Eine erhöhte Anzahl von einigen Chromosomen ist der Grund für bestimmte Erbkrankheiten oder andere Störungen, dagegen ist für die Ausbildung anderer Erbkrankheiten nur die Verdoppelung eines rezessiven Gens notwendig. Diese Methode soll sich sowohl zur Krebs- wie auch zur pränatalen Diagnostik eignen. Sie soll sich ferner zum Nachweis von Stoffen eignen, die eine Gen-Amplifikation hervorrufen, ferner zum Nachweis der Entwicklung einer Arzneistoff-Resistenz und zur Bestimmung des Expressionswertes von mRNA. Das Verfahren der Erfindung erfordert eine Standard-Nucleinsäure mit konstanter Kopienzahl. Es eignet sich zur Bestimmung der Anzahl von Nucleinsäure-Molekülen pro Einheit, z. Bestimmungen, in denen sowohl die Test- als auch die Standard-Nucleinsäuren entweder als DNA oder RNA vorliegen, können in einer nichtgetrennten Probe durchgeführt werden. Bestimmungen, in denen die Test-Nucleinsäuren als DNA und die Standard-Nucleinsäuren als RNA oder umgekehrt vorliegen, müssen in einer getrennten Probe durchgeführt werden, da für die Weiterbehandlung verschiedene Methoden notwendig sind. Bei der Behandlung von Krebs entwickeln Zellen eine Resistenz gegen den eingesetzten Aizneistoff, indem sie jenes Gen amplifizieren, dessen Expressionsprodukt den Arzneistoff inaktiviert. Benutzt man die in Beispiel 2 beschriebenen Test-Proben, so ist es auch möglich, die Menge der mRNA, die von der DHFR-cDNA abgeschrieben wird, zu messen. Das pMTVdhfr-Plasmid ist so konstruiert, daß die Transkription des DHFR-Gens an dem MMTV-Promotor beginnt. Die entstandenen mRNAs besitzen eine Länge von etwa 1,0 kb. Davon stammen etwa 0,25 kb von dem MMTV Promotorbereich und der Rest von der DHFR-cDNA ab; vgl. Zur Bestimmung der Standardkurve wurde jene Probe verwendet, die infolge der in vitro Transkription des Dihydrofolat-Reduktase-Gens die entsprechende mRNA besitzt Die für die Transkription verwendete DNA wurde hergestellt, indem das 1,4 kb Hind ΠΙ-Fragment des MMTV Promotors aus dem Plasmid pMTVdhfr zusammen mit dem 0,75 kb Hind ΙΠ-Bgl Q-Fragment (DHFR-cDNA) in den Restriktionsstellen Hind III und Bam HI des Plasmids pSP64 (Promega Biotec) subkloniert wurden. Die Proben-RNA wurde in wäßriger 0,2 % SDS-Lösung aufbewahrt Der "sandwich"-Hybridisierungs-Test wurde in der Weise durchgefiihrt, wie er in den Beispielen 1 b) und 2 b) beschrieben wurde, lediglich die Denaturierung wurde weggelassen. Die Zellen wurden durch SDS lysiert und ihre DNA leicht gesehnt, indem sie durch eine feine Nadel einer Spritze für subkutane Injektion gedrückt wurde. Eine 250 μΐ Probe mit etwa 5 x 10^ Zellen wurde von dem Homogenat genommen. Das Homogenat wurde dann ohne Denaturierung in den "sandwich"-Hybridisierungs-Test eingesetzt Die "sandwich"-Hybridisierung wurde durchgeführt, wie in Beispiel 2 c) und 1 b) beschrieben, mit der Ausnahme, daß lediglich die DHFR-Proben der Hybridisierungs-Lösung zugesetzt wurden. In einer Parallelprobe von 250 μΐ Homogenat wurde die Zellzahl bestimmt indem die in Beispiel 2 c) beschriebene Metallothionein-Probe verwendet wurde. Die Hybride wurden während 15 Minuten bei 37 °C an der Streptavidin-Agarose gesammelt Sodann wurde die Agarose einmal während 5 Minuten mit der gepufferten 1 M NaCl-Lösung bei 37 °C und zweimal während 2 Minuten mit 15 mM NaCl, 1,5 mM Natriumcitrat bei 55 °C gewaschen. Die Menge an gebundenen Hybriden wurde durch Messung der Agarose in einem Gamma-Zähler bestimmt vgl. Syvänen et al., Nucleic Acids Res., 14 (1986), 5037-5048. Proben der in Beispiel 2 beschriebene Zellinie wurden in ähnlicher Weise behandelt, das Probenvolumen, entsprechend etwa 2 x 10^ Zellen, betrug 125 μΐ.
