Verfahren zur Herstellung neuer Alkoxypiperidin-Derivate und ihrer Salze
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Alkoxypiperidin-Derivate der allgemeinen Formel :

Diese Verbindungen besitzen interessante pharmakologische Eigenschaften und sollen auf Grund ihrer psychotropen Effekte in der psychiatrischen Pharmakotherapie Verwendung finden.
Zur Herstellung der neuen Alkoxypiperidin-Derivate verfährt man gemäss der Erfindung so, dass man einen reaktionsfähigen Ester der allgemeinen Formel :

in welcher A, X und Z die obige Bedeutung haben und Y einen reaktionsfähigen Säurerest vorstellt, mit einem Alkoxypyperidin umsetzt, worauf man die erhaltenen Verbindungen gewünschtenfalls in an sich bekannter Weise in ihre Salze überführt. Die Reaktion wird zweckmässig in einem geeigneten Lösungsmittel (z. B. Aceton, Butanon) unter Zusatz von säureabfangenden Mitteln, wie Alkalicarbonaten, durchgeführt.
Bei der Herstellung der neuen Verbindungen mit X = Acyl empfiehlt es sich, die entsprechenden Äthylenketale als Ausgangsmaterialien einzusetzen und am Ende des Syntheseweges mit verdünnter Mineralsäure zu den gewünschten Acyl-Derivaten zu verseifen.

Die neuen Alkoxypiperidin-Derivate können in ihre Salze übergeführt werden, wenn man sie in an sich bekannter Weise mit einer Mineralsäure (z. B. Chlorwasserstoff) oder mit einer organischen Säure (z. B. Oxalsäure) umsetzt.
In den nachfolgenden Beispielen wird das erfindungsgemässe Verfahren näher erläutert.

liumcarbonat zugibt und weitere 4 h kocht. Danach saugt man die anorganischen Salze ab, dampft das Filtrat zur Trockne ein, löst in Benzol und extrahiert mit 2n-Salzsäure. Den Cl-Extrakt wäscht man mit Benzol, macht mit verdünnter Natronlauge alkalisch und nimmt die Base in Benzol auf. Der Benzol-Auszug wird getrocknet und eingedampft. Den Rückstand löst man in Äther und fällt mit ätherischer Salzsäure das Hydrochlorid, welches aus Isopropanol umkristallisiert wird. Man erhält 19, 2 g (570/0 d. Th.) 2-Acetyl-10-[3-(4-methoxy-piperdyl)-prepyl]-phenothiazin-hydrochlorid vom Fp. I30 C.
Das als Ausgangsmaterial verwendete 2-Acetyl-IO- (3-chlorpropyl)-phenothiazin-äthylenketal erhält man aus 2-Acetyl-phenothiazin-äthylenkertal (J. Nakazawa u. M. Watatani, Takamine Kenkyusho Nempo 12 [1960], S. 25 ; vgl. Chemical Abstracts 55 [1961], S. 6483) durch Umsetzung mit 1, 3-Chlor-brom-pro- pan und Natriumamid in flüssigem Ammoniak. Man verdunstet das Ammoniak, extrahiert den Rückstand mit Äther und kristallisiert aus Äther/Ligroin ; Fp. : 870C.
Beispiel 2: I0-[3-(4-Methoxy-piperidyl)-propyl]-phenoxazin.

Eine Mischung von 25, 9 g 10-(3-Chlorpropyl)-phenoxazin, 11,5g 4-Methoxy-piperidin,13 g Kaliumcarbonat, 1 g Natriumjodid und 250 cm3 Butanon wird 8 h unter Rückfluss gekocht. Man saugt ab und engt das Filtrat im Vakuum ein. Der Rückstand wird mit verdünnter Salzsäure aufgenommen und. ausge- äthert. Danach macht man die saure Lösung alkalisch und extrahiert mit Methylenchlorid. Der Eindampfrückstand des Methylenchlorid-Extraktes kann aus Isopropanol umkristallisiert werden, und man erhält 27 g (800/0 d. Th.) 10- [3- (4-Methoxy-piperidyl)-propyl]-phenoxazin vom Schmelzpunkt 46-470C. Das Hydrochlorid schmilzt bei 192 - 1930C.
