  Composition à usage buccal antibactérien antiplaque et son procédé d'utilisation 
La présente invention concerne une composition dentifrice   antibactérienne, antiplaque, à   usage buccal. Plus particulièrement, l'invention concerne une composition dentifrice à usage buccal contenant un agent antibactérien non cationique sensiblement insoluble dans   l'eau,   efficace pour inhiber la plaque dentaire. 



   La plaque dentaire est un dépôt mou qui se forme sur les dents, contrairement au tartre qui est une formation minéralisée dure déposée sur les dents. 



  A la différence du tartre, la plaque peut se former partout sur la surface des dents, y compris notamment au niveau de la lisière gingivale. Par suite, outre son aspect inesthétique, la plaque dentaire est impliquée dans l'apparition de gingivite. 



   Il est par conséquent très souhaitable d'inclure des agents antimicrobiens, dont on sait déjà qu'ils réduisent la plaque dentaire, dans des compositions à usage buccal. Des agents antibactériens de type cationique ont souvent été proposés. En outre, dans le brevet des E. U. A. NO 4 022   8801   un composé fournissant des ions zinc servant d'agent antitartre est mélangé avec un agent antibactérien efficace pour retarder le développement des bactéries responsables de la plaque dentaire. Cependant, ils sont généralement inefficaces lorsqu'ils sont utilisés avec des substances anioniques. Des substances antibactériennes non cationiques peuvent, par contre, être compatibles avec les composants anioniques d'une composition à usage buccal.

   Un autre avantage de la présente invention réside en ce que   l'ASA   améliore la distribution et la rétention d'une quantité antiplaque, faible mais efficace, de l'agent antibactérien sur les dents et sur les tissus mous de la cavité buccale. 



   Un autre avantage de la présente invention réside en ce qu'elle fournit une composition antiplaque à usage buccal qui est efficace pour réduire l'apparition de gingivite. 



   D'autres avantages de la présente invention se dégageront de la description suivante. 



   Sous certains de ses aspects, la présente invention concerne une composition à usage buccal comprenant une quantité efficace contre la plaque dentaire d'un agent antibactérien non cationique sensiblement insoluble dans   l'eau, environ 0, 005   à 4 % en poids d'un agent de stimulation antibactérienne qui améliore la   disericution   et la rétention dudit agent antibactérien

   Comme on l'entend   ici, l'expression"groupe   améliorant la distribution"se rapporte à un groupe qui fixe ou lie de manière substantive, adhésive ou cohésive ou   o'une   autre façon l'ASA (transportant l'agent antibactérien) aux surfaces de la cavité buccale (par exemple des dents et les gencives), en"délivrant"ou "distribuant"ainsi l'agent antibactérien à ces surfaces. Le groupe organique améliorant la rétention, généralement hydrophobe, fixe ou lie d'une autre manière l'agent antibactérien à   l'ASA, en   améliorant ainsi la rétention de l'agent antibactérien à   l'ASA   et, indirectement, aux surfaces de la cavité buccale.

   Dans certains cas, la fixation de l'agent antibactérien s'effectue par son empiègement physique par   l'ASA, en   particulier lorsque   l'ASA   est un polymère réticulé, dont la structure offre par nature davantage de sites convenant à   un     tel ampiègement.   La présence d'un fragment réticulant plus hydrophobe, de plus haut poids moléculaire, dans le polymère réticulé favorise encore   l'empiègement   physique de l'agent antibactérien par le polymère réticulé formant l'ASA. 



   De préférence,   l'ASA   est un polymère anionique comprenant une chaîne ou squelette ayant des motifs récurrents qui contiennent chacun, de préférence, au

 moins un atome de carbone et, de préférence, au moins un groupe monovalent améliorant la distribution fixé latéralement directement ou indirectement et au moins un groupe monovalent améliorant la rétention fixé latéralement directement ou indirectement, qui sont liés en relation géminale, vicinale ou, moins préférablement,   d'une   autre manière à des atomes de la chaîne, de préférence des atomes de carbone.

   Moins préférablement, le polymère peut contenir des groupes améliorant la distribution et/ou des groupes améliorant la rétention et/ou d'autres atomes ou groupes divalents comme maillons de la chaîne polymère à la place ou en plus des atomes de carbone, ou comme fragments réticulants. 



   On comprendra que tous les exemples ou illustrations d'ASA présentés ici qui ne contiennent pas à la fois des groupes améliorant la distribution et des groupes améliorant la rétention peuvent, et de préférence doivent, être chimiquement modifiés d'une manière connue pour former les ASA préférés contenant ces deux

 types de groupes, et de préférence plusieurs de chaque type de ces groupes. Dans le cas des ASA polymères pré- férés, il est avantageux, pour maximaliser la fixation substative et la distribution de l'agent antibactérien aux surfaces de la cavité buccale, que les motifs récurrents de la chaîne ou squelette polymère comportant les groupes acides améliorant la distribution constituent au moins 10 %, de préférence au moins environ 50 %, et mieux encore au moins environ 80 % à 95 % ou 100 % en poids du polymère. 



   Selon une forme de réalisation préférée de la présente invention,   l'ASA   consiste en un polymère contenant des motifs récurrents dans lesquels un ou plusieurs groupes acide phosphonique améliorant la distribution sont liés à un ou plusieurs atomes de carbone de la chaîne polymère. Un exemple d'un tel ASA est un

   D'autres polymères contenant des groupes phosphoniques comprennent, par exemple, un polymère d'éthylène phosphoné ayant des motifs de formule :

   Sous un aspect de l'invention, la composition à usage buccal comprend un véhicule acceptable en usage buccal, une quantité efficace contre la plaque dentaire

 d'un agent antibactérien non cationique sensiblement insoluble dans l'eau et un agent de stimulation antibactérienne dont le poids moléculaire est d'environ 1000 à environ 1 000 000 et qui contient au moins un groupe fonctionnel améliorant la distribution et au moins un groupe organique améliorant la rétention, ledit agent contenant lesdits groupes étant dépourvu ou sensiblement dépourvu de tout polycarboxylate polymère linéaire anionique synthétique hydrosoluble de métal alcalin ou d'ammonium ayant un poids moléculaire d'environ 1000 à environ 1 000 000. 



   Selon une autre forme de réalisation préfé-   rée ; l'ASA   peut contenir un polycarboxylate polymère anionique synthétique. Bien qu'il ne soit pas utilisé dans la présente invention pour agir de concert avec un agent antitartre du type polyphosphate, un polycarboxylate polymère anionique synthétique ayant un poids moléculaire d'environ 1000 à environ 1 000 000, de préférence d'environ 30 000 à environ 500 000, a été utilisé comme inhibiteur de phosphatase alcaline pour optimiser l'activité antitartre de polyphosphates salins linéaires à déshydratation moléculaire, comme décrit dans le brevet des E. U. A. NO 4 627 977.

   En effet, dans le brevet britannique publié sous le NO 2 200 551, le polycarboxylate polymère est proposé comme un ingrédient facultatif dans des compositions à usage buccal contenant des polyphosphates salins linéaires à déshydratation moléculaire et un agent antibactérien non cationique sensiblement insoluble dans l'eau.

   On a encore observé, dans le contexte de la présente invention, que ce polycarboxylate, est très' efficace pour améliorer la distribution et la rétention de l'agent antiplaque antibactérien non ionique sur les surfaces

 dentaires lorsqu'il n'y a pas d'autre ingrédient avec lequel le polycarboxylate polymère est susceptible de réagir (c'est-à-dire un polyphosphate à déshydratation moléculaire) ; par exemple, lorsque l'ingrédient avec lequel le polycarboxylate polymère agit de concert est notamment l'agent antibactérien non cationique. 



   Les polycarboxylates polymères anioniques synthétiques et leurs complexes formés avec divers germicides cationiques, du zinc et du magnésium ont été antérieurement proposés comme agents antitartre par eux-mêmes, par exemple dans les brevets des E. U. A. NO 3 429 963,4 152 420,3 956 480,4 138 477 et 4 183 914. On comprendra que les polycarboxylates polymères anioniques synthétiques ainsi proposés dans ces divers brevets, lorsqu'ils contiennent ou sont modifiés pour contenir des groupes améliorant la rétention, sont utilisables dans les compositions et procédés de la présente invention et, dans cette mesure, les exposés pertinents de ces brevets sont incorporés ici par référence à ceux-ci. 



   Les polycarboxylates polymères anioniques synthétiques utilisés ici sont bien connus, étant donné qu'ils sont souvent utilisés sous forme de leurs acides libres ou de leurs sels de métaux alcalins (par exemple potassium et, de préférence sodium) ou d'ammonium hydrolubles de préférence partiellement ou, mieux encore, totalement neutralisés. On préfère les copolymères de 1 : 4 à 4 : 1 d'anhydride ou acide maléique avec un autre monomère polymérisable éthyléniquement insaturé, de préférence un copolymère éther de méthyle et de vinyle/ anhydride maléique ayant un poids moléculaire (P. M.) d'environ 30 000 à environ 1 000 000 et mieux encore d'environ 30 000 à environ 500 000. Ces copolymères sont disponibles, par exemple, sous les désignations de produits Gantrez, par exemple AN 139 (P. M. 500 000),

   D'autres polycarboxylates polymères utilisables comme ASA, contenant ou modifiés pour contenir des groupes améliorant la rétention comprennent ceux proposés dans le brevet des E. U. A.   NO 3   956 480 précité, tels que les copolymères à l : l d'anhydride maléique avec l'acrylate d'éthyle, le méthacrylate d'hydroxyéthyle, la N-vinyl-2-pyrrolidone ou l'éthylène, ce dernier type de copolymère étant disponible, par exemple, sous les désignations EMA   NO 1103,   P. M. 10 000,   et EMA   Grade 61 de Monsanto, et les copolymères à 1 : 1 d'acide acrylique avec le méthacrylate de méthyle ou d'hydroxyéthyle, l'acrylate de méthyle ou d'éthyle, l'éther   d'isobutyle   et de vinyle ou la N-vinyl-2-pyrrolidone. 



   D'autres polycarboxylates polymères utilisables proposés dans les brevets des E. U. A. NO 4 138 477 et 4 183 914 précités, contenant ou modifiés pour contenir des groupes améliorant la rétention, comprennent des copolymères d'anhydride maléique avec le styrène,   l'isobutylène   ou l'éther d'éthyle et de vinyle, des acides polyacryliques, polyitaconiques et polymaléiques, et des oligomères sulfoacryliques de P. M. aussi bas que 1000, disponibles sous la désignation ND-2 de Uniroyal. 



   Des monomères appropriés d'une façon générale sont les acides carboxyliques oléfiniquement ou éthyléniquement insaturés contenant un groupe améliorant la rétention, qui comportent une double liaison oléfinique carbone-carbone activée opérant facilement une polymérisation du fait qu'elle est présente dans la molécule de monomère soit en position alpha-bêta par rapport à un groupe carboxyle, soit comme partie d'un groupement méthylène terminal.

   Des exemples représentatifs de tels

 acides sont les acides acrylique, méthacrylique, éthacrylique, alpha-chloracrylique, crotonique, bêta-acryloxypropionique, sorbique, alpha-chlorosorbique, cinnamique, bêta-styrylacrylique, muconique, itaconique, citraconique, mésaconique, glutaconique, aconitique, alpha-phénylacrylique, 2-benzylacrylique, 2-cyclohexylacrylique, angélique, umbellique, fumarique, maléique et leurs anhydrides. D'autres monomères oléfiniques différents copolymérisables avec ces monomères carboxyliques comprennent l'acétate de vinyle, le chlorure de vinyle, le maléate de diméthyle, etc. Les copolymères contiennent suffisamment de groupes carboxyliques salifiés pour assurer leur solubilité dans l'eau. 



   Des substances également utiles ici sont des polymères dits carboxyvinyliques proposés comme composants de pâtes dentifrices dans les brevets des E. U. A. 



  NO 3 980 767,3 935 306,3 919 409,3 911 904 et 3 711 604. Ces polymères sont disponibles dans le commerce, par exemple sous les marques de fabrique Carbopol 934,940 et 941 de B. F. Goodrich, ces produits étant essentiellement constitués d'un polymère colloidalement hydrosoluble d'acide polyacrylique réticulé par environ 0,75 % à environ 2,0 % de polyallyl-saccharose ou de polyallyl-pentaérythritol comme agent réticulant, la structure réticulée et les liaisons de réticulation assurant l'amélioration de rétention désirée par hydrophobie et/ou empiègement physique de l'agent antibactérien ou autre.

   Un polymère carbophile est assez similaire s'agissant d'un acide polyacrylique réticulé par moins de 0, 2 % de divinylglycol, la proportion, le poids moléculaire et/ou l'hydrophobie plus faibles de cet agent réticulant ayant tendance à n'offrir que peu sinon pas d'amélioration de la rétention. Le 2, 5dimethyl-1, 5-hexadiène est un exemple d'agent réticulant améliorant la rétention qui est plus efficace.

   Le composant polycarboxylate polymère anionique synthétique est principalement un hydrocarbure comportant des substituants et liaisons halogénés et oxygénés facultatifs, comme par exemple des groupes ester, éther et OH, et on l'emploie généralement dans les compositions de l'invention en des proportions pondérales approximatives de 0,05 à 4 %, de préférence de 0,05 à 3 %, et mieux encore de 0, 1 à 2 %. 



   L'ASA peut également comprendre des polycarboxylates polymères anioniques naturels contenant des groupes améliorant la rétention. La carboxyméthylcellulose et autres agents liants, gommes et agents filmogènes dépourvus des groupes améliorant la distribution et/ou améliorant la rétention ci-dessus définis sont inefficaces en tant qu'ASA. 



   A titre d'exemples représentatifs de ASA contenant des groupes acide phosphinique et/ou acide sulfonique améliorant la distribution, on peut mentionner des polymères et copolymères contenant des motifs ou fragments provenant de la polymérisation d'acides vinyl-ou allyl-phosphiniques et/ou sulfoniques substitués, selon les besoins, sur l'atome de carbone en 1 ou 2 (ou 3) par un groupe organique améliorant la rétention, répondant par exemple à la    formule- (X) n-R   définie ci-dessus. On peut employer des mélanges de ces monomères et leurs copolymères formés avec un ou plusieurs monomères polymérisables inertes éthyléniquement insaturés tels que ceux décrits ci-dessus à propos des polycarboxylates polymères anioniques synthétiques utilisables.

   Comme on le remarquera, dans ces ASA polymères ainsi que d'autres utilisables ici, il n'y a en général qu'un seul groupe acide améliorant la distribution lié à un quelconque atome donné de carbone ou autre du squelette du polymère ou d'une ramification de celui-ci. On peut également utiliser ici comme ASA des

 polysiloxanes contenant ou modifiés pour contenir des groupes latéraux améliorant la distribution et des groupes latéraux améliorant la rétention. Des ionomères contenant ou modifiés pour contenir des groupes améliorant la distribution et la rétention sont également efficaces ici comme ASA.

   Ces ionomères sont décrits aux pages 546 à 573 du volume supplémentaire   de"Kirk   Othmer Encyclopedia of Chemical Technology", troisième édition, John Wiley & Sons, Inc., copyright 1984, dont la description est incorporée ici par référence. Des substances également efficaces ici comme ASA sont des polyesters, polyuréthannes et polyamides synthétiques et naturels, y compris des protéines et des matières protéiques telles que le collagène, une poly (arginine) et d'autres amino-acides polymérisés, pourvu qu'ils contiennent ou soient modifiés pour contenir des groupes améliorant la rétention. 



   Sous un aspect de la présente invention,   l'ASA   qui a un poids moléculaire moyen d'environ 1000 à environ 1 000 000 contient au moins un groupe fonctionnel améliorant la distribution et au moins un groupe

 organique améliorant la rétention, ledit agent conte- - nant lesdits groupes étant dépourvu ou sensiblement dépourvu de polycarboxylate polymère linéaire anionique synthétique hydrosoluble de métal alcalin ou d'ammonium ayant un poids moléculaire d'environ 1000 à environ 1 000 000. 



   Dans la présente invention, une composition à usage buccal préférée est un dentifrice contenant environ 0,3 % en poids de l'agent antibactérien (par exemple le Triclosan) et environ 1,5 à 2 % en poids du polycarboxylate servant d'ASA. 



   Sans vouloir être tenu à une théorie, on considère que l'ASA, en particulier un ASA polymère, est généralement une substance anionique filmogène et

 l'on pense qu'il se fixe aux surfaces dentaires et forme une pellicule continue sur les surfaces, en empêchant ainsi une fixation bactérienne à la surface des dents. Il est possible que l'agent antibactérien non cationique forme un complexe ou une autre sorte d'association avec   l'ASA, en   formant ainsi une pellicule d'un complexe ou autre sur les surfaces dentaires.

   Il apparaît que la propriété filmogène de   l'ASA   et les meilleures distribution et rétention de l'agent antibactérien sur les surfaces dentaires sous l'effet de l'ASA rendent les surfaces dentaires non réceptives à une accumulation bactérienne en particulier du fait que l'action bactériostatique directe de l'agent antibactérien inhibe la croissance bactérienne. Par conséquent, par la combinaison des trois modes d'actions, à savoir   l)   la meilleure distribution, 2) le long temps de rétention sur les surfaces dentaires et 3) l'empêchement de la fixation bactérienne aux surfaces dentaires, la composition à usage buccal est rendue efficace pour réduire la plaque dentaire. On obtient une efficacité antiplaque analogue sur les tissus mous de la bouche au niveau ou à proximité de la lisière gingivale. 



   Selon la présente invention, le véhicule acceptable en usage buccal est efficace pour permettre à l'agent antibactérien non cationique sensiblement insoluble dans l'eau de se dissoudre dans la salive en une quantité efficace contre la plaque dentaire. 



   Dans la préparation à usage buccal, un véhicule acceptable en usage buccal comprend une phase aqueuse en présence d'un agent humectant. Dans un dentifrice en gel, contenant typiquement environ 5 à 30 % en poids d'un agent de polissage siliceux, l'eau est habituellement présente en une quantité d'au moins environ 3 % en poids, généralement d'environ 3 à 35 %, et l'humectant, de préférence le glycérol et/ou le

 sorbitol, constitue généralement au total environ 6,5 à 75 % ou 80 % en poids de la composition de gel dentifrice à usage buccal. Le sorbitol dont il est question ici désigne le produit généralement disponible dans le commerce en solutions aqueuses à 70 %. 



   Lorsque la quantité d'agent antibactérien est d'environ   0, 25   à 0,35 % en poids, les dentifrices en gel ne nécessitent pas d'autre ingrédient dans le véhicule à usage buccal pour solubiliser l'agent antibactérien, bien que la présence d'un tel agent solubilisant soit facultative. Lorsque la quantité d'agent antibactérien est inférieure à environ 0,25 % en poids, par exemple d'environ 0, 01 à environ   0, 25   % en poids, l'agent solubilisant doit donc être présent pour assurer une solubilisation suffisante dans la salive pour produire l'activité antiplaque.

   Lorsque la quantité d'agent antibactérien est supérieure à environ 0,35 % en poids, par exemple d'environ 0,35 à environ 0,5 % ou plus, par exemple de 5 %, l'agent solubilisant doit par conséquent être présent car, sinon, une partie importante de l'agent antibactérien resterait insoluble. 



   Lorsque la composition à usage buccal est un dentifrice contenant environ 30 à 75 % en poids d'un agent de polissage acceptable pour les dents, la présence d'un tel agent solubilisant est également facultative. 



   Lorsque la composition à usage buccal est un bain de bouche ou un dentifrice liquide, le véhicule à usage buccal contient l'un au moins d'un agent tensioactif, d'une essence aromatisante ou d'un alcool non toxique, chacun aidant à dissoudre l'agent antibactérien et, ici encore, la présence de l'agent solubilisant est facultative. 



   Lorsqu'un agent solubilisant est présent dans les compositions à usage buccal de la présente inven-

 tion, sa quantité est généralement d'environ 0,5 à 20 % en poids, une quantité aussi faible qu'environ 0,5 % en poids étant suffisante lorsque l'agent antibactérien non cationique sensiblement insoluble dans l'eau est présent en faible quantité, à savoir d'environ 0,3 % en poids. Lorsque l'agent antibactérien est présent en des quantités supérieures comme d'au moins environ 0,5 % en poids et, en particulier, lorsqu'un agent de polissage siliceux est également présent en une quantité d'environ 5 à 30 % en poids, il est souhaitable qu'il y ait au moins environ 5 % en poids, et jusqu'à 20 % en poids ou plus, de l'agent solubilisant.

   Il est à remarquer que le dentifrice peut avoir tendance à se séparer en des parties liquide et solide lorsqu'il y a plus d'environ 5 % en poids de l'agent solubilisant. 



   L'agent qui est facultativement présent pour solubiliser l'agent antibactérien dans la salive peut être incorporé au véhicule eau-humectant. Ces agents solubilisants comprennent des humectants du type polyol tels que le propylène-glycol, le dipropylène-glycol et

 l'hexylène-glycol, des produits du type Cellosolve tels que le Méthylcellosolve et l'Ethylcellosolve, des huiles végétales et des cires contenant au moins environ 12 atomes de carbone dans une configuration de chaîne droite, telles que l'huile d'olive, l'huile de ricin et la vaseline, et des esters tels que l'acétate d'amyle, l'acétate d'éthyle et le benzoate de benzyle. Comme on l'emploie ici, le terme"propylène-glycol"inclut le 1, 2-propylène-glycol et le 1,3-propylène-glycol.

   Des quantités importantes de polyéthylène-glycol, en particulier d'un poids moléculaire égal ou supérieur à 600, sont à éviter car le polyéthylène-glycol inhibe fortement l'activité antibactérienne de l'agent antibactérien non cationique. Par exemple, le polyéthylèneglycol (PEG) 600, lorsqu'il est présent avec du

 Triclosan en un rapport en poids Triclosan : PEG 600 de 25 : 1, réduit l'activité antibactérienne du Triclosan d'un facteur d'environ 16 par rapport à celle qui s'excerce en l'absence du polyéthylène-glycol. 



   Sous certains aspects de la présente invention, la composition dentifrice à usage buccal peut avoir sensiblement la nature d'un gel, s'agissant par exemple d'un gel dentifrice. Ces préparations en gel à usage buccal contiennent une matière siliceuse de polissage dentaire. Des matières de polissage préférées comprennent une silice cristalline ayant une dimension granulométrique limitée à un maximum d'environ 5 micromètres, une dimension granulométrique moyenne limitée à un maximum d'environ   1, 1   micromètre et une surface spécifique limitée à un maximum d'environ 50 000 cm2/g, un gel de silice ou une silice colloidale, et un aluminosilicate amorphe complexe de métal alcalin. 



   Lorsqu'on utilise des gels visuellement transparents ou opacifiés, une silice colloïdale telle que celles vendues sous la marque commerciale SYLOID, comme Syloid 72 et Syloid   74, ou   sous la marque commerciale SANTOCEL, comme Santocel 100, ou des complexes aluminosilicatés de métaux alcalins (c'est-à-dire de la silice contenant de l'alumine combinée dans sa matrice) sont particulièrement utiles comme agents de polissage car ils sont compatibles avec la texture d'un gel et leur indice de réfraction est proche de l'indice de réfraction des systèmes agent gélifiant-liquide (comprenant de l'eau et/ou un humectant) couramment utilisés dans les dentifrices. 



   La matière de polissage est généralement présente dans les compositions de dentifrice à usage buccal, telles que des compositions de pâte ou gel dentifrice, en des concentrations pondérales d'environ 5 % à environ 30 %.

   Sous l'aspect de la présente invention dans lequel la préparation à usage buccal est un dentifrice, cette préparation contient un véhicule acceptable en usage buccal comprenant une phase aqueuse avec un humectant qui est de préférence le glycérol et/ou le sorbitol, dans lequel l'eau est généralement présente en une quantité d'environ 15 à 35 % ou 40 % en poids et le glycérol et/ou le sorbitol constituent généralement au total environ 20 à 75 % en poids, et plus typiquement 25 à 60 %, de la préparation dentifrice à usage buccal. La mention du sorbitol désigne ici encore le produit généralement disponible dans le commerce en solutions aqueuses à 70 %. 



   Dans la présente invention, la composition dentifrice à usage buccal peut être de nature sensiblement pâteuse, s'agissant par exemple d'une pâte dentifrice (crème dentaire), bien que si l'on utilise un agent de polissage siliceux (ce qui n'est généralement pas le cas puisque cette matière n'est habituellement pas utilisée en une quantité supérieure à environ 30 % en poids), elle puisse avoir la nature d'un gel. Le véhicule de la composition dentifrice à usage buccal contient une matière de polissage acceptable pour les dents.

   Des exemples de matières de polissage sont les variétés insolubles dans l'eau de métaphosphate de sodium et de métaphosphate de potassium, le phosphate tricalcique, le phosphate dicalcique dihydraté, le phosphate dicalcique anhydre, le carbonate de calcium, le silicate d'aluminium, l'alumine hydratée, la silice, la bentonite et leurs mélanges entre eux ou avec des matières de polissage dures telles que l'alumine calcinée et le silicate de zirconium, une matière contenant les résines thermodurcissables particulaires décrites dans le brevet des E. U. A. NO 3 070 510, telles que des résines mélamine-phénol et urée-formaldéhyde, et des

 polyépoxydes et polyesters réticulés. Les matières de polissage préférées comprennent les métaphosphates de sodium insolubles, le phosphate dicalcique et l'alumine hydratée. 



   Un grand nombre des matières de polissage dites"insolubles dans l'eau"sont de caractère anionique et renferment également de petites quantités de matière soluble. Ainsi, un métaphosphate de sodium insoluble peut être préparé par toute méthode appropriée comme expliqué, par exemple, dans l'ouvrage "Dictionary of Applied   Chemistry"de   Thorpe, Volume 9, 4ème édition, pages 510-511. Les formes de métaphosphate de sodium insoluble connues en tant que sel de Madrell et sel de Kurrol sont d'autres exemples de matières qui conviennent. Ces métaphosphates salins ne manifestent qu'une infime solubilité dans l'eau et sont par conséquent couramment appelés des métaphosphates insolubles (MPI).

   On y rencontre, comme impuretés, une proportion mineure de matières phosphatées solubles, ordinairement de quelques pour cent, par exemple 4 % en poids tout au plus. La quantité de matières phosphatées solubles, qui sont supposées inclure un trimétaphosphate de sodium soluble dans le cas d'un métaphosphate insoluble, peut être éventuellement réduite ou éliminée par lavage à l'eau. Le métaphosphate insoluble de métal alcalin est typiquement utilisé sous forme d'une poudre dont la dimension granulométrique est telle qu'un maximum de 1 % de la matière soit plus grosse que 37 micromètres. 



   L'alumine hydratée est un exemple de matière de polissage de nature essentiellement non ionique. En général, sa dimension granulométrique est petite, c'est- à-dire qu'au moins environ 85 % des particules mesurent moins de 20 micromètres, et elle est classée en tant que gibbsite (alpha-alumine trihydratée) et normalement

 représentée chimiquement par Al2O3.3H2O ou Al(OH)3. La dimension granulométrique moyenne de la gibbsite est généralement d'environ 6 à 9 micromètres.

La matière de polissage est généralement présente dans les compositions en gel ou en pâte ou crème en des concentrations pondérales d'environ 30 % à environ 75 %. 



   Les pâtes, crèmes et gels dentifrices contiennent typiquement un agent gélifiant ou épaississant naturel ou synthétique en des proportions d'environ 0, 1 à environ 10 %, de préférence d'environ   0, 5   à environ 5 %. Un épaississant approprié est une argile collaidale synthétique formée par un silicate complexe de métaux alcalins et de magnésium, disponible par exemple sous la désignation Laponite (par exemple   CP/SP,   2002, D), ces produits étant commercialisés par Laporte Industries Limited. L'analyse de la Laponite D indique, approximativement, en poids,   58, 00   % de   Si02'25,     40   % de MgO, 3,05 % de   Na 201   0,98 % de   Li20,   de l'eau et des traces de métaux.

   Sa densité réelle est de 2,53 et sa masse volumique apparente (à 8 % d'humidité) de 1,   0 g/ml.   



   D'autres épaississants ou agents gélifiants appropriés comprennent la mousse d'Irlande, la iotacarraghénine, la gomme adragante, l'amidon, la polyvinylpyrrolidone,   l'hydroxyéthylpropylcellulose,   l'hydroxybutylméthylcellulose, l'hydroxypropylméthylcellulose, l'hydroxyéthylcellulose (disponible par

 exemple sous la désignation Natrosol), la carboxyméthylcellulose sodique et, en particulier lorsqu'un agent de polissage siliceux est présent, une silice colloïdale telle que les silices finement broyées disponibles sous les désignations Syloid 244 et Sylodent 15. 



   Sous l'aspect de la présente invention dans lequel la composition à usage buccal est un bain de bouche ou un dentifrice liquide, de nature sensiblement liquide, le véhicule est typiquement un mélange eaualcool, notamment dans un bain de bouche. Généralement, le rapport en poids de l'eau à l'alcool se situe dans l'intervalle d'environ 1 : 1 à environ 20 : 1, de préférence d'environ 3 : 1 à 10 : 1, et mieux encore d'environ 4 : 1 à environ 6 : 1. La quantité totale de mélange eaualcool dans ce type de préparation est généralement comprise dans l'intervalle d'environ 70 à environ 99,9% en poids. L'alcool est un alcool non toxique tel que l'éthanol ou l'isopropanol. Un humectant tel que le glycérol ou le sorbitol peut être présent en une quantité d'environ 10 à 30 % en poids.

   Les dentifrices liquides contiennent généralement environ 50 à 85 % d'eau, ils peuvent contenir environ 0,5 à 20 % en poids d'alcool non toxique et peuvent également contenir environ 10 à 40 % en poids d'un humectant tel que le glycérol et/ou le sorbitol. La mention faite ici au sorbitol désigne le produit qui est généralement disponible dans le commerce en solutions aqueuses à 70 %. 



  L'éthanol est l'alcool non toxique préféré. On pense que l'alcool facilite la dissolution de l'agent antibactérien non cationique insoluble dans l'eau et l'on pense également qu'une essence aromatisante agit de la même façon. 



   Comme indiqué, l'agent antibactérien non cationique est sensiblement insoluble dans l'eau.

  Cependant, dans la présente invention, on pense qu'avec l'ASA, tel qu'un polycarboxylate, qui est présent dans le bain ce bouche ou le dentifrice liquide, un agent tensio-actif organique, une essence aromatisante ou un alcool non toxique facilitent la dissolution de l'agent antibactérien en l'aidant à atteindre les tissus buccaux mous au niveau ou à proximité des gencives ainsi que les surfaces dentaires. Des agents tensio-actifs organiques et/ou des essences aromatisantes qui peuvent également faciliter la dissolution des agents antibactériens sont des ingrédients facultatifs des compositions dentifrices à usage buccal. 



   Des agents tensio-actifs organiques sont également utilisés dans les compositions de la présente invention pour produire une action prophylactique accrue, aider à réaliser une dispersion complète et uniforme de l'agent antibactérien antiplaque dans toute la cavité buccale, et rendre les présentes compositions plus acceptables au point de vue cosmétique. La substance tensio-active organique est de préférence de caractère anionique, non ionique ou ampholytique, et l'on préfère employer comme agent tensio-actif une substance détersive qui confère à la composition des propriétés détersives et moussantes.

   Des exemples de surfactifs anioniques appropriés sont les sels hydrosolubles de monosulfates de monoglycérides d'acides gras supérieurs, tels que le sel de sodium du monoglycéride   monosulfaté   d'acides gras d'huile de coprah hydrogénée, des alkylsulfates supérieurs tels que le laurylsulfate de sodium, des alkylarylsulfonates tels que le dodécylbenzène-sulfonate de sodium, les alkylsulfoacétates supérieurs, les esters d'acides gras supérieurs de 1,   2-dihydroxypropane-sulfonates,   et les acylamides aliphatiques supérieurs sensiblement saturés formés avec des acides aminocarboxyliques aliphatiques

 inférieurs, tels que ceux dont les radicaux acide gras, alkyle ou acyle comptent 12 à 16 atomes de carbone, etc.

   Des exemples des amides mentionnés en dernier lieu sont la   N-lauroylsarcosine, et   les sels de sodium, de potassium et d'éthanolamine de N-lauroyl-, N-myristoylou   N-palmitoyl-sarcosine   qui doivent être sensiblement exempts de savon ou de dérivés similaires d'acides gras supérieurs. L'utilisation de ces sarcosinates dans les compositions à usage buccal de la présente invention est particulièrement avantageuse car ces substances exercent un effet marqué et prolongé d'inhibition de la formation d'acides par décomposition d'hydrates de carbone dans la cavité buccale, et exercent de plus une certaine réduction de la solubilité de l'émail dentaire dans les solutions acides.

   Des exemples de surfactifs non ioniques hydrosolubles sont des produits de condensation de l'oxyde d'éthylène avec divers composés à hydrogène labile réactifs avec l'oxyde d'éthylène qui possèdent de longues chaînes hydrophobes (par exemple des chaînes aliphatiques d'environ 12 à 20 atomes de carbone), ces produits de condensation ("éthoxamères") contenant des fragments polyoxyéthylène hydrophiles, par exemple les produits de condensation de poly (oxyde d'éthylène) avec des acides gras, des alcools gras, des amides gras, des polyalcools (par exemple le monostéarate de sorbitanne) et le poly (oxyde de propylène) (par exemple les produits de la série   Pluronic).   



   L'agent tensio-actif est généralement présent en une quantité d'environ 0,5 à 5 % en poids, de préférence d'environ l à 2,5 % en poids. 



   Lorsque la composition à usage buccal est un dentifrice liquide, l'épaissant ou agent gélifiant naturel ou synthétique tel que décrit est généralement présent en des proportions d'environ 0, 1 à environ 10%, de préférence d'environ 0,5 à environ 5 %.

   En général, les dentifrices liquides ne contiennent pas d'agent de polissage. Cependant, comme décrit dans le brevet des E. U. A. NO 3 506 757, on peut utiliser environ 0, 3 à 2,0 % en poids d'un polysaccharide ayant un poids moléculaire élevé supérieur à l 000 000, contenant des fragments de mannose, glucose, glucuronate de potassium et acétyle dans le rapport approximatif de 2 : 1 : 1 : 1, à titre d'agent suspendant et épaississant, dans un dentifrice qui peut alors contenir également environ 10 à 20 % d'une matière de polissage telle que l'alumine hydratée, le phosphate dicalcique dihydraté, le pyrophosphate de calcium, un métaphosphate de sodium insoluble, le phosphate dicalcique anhydre, le carbonate de calcium, le carbonate de magnésium, l'oxyde de magnésium, la silice, leurs mélanges, etc. 



   Sans vouloir être tenu à une théorie quelconque concernant la manière dont les avantages de la présente invention sont obtenus, on pense qu'un véhicule aqueux humectant est normalement solubilisé dans les micelles de surfactif dans la phase mobile (c'est- à-dire n'incluant pas l'agent gélifiant et l'agent de polissage s'ils sont présents dans une formule de dentifrice). La solution de phase mobile du dentifrice peut, à l'emploi, être diluée par la salive, ce qui provoque la précipitation du Triclosan. Ainsi, on constate que même en l'absence d'un agent de solubilisation spécial pour le Triclosan, lorsque la quantité de Triclosan est d'environ 0,25 à 0,35 % en poids et qu'un ASA tel que le polycarboxylate est présent, il y a suffisamment de Triclosan pour exercer un excellent effet antiplaque sur les tissus mous au niveau de la lisière gingivale.

   Des observations similaires s'appli-   quent   aux autres agents antibactériens non cationiques insolubles dans l'eau décrits ici.

   La composition dentifrice à usage buccal peut également contenir une source d'ions fluorure ou de composant donneur de fluor comme agent anticarie en une quantité suffisante pour fournir environ 25 x 10-4 % à 5000 x    10-4     %   d'ions fluorure. Ces composés peuvent être légèrement solubles dans l'eau ou peuvent être totalement solubles dans l'eau. Ils sont caractérisés par leur faculté de libérer des ions fluorure dans l'eau et par leur quasi-absence de réaction indésirable 'avec les autres composés contenus dans la composition à usage buccal.

   Parmi ces substances, on compte des fluorures salins minéraux, tels que des sels solubles de métaux alcalins et de métaux alcalino-terreux, comme par exemple le fluorure de sodium, le fluorure de potassium, le fluorure d'ammonium, le fluorure de calcium, un fluorure de cuivre tel que le fluorure cuivreux, le fluorure de zinc, le fluorure de baryum, le fluorosilicate de sodium, le fluorosilicate d'ammonium, le fluorozirconate de   sodium, le fluorozirconate     d'ammonium, le monofluorophosphate   de sodium, les monoet difluorophosphates d'aluminium, et le pyrophosphate de sodium et de calcium. On préfère les fluorures de métaux alcalins et d'étain, tels que les fluorures sodique et stanneux, le monofluorophosphate de sodium (MFP) et leurs mélanges. 



   La quantité de composé fournissant du fluor dépend dans une certaine mesure du type de composé, de sa solubilité et du type de préparation à usage buccal, mais ce doit être une quantité non toxique, généralement d'environ   0, 0005   à environ   3, 0   % dans la préparation. Dans une préparation de dentifrice, par exemple un gel dentaire, on considère comme satisfaisante une quantité d'un tel composé qui libère un maximum d'environ 5000 x    10-4   % en poids d'ions F, par rapport au poids de la préparation.  composé peut être utilisée, mais il est préférable d'en employer suffisamment pour libérer environ 300 à 2000 x io-4 % et mieux encore environ 800 à environ 1500 x - < 1 10 - % d'ions fluorure. 



   Dans le cas des fluorures de métaux alcalins, ce composant est typiquement présent en une quantité limitée à environ 2 % en poids, par rapport au poids de la préparation, et de préférence dans l'intervalle d'environ   0, 05   % à l %. Dans le cas du monofluorophosphate de sodium, ce composé peut être présent en une quantité d'environ   0, 1   à 3 %, plus couramment d'environ 0,76 %. 



   On comprendra que, comme cela est habituel, les préparations à usage buccal doivent être vendues ou autrement distribuées dans des emballages convenablement étiquetés. Ainsi, un gel dentifrice est habituellement conditionné dans un tube compressible, typiquement en aluminium, en plomb revêtu intérieurement ou en matière plastique, ou dans un autre distributeur compressible, à pompe ou sous pression, destiné à délivrer son contenu d'une façon dosée et portant une étiquette le décrivant pour l'essentiel comme étant un gel dentifrice, etc. 



   Diverses autres substances peuvent être incorporées dans les préparations à usage buccal de la présente invention, telles que des agents de blanchiement, des conservateurs, des silicones, des composés   chlorophyllés   et/ou des substances ammoniées telles que l'urée et le phosphate diammonique, et leurs mélanges. Ces adjuvants, s'ils sont présents, sont incorporés dans les préparations en des quantités qui ne nuisent sensiblement pas aux propriétés et caractéristiques désirées. Il faut éviter d'importantes proportions de sels et substances, généralement solubles, contenant du zinc, du magnésium et d'autres métaux, qui

 formeraient des complexes avec les composants actifs des préparations de la présente invention. 



   Toute substance aromatisante ou édulcorante convenable peut également être utilisée. Des exemples de constituants aromatisants appropriés sont des huiles essentielles aromatisantes, par exemple les essences de menthe verte, menthe poivrée, wintergreen, sassafras, girofle, sauge, eucalyptus, marjolaine, cannelle, citron et   orange,'et   le salicylate de méthyle. Des agents édulcorants appropriés comprennent le saccharose, le lactose, le maltose, le xylitol, le cyclamate de sodium, la périllartine, AMP (ester méthylique d'aspartyl-phényl-alanine), la saccharine, etc. Les agents aromatisants et édulcorants peuvent convenablement constituer ensemble environ 0,1 % à 5 % ou plus de la préparation.

   En outre, de même que l'agent tensioactif, l'essence aromatisante est supposée faciliter la dissolution de l'agent antibactérien, en la présence ou même en l'absence d'agent tensio-actif. 



   Dans l'application pratique préférée de la présente invention, une composition à usage buccal selon l'invention est de préférence appliquée à intervalles réguliers sur l'émail dentaire et les tissus mous de la cavité buccale, en particulier au niveau ou à proximité de la lisière gingivale, par exemple tous les jours ou tous les deux ou trois jours, ou de préférence l à 3 fois par jour, à un pH d'environ 4,5 à environ   9 ou 10,   généralement d'environ 5, 5 à environ 8, de préférence d'environ 6 à 8 et, au mieux, d'environ 6,5 à environ 7,5, pendant au moins 2 à 8 semaines ou davantage, voire à vie. Même à des pH inférieurs à 5, l'émail n'est pas décalcifié ni autrement endommagé. 



  Le pH peut être ajusté avec un acide (par exemple l'acide citrique ou l'acide benzoïque) ou une base (par exemple l'hydroxyde de sodium) ou tamponné par exemple

 avec du citrate, benzoate, carbonate ou bicarbonate de    sodium, de l'hydrogénophosphate disodique, du dihydro-     génophosphate   de   sodium, etc.).   



   Les compositions de la présente invention peuvent être incorporées dans des pastilles ou dans des chewing-gums ou d'autres produits, par exemple en les agitant dans une base de gomme chaude ou en les appliquant sur la surface extérieure d'une base de gomme dont des exemples représentatifs que l'on peut mentionner sont le   jelutong, le   latex de caoutchouc, des résines Vinylite, etc., avantageusement associée à des plastifiants ou amollissants classiques, du sucre ou d'autres édulcorants ou des hydrates de carbone tels que le glucose, le sorbitol, etc. 



   Les exemples suivants illustrent davantage la nature de la présente invention sans toutefois la limiter. Toutes les quantités et proportions qui y sont mentionnées sont exprimées en poids sauf spécification contraire.

Le oentifrice A ci-dessus distribue le Triclosan aux dents et aux tissus gingivaux mous essentiellement aussi efficacement que le dentifrice B contenant un agent spécial de solubilisation du Triclosan. En d'autres termes, pour que le dentifrice de la présente invention soit efficace, aucun agent de solubilisation spécial n'est nécessaire. De plus, un dentifrice correspondant d'où le polycarboxylate Gantrez est absent distribue le Triclosan sensiblement moins bien. 



   Dans l'exemple ci-dessus, on peut également obtenir de meilleurs résultats en remplaçant le Triclosan par d'autres agents antibactériens décrits ici tel qu'un phénol, le thymol, l'eugénol et le   2, 2'-méthy-   lène-bis (4-chloro-6-bromophénol) et/ou en remplaçant Gantrez par d'autres ASA tels qu'un copolymère 1 : 1 d'anhydride maléique et d'acrylate d'éthyle, un oligomère sulfoacrylique, l'un des produits de la série Carbopol (par exemple 934) et des polymères d'acides   alpha-ou bêta-styrène-phosphoniques   monomères et des copolymères de ces monomères entre eux ou avec d'autres monomères éthyléniquement insaturés polymérisables tels que l'acide vinylphosphonique.

Les résultats ci-dessus montrent que la solution (D) contenant de l'alcool polyvinylique, et non un ASA ci-dessous, produit une fixation de Triclosan de seulement 36,0, tout à fait similaire à la fixation de 31,0 produite par la solution témoin (A) sans additif. 



  Par contre, la solution (C) contenant un poly (acide alpha-styrène-phosphonique) produit une fixation de 86,0, plus de deux fois celle des solutions (A) et (D), et la solution (B) contenant du poly (acide bêtastyrène-phosphonique) produit une fixation égale à

 environ 5 fois celle des solutions (A) et (D), ce qui tend à démontrer en outre que la substitution vicinale du groupe améliorant la distribution donne des résultats supérieurs. Les résultats ci-dessus mettent également en évidence la rétention étonnamment bonne dans le temps du Triclosan sur les disques de HA obtenue avec la solution (B) contenant l'acide poly (bêta-styrènephosphonique) (poids moléculaire d'environ 3000 à 10 000). 



   EXEMPLE 3
L'effet du polycarboxylate linéaire anionique synthétique sur la fixation, la rétention et la libération de l'agent antibactérien non cationique insoluble dans l'eau sur et à partir des surfaces dentaires est   évalué"in vitro"sur   un disque d'hydroxyapatite enduit de salive et sur des cellules épithéliales exfoliées de la cavité buccale. Les évaluations"in vitro"peuvent être mises en corrélation avec la distribution et la   rétention"in vivo"sur   les surfaces de la cavité buccale. 



   Pour l'essai de distribution de l'agent antibactérien sur un disque d'hydroxyapatite enduit de salive, on lave abondamment à l'eau distillée de l'hydroxyapatite (HA) fournie par Monsanto Co., la recueille par filtration sous vide et la met à sécher durant une nuit à   37OC.   On broie en poudre la HA séchée à l'aide d'un mortier et d'un pilon. On place 150,00 mg de HA dans la cavité d'une matrice à pastille de KBr   (Barnes Analytical, Stanford, CT. ) et l'on comprime   pendant 6 minutes sous une pression de 4540 kg dans une presse de laboratoire de Carver. Les disques de 13 mm résultants sont frittés pendant 4 heures à 800 C dans un four Thermolyne. On recueille de la salive entière produite par stimulation au parafilm dans un bécher en verre refroidi à la glace.

 salive par centrifugation à 15 000 x g pendant 15 minutes à 4OC. On effectue une stérilisation de la salive clarifiée, sous agitation à   4OC, par   irradiation de l'échantillon avec une lampe UV pendant 1, 0 heure. 



   On hydrate chaque disque fritté avec de l'eau stérile dans un tube à essai en polyéthylène. On retire

 ensuite l'eau et la remplace par 2, 00 ml de salive. Une pellicule de salive est formée par incubation du dis- que pendant une nuit à   37 C   dans un bain-marie sous agitation continue par secousses. Après ce traitement, on retire la salive et traite les disques avec 1, 00 ml d'une solution contenant la solution en phase liquide de dentifrice contenant l'agent antibactérien (Triclosan) et les met à incuber à   37 C   dans le bain-marie sous agitation continue par secousses. Au bout de 30 minutes, on transfère le disque dans un nouveau tube et l'on ajoute 5, 00 ml d'eau, puis on secoue doucement le disque avec un appareil Vortex.

   On transfère ensuite le disque dans un nouveau tube et répète deux fois l'opération de lavage. Enfin, on transfère le disque avec précaution dans un nouveau tube pour éviter un transfert simultané de tout liquide avec le disque. Ensuite, on ajoute 1, 00 ml de méthanol au disque et l'on secoue énergiquement avec un appareil Vortex. On laisse l'échantillon à la température ambiante pendant 30 minutes pour extraire le Triclosan adsorbé en le faisant passer dans le méthanol. On retire ensuite le méthanol par aspiration et le clarifie par centrifugation dans un appareil Beckman Microfuge 11 à 10 000 tr/min pendant 5 minutes. Après ce traitement, on transvase le méthanol dans des flacons de CLHP (chromatographie en phase liquide à haute performance) pour le dosage de l'agent antibactérien.

   Des échantillons en triple exemplaire sont utilisés dans toutes les expériences.

   Pour l'essai de rétention de l'agent antibactérien sur un disque de HA enduit de salive, on traite un disque de HA enduit de salive avec des suspensions de dentifrice comme décrit ci-dessus. Après incubation pendant 30 minutes à   37OC, on   retire le disque de HA de la suspension de dentifrice, le lave deux fois à l'eau, puis le remet à incuber avec de la salive entière humaine produite par stimulation au parafilm, ayant été clarifiée par centrifugation. Après incubation à   370C   sous agitation constante par secousses pendant diverses périodes de temps, on retire le disque de HA de la salive et l'on détermine la quantité d'agent antibactérien (Triclosan) retenue sur le disque et libérée dans la salive par une méthode analytique utilisant la CLHP. 



   Pour l'essai de distribution de l'agent antibactérien sur les cellules épithéliales de la cavité buccale, on mesure la distribution afin de déterminer l'effet du copolymère éther de méthyle et de vinyle/anhydride maléique sur la distribution de l'agent antibactérien (Triclosan) aux tissus mous de la cavité buccale à partir d'un produit dentifrice. On recueille des cellules épithéliales de la cavité buccale à l'aide d'un bâtonnet applicateur en bois en frottant doucement la muqueuse buccale. On met les cellules en suspension dans un tampon de Sels de Salive au Repos (SSR)   (NaCl     50nM, Cad-l, lnM et KHPO. 0, 6nM, pH 7,0) à raison de 5-6 x 105 cellules/ml en utilisant un hématiomètre pour   dénombrer les cellules et les maintient dans la glace jusqu'à l'emploi.

   On ajoute 0, 5 ml de suspension cellulaire, préalablement incubée à   37 C   dans un bain-marie, à   0, 5   ml de la solution d'agent antibactérien à l'essai et l'on met à incuber à   37 C.   On dilue au moins au dixième la solution d'agent antibactérien dans le mélange en incubation afin d'abaisser la concentration de surfactif et d'empêcher une destruction des mem-

 branes cellulaires par le surfactif. Au bout de 30 minutes d'incubation, on récolte les cellules par centrifugation dans un appareil Beckman Microfuge 11 à 5000 tr/min pendant 5 minutes. On lave 3 fois au tampon SSR les cellules recueillies sous forme du culot et les traite avec 1, 5 ml de méthanol.

   On mélange énergiquement l'échantillon et l'analyse par la méthode de CLHP pour déterminer sa teneur en agent antibactérien.

   EXEMPLE 4
Dans des essais avec des disques d'hydroxyapatite enduits de salive et des cellules épithéliales buccales exfoliées différents de ceux présentés dans l'Exemple 3 ci-dessus, ledit Dentifrice B contenant 2,00 % de Gantrez S-97 et 0,20 % de   Triclosan, 10,   00 % de propylène-glycol et 2,00 % de laurylsulfate de sodium, et un dentifrice formulé de façon équivalente   (B'), si   ce n'est la présence de 0, 30 % de Triclosan, sont comparés à un Dentifrice (C) du commerce contenant de l'alumine hydratée comme agent de polissage et (a) 0, 2 % de Triclosan, (b) pas de Gantrez, (c) pas de propylène-glycol, (d) 0, 5 % de citrate de zinc, (e) 2,5 % d'agents tensio-actifs, (f) du monofluorophosphate de sodium et de l'alumine hydratée comme agent de polissage, et à une formulation de Dentifrice (C')

Une   étude"à domicile"est   conduite sur un groupe de volontaires pour évaluer les effets de dentifrices particuliers sur une nouvelle croissance de plaque dentaire selon la méthode décrite par Addy, Wilis et Moran, J. Les surnageants des suspensions dentifrice/eau à 1 : 1 des dentifrices ci-dessus sont mis à l'essai pour déterminer la concentration du Triclosan dans le surnageant et la fixation de Triclosan sur des disques de HA enduits de salive.

   Les résultats montrent que l'utilisation de 50 % d'alumine abrasive augmente sensiblement le Triclosan dans des conditions de faible dilution de 1 : 1 (de 1650 à 1905), ce qui se traduit par une augmentation importante de la fixation de Triclosan (de 52 à 74).

The present invention relates to an antibacterial, anti-plaque toothpaste composition for oral use. More particularly, the invention relates to a dentifrice composition for buccal use containing a non-cationic antibacterial agent substantially insoluble in water, effective for inhibiting dental plaque.



   Dental plaque is a soft deposit that forms on the teeth, unlike tartar which is a hard mineral formation deposited on the teeth.



  Unlike tartar, plaque can form anywhere on the surface of the teeth, including in particular at the gingival margin. As a result, in addition to its unsightly appearance, dental plaque is involved in the appearance of gingivitis.



   It is therefore very desirable to include antimicrobial agents, which are already known to reduce plaque, in compositions for oral use. Cationic type antibacterial agents have often been proposed. In addition, in U.S. Patent No. 4,022,8801, a compound providing zinc ions serving as a scale inhibitor is mixed with an effective antibacterial agent to retard the development of bacteria responsible for dental plaque.    Suitable monomers in general are the olefinically or ethylenically unsaturated carboxylic acids containing a retention-improving group, which have an activated carbon-carbon olefin double bond which easily polymerizes because it is present in the monomer molecule either in the alpha-beta position relative to a carboxyl group, or as part of a terminal methylene group.

   The synthetic anionic polymeric polycarboxylate component is mainly a hydrocarbon comprising optional halogenated and oxygenated substituents and bonds, such as for example ester, ether and OH groups, and it is generally used in the compositions of the invention in approximate weight proportions of 0.05 to 4%, preferably 0.05 to 3%, and more preferably 0.1 to 2%.



   The ASA may also include natural anionic polymeric polycarboxylates containing retention enhancing groups. Carboxymethylcellulose and other binding agents, gums and film-forming agents lacking the distribution improving and / or retention enhancing groups defined above are ineffective as ASAs.

   In the present invention, a preferred oral composition is a toothpaste containing about 0.3% by weight of the antibacterial agent (for example Triclosan) and about 1.5 to 2% by weight of the polycarboxylate serving as ASA.



   Without wishing to be bound by theory, it is considered that ASA, in particular a polymeric ASA, is generally a film-forming anionic substance and

   It appears that the film-forming property of ASA and the better distribution and retention of the antibacterial agent on dental surfaces under the effect of ASA make dental surfaces not receptive to bacterial accumulation, in particular because direct bacteriostatic action of the antibacterial agent inhibits bacterial growth. Consequently, by the combination of the three modes of action, namely l) the best distribution, 2) the long retention time on the dental surfaces and 3) the prevention of bacterial attachment to the dental surfaces, the composition for use buccal is made effective in reducing dental plaque. A similar antiplaque effect is obtained on the soft tissues of the mouth at or near the gingival margin.

The effect of synthetic anionic linear polycarboxylate on the fixation, retention and release of the water-insoluble non-cationic antibacterial agent on and from dental surfaces is evaluated "in vitro" on a hydroxyapatite disk coated with saliva and on epithelial cells exfoliated from the oral cavity. The supernatants of the 1: 1 toothpaste / water suspensions of the above toothpastes are tested to determine the concentration of Triclosan in the supernatant and the binding of Triclosan to HA disks coated with saliva.

   The results show that the use of 50% abrasive alumina appreciably increases Triclosan under low dilution conditions by 1: 1 (from 1650 to 1905), which results in a significant increase in the fixation of Triclosan (from 52 to 74).

Revendications 1.
