Abstract

<P>PROBLEM TO BE SOLVED: To provide a method for identifying and producing a nucleic acid ligand improved on the basis of SELEX method, and to provide a nucleic acid ligand to HIV-RT, HIV-1 Rev, HIV-1 tat, thrombin and basic fibroblast growth factor protein. <P>SOLUTION: Disclosed is a method for producing an improved nucleic acid ligand which is a ligand for a target from a candidate mixture of nucleic acid as an embodiment, wherein the method comprises: (a) bringing the candidate mixture into contact with the target, wherein nucleic acids which have increased affinity to the target compared with the candidate mixture may be fractionated from the residue of the candidate mixture; (b) fractionating affinity-increasing nucleic acids from the residue of the candidate mixture; (c) multiplying the affinity-increasing nucleic acid to obtain the condensed mixture of the ligand of the nucleic acid; (d) if necessary identifying the nucleic acid ligand by repeating the processes (a) to (c); (e) measuring the nucleic acid residue of the nucleic acid which is essential to bind to the target; and (f) producing the improved nucleic acid ligand based on the measurement. <P>COPYRIGHT: (C)2010,JPO&INPIT
