, la demanderesse a constaté qu'il est commode d'utiliser le sel de l'acide chlorhydrique. Toutefois, d'autres sels appropriés acceptables au point de vue pharmacodynamique et entrant dans le cadre de la présente inven tion sont ceux qui proviennent d'acides minéraux comme l'acide bromhydrique, 1' acide iodhydrique, l'acide nitrique', l'acide phosphorique et l'acide sulfurique, et d'acides organiques comme l'acide acétique, l'acide citrique, l'acide tartri- que, l'acide lactique, l'acide éthanesulfonique, l'acide quinique, etc... qui don- nent les bromhydrate, iodhydrate, nitrate, phosphate, sulfate, acétate, citrate, tartrate, lactate, éthanesulfonate et quinate, respectivement.
On prépare les sels d'addition avec un acide soit en dissolvant la base libre dans une solution aqueuse contenant l'acide approprié et en isolant le sel par évaporation de la solution, soit en faisant réagir la base libre et l'acide dans un solvant organique, auquel cas le sel se sépare directement ou bien peut être obtenu par concentration de la solution.
Bien que des sels acceptables au point àe vue pharmacodynamique soient préférés, tous les sels d'addition avec un acide entrent dans le cadre de la pré- sente invention. Tous les sels d'addition avec un acide sont utiles comme sources de base libre du produit, même si on ne désire pas obtenir le sel particulier per se comme produit final, comme, par exemple, dans le cas où le sel est formé uniquement dans un but de purification ou d'identification, ou dans le cas où on l'utilise comme intermédiaire pour préparer un sel acceptable au point de vue pharmacodynamique par des processus faisant appel à des échanges d'ions.

Les N-(g,2-àialkoxyéthyl)-(halogèno-phényl)alkylamines décrites ci- dessus ont des propriétés chimiothérapeutiques intéressantes, en particulier une activité antihelminthique, qu'on peut déterminer sur la souris à l'aide des pro- cédés normalisés.

A. N-(2,2-dzalkoxyéthyl)-(phényl substitué) alkylamines
La préparation de ces intermédiaires est illustrée par la préparation de la N-(2,2-diéthyoxyéthyl)-2,4- dichlorobenzylamine qui est la suivante : onchauffe un mélange de 80 g de 2,4-diçhlorobenzylamine et de 30,5 g de diéthylacé- tal de chloracétaldéhyde sur un bain-marie, tout en agitant, pendant environ 44 heures. On ajoute un volume égal d'éther et on recueille par filtration le produit solide qui se sépare (42 g de chlorhydrate de 2,4-dichlorobenzylamine). On élimine

l'éther du filtrat par distillation sous vide et on distille le produit résiduel, sous pression réduite.
On obtient ainsi 23,5 g de N-(2,2-diéthyoxyéthyl)-2,4- dichlorobenzylamine distillant à 124 - 132 C sous 0,5 mm; n25D = 1,5160.

Par traitement de la N-(2,2-diéthoxyéthyl)-2,4- dichlorobenzylamine par un acide, par exemple HCl, on forme le sel correspondant d'addition avec un acide, par exemple le chlorhydrate.
On mentionne sur le tableau A d'autres N-(2,2-dialkoxyéthyl)-(phényl substitué) alkylamines qu'on a préparées par le processus décrit au sujet de la préparation de la N-(2,2-diéthoxyéthyl)-2,4-dichlorobenzylamine, en utilisant les réactifs appropriés.

On peut isoler les intermédiaires constitués par les N-(2,2-dialkoxyé- thyl)- (phényl substitué) alkylamines sous forme de leur base libre ou sous forme de leurs sels d'addition avec un acide, par exemple sous forme de leur chlorhydra- te. La préparation suivante illustre la formation et l'isolement de l'un de ces composés sous forme de son sel.
On chauffe sur un bain-marie tout en agitant pendant 7 heures environ, un mélange de 7,5 g de 3,4-di-chlorobenzylamine et de 30,5 g de diéthylacétal de chloracétaldéhyde. On ajoute un volume égal d'éther et on recueille par filtration

A 5 g de N-(2,2-diéthoxyéthyl)-3,4-dichlorobenzylamine, sous forme de sa base libre, on ajoute 2,6 cm3 d'acide chlorhydrique éthanolique 6,5 N dans de l'éther. On recueille le produit solide qui se sépare et on le lave à fond avec de l'éther. On obtient ainsi 3,0 g de chlorhydrate de N-(2,2-diéthoxyéthyl) 3,4-dichlorobenzylamine, point de fusion : 135,8 - 137,0 C (corrigé).

Quand les N-(2,2-dialkoxyéthyl)-halogéno-benzyl-amines (formule III) obtenues par le procédé de l'invention sont administrées par voie buccale à des souris infestées du microbe Hymenolepis nana, on constate qu'elles guérissent la moitié des animaux quand elles sont administrées à raison d'environ 500 mg/kg toutes les 12 heures, pendant 4 jours ;

On peut formuler les N-(2,2-dialkoxyéthyl)-halogéno-benzylamines obtenues conformément à la présente invention de la même manière que les produits antihelminthiques classiques tels que les préparations à base de pipérazine ou d'hexylrésorcine.

B. N-(2.2-dialkoxyéthyl)-N-'(phényl substitué)alkyl7-dihalogénoacétamides.
A titre d'exemple de la préparation de ces composés, on va décrire celle de la N-(2,4-dichlorobenzyl)- N-(2e2-diéthoxyéthyl)dichloracétamide. Une solution de 16,2 g de chlorure de dichloracétyle dans 50 cm3 de dichloréthylène est ajoutée goutte à goutte, tandis qu'on agite et qu'on refroidit, à un mélange de 29,2 g de N-(2,2-diéthoxyéthyl)-2,4-diohlorobenzylamine dans 200 cm3 de di- chloréthylène et 200 cm3 d'une solution de soude caustique 1N. On maintient la température au-dessous de 5 C au moyen d'un bain de glace et de sel.
Lorsque l' addition est terminée, on laisse le mélange se réchauffer jusqu'à la température ambiante, tout en l'agitant. On travaille de la manière suivante la moitié de la couche de dichloréthylène; on la lave deux fois à l'eau, deux fois à l'acide chl hydrique 2N et ensuite à l'eau, puis on la sèche sur du sulfate de calcium anhy- dre. Lorsqu'on a éliminé le solvant par distillation sous pression réduite, on laisse reposer les 16,5 g de produit résiduel (rendement 82%) jusqu'à cristallis tion.
On recristallise alors trois fois le produit cristallisé dans l'isopropano
