Abstract
Description
この性質を利用して、銀ナノプレートは、被験物質の検出試薬の標識(例えば、目的タンパク質の検出に用いられる抗体の標識)として用いられている。
一方、銀ナノプレートは酸化により溶解してその形状が変化する。この酸化による銀ナノプレートの形状変化は、意図していた色の変化を引き起こし得る。
そこで、銀ナノプレートを酸化に対して安定化するため、銀ナノプレートの表面を金で被覆することが行われている(特許文献1及び非特許文献1~2)。
また、被験物質に対する特異的結合物質を担持した着色ラテックス粒子の懸濁液を用いた診断薬が知られている。ラテックス粒子径0.02~2μmのポリスチレン粒子などが知られており、有機色素によって赤色や青色に着色されている(特許文献2及び特許文献3)。

本発明の金被覆銀ナノプレート懸濁液は、0~50μMの水溶性高分子を含み、pHが10以下であることを特徴とする。
本発明では、被験物質の検出技術分野で着色標識として用いられている銀ナノプレートを特に制限なく用いることができるが、以下に好ましい態様を詳述する。
銀ナノプレートとは、銀から製造されたナノプレート(板)をいう。
銀ナノプレートの主面の最大長さとなる粒子径(円形の場合は直径に相当し、三角形の場合は最大辺の長さに相当する)は、通常10~1000nmであり、10~150nmが好ましい。更に、銀ナノプレートのアスペクト比(粒子径/厚み)は、通常1.5以上であり、可視光領域にLSPRの吸収波長が発現して多色設計が可能な1.5~10が好ましい。近赤外光での検出に用いる場合には、LSPRが800~2000nmで発現するようなアスペクト比(例えば、アスペクト比11で900nm付近にLSPRが発現)のプレート状銀ナノ粒子を用いればよい。
色を定量的に表す体系である表色系の一つであるマンセル・カラー・システムのマンセル値(以下、単にマンセル値ともいう)が5Y 8.5/14で、CIE1931xy色度図の座標(以下、単に色度座標ともいう)がx:0.4498、y:0.4811であるイエロー(イエロー系色、400~500nm付近にLSPRを発現するプレート状銀ナノ粒子)、マンセル値が5RP 5/14で、色度座標がx:0.4142、y:0.2428であるマゼンタ(マゼンタ系色、500~600nm付近にLSPRを発現するプレート状銀ナノ粒子)、マンセル値が7.5B 6/10で、色度座標がx:0.1934、y:0.2374であるシアン(シアン系色、600~750nm付近にLSPRを発現するプレート状銀ナノ粒子)など、プレート状銀ナノ粒子のアスペクト比を調整することで任意のLSPRの吸収波長を選択できる。なお、図1は、CIE1931xy色度図におけるイエロー、マゼンタ及びシアンの色度座標を示す。アスペクト比の異なる2種以上のプレート状銀ナノ粒子を混合して色を設計してもよい。例えば、イエローとマゼンタを混合し赤色(マンセル値:5R 4/14、色度座標 x:0.5734、y:0.3057)、マゼンタとシアンを混合し青色(マンセル値:10B 4/14、色度座標 x:0.1310、y:0.1580)、イエローとシアンを混合し緑色(マンセル値:2.5G 6.5/10、色度座標 x:0.3000、y:0.6000)などが設計できる。なお、図2は、CIE1931xy色度図における赤色、青色及び緑色の色度座標を示す。さらに、イエロー系色、マゼンタ系色、シアン系色が発現するアスペクト比の異なる3種以上のプレート状銀ナノ粒子を三原色とした減法混合を適用した多色設計を用いることができる。例えば、イエロー系色、マゼンタ系色、シアン系色のプレート状銀ナノ粒子を任意の割合で混合して黒色を設計した場合、イムノクロマト試験時の被験物質検出ラインは背景(白色)とのコントラスト差が大きくなるため視認性(検出感度)が高くなる。
銀ナノプレートの形状は、プラズモン吸収することができるものであれば特に制限されず、意図する色に応じた形状を採用することができる。具体例としては三角形状、五角形状、六角形状等の多角形状や、角がカーブ状となった円形状等の形状があげられる。
本発明では、単一種類(単一形状)の銀ナノプレートを用いてもよく、形状の異なる複数種類の銀ナノプレートの混合物を用いてもよい。
銀ナノプレートの大きさ(主面の最大長さ)並びに形状は、意図する色又は最大吸収波長に応じて適宜設定することができる。銀ナノプレートの最大吸収波長は、430~2000nmの範囲で調整してもよく、好ましくは430~1500nm、特に好ましくは430~1000nmの範囲で調整してもよい。銀ナノプレートの大きさ及び形状と色との関係は、例えば特表2011-508072号公報に記載されている。例えば、銀ナノプレートを三形状および六角形状(主面の最大長さ:20nm、厚さ:5.1nm)とすると、マゼンタ(極大吸収波長:538nm)を呈することができる。銀ナノプレートの最大吸収波長は、銀ナノプレート形成後の金による被覆、懸濁液のpHの調整、及び/又は、被験物質に対する特異的結合物質の担持後に安定する。
銀ナノプレートは市販品を用いてもよく、公知の製造方法や後述の実施例に記載の方法に従って製造したものを用いてもよい。
金の厚さの下限は、銀ナノプレート表面の金による被覆という目的を達成できるものであれば特に制限されないが、厚みの平均が、好ましくは0.1nm以上又は0.3nmである。なお、本発明における金の厚みの平均とは、高角散乱環状暗視野走査透過顕微鏡法(HAADF-STEM)を用いて測定された、銀ナノプレート表面の金の厚みより算出すれば良い。具体的には、HAADF-STEM像から任意の粒子10個について各粒子の任意の部位10点について、金の厚みを測定した計100点のデータの内、上下10%を除いた80点の平均値を金の厚みの平均とすれば良い。
被覆方法は、銀ナノプレート表面の金による被覆という目的を達成できるものであれば特に制限されない。したがって、公知の被覆方法や後述の実施例に記載の被覆方法を用いることができる。
本発明の金被覆銀ナノプレートは、銀ナノプレートの全ての表面が金で被覆されているものであってもよく、銀ナノプレートの表面の一部が金で被覆されているものであってもよい。銀ナノプレートの表面の全て又は一部が金で被覆されたことは、電子顕微鏡による観察及び物理化学的性質の測定など、通常用いられる種々の方法によって確認することができる。例えば、銀ナノプレートの表面の全て又は一部が金で被覆されると、酸又はナトリウム若しくは塩化物イオンに対する安定性が上昇し、酸化に対して安定なものとなる。そうすると、金被覆処理後に、酸性溶液(例えば、2%過酸化水素水)又は緩衝液(例えば、10mMのリン酸緩衝生理食塩水(二価イオンあり又はなし))中という銀ナノプレートにとって過酷な条件下で銀ナノプレートの懸濁液の分光特性(最大吸収波長)を測定しても、その分光特性が水中で測定したときと比較して僅かしか変化しない場合には、その銀ナノプレートは金で被覆されていると判断できる。
1.金被覆銀ナノプレート懸濁液を遠心分離(25,000rpm、2
6,000g)後、上澄み液を除去し、得られた沈殿物を、除去し
た上澄み液と同量の超純水で再度懸濁する。
2.上記工程1で得られた懸濁液に王水を添加後、5分間煮沸して、金
及び銀を王水中へ溶解させる。
3.上記工程2で得られた溶液を、ICP発光分析装置を用いて測定す
る。
なお、各金属の濃度は、任意濃度の標準サンプルを上述と同様に測定することで作成した検量線より算出する。
得られた各金属の濃度から、金と銀の比率が明らかとなり、銀ナノプレート表面が金で被覆されていること、そして、その金の厚さを確認することができる。例えば、三角形の銀ナノプレート上の金の厚さは、以下のようにして算出することができる。

このように、ICP発光分析装置を用いた分析の結果、金濃度と銀濃度の比率が1:4であることがわかった場合には、金被覆処理を施した銀ナノプレート(形状:正三角形、高さ:30nm、厚さ8nmの粒子の場合)の表面の金の厚さは、0.30nmであると計算できる。これは、金原子が一層から二層被覆していることを意味している。
金被覆銀ナノプレートは市販品を用いてもよく、公知の製造方法や後述の実施例に記載の方法に従って製造したものを用いてもよい。
本発明では、単一種類の金被覆銀ナノプレートを用いてもよく、形状や大きさの異なる複数種類の金被覆銀ナノプレートの混合物を用いてもよい。
分散媒としては、金被覆銀ナノプレートを分散できるものであれば特に制限なく用いることができる。具体例としては、水、水性緩衝液(リン酸緩衝生理食塩水、トリス塩酸緩衝液、HEPES緩衝液など)、エタノールやメタノール等のアルコール類、アセトンやメチルエチルケトン等のケトン類、テトラヒドロフラン等が挙げられる。分散媒は、生化学実験において好適であるため、好ましくは水である。
分散媒は一種類であってもよく、複数種類の混合物であってもよい。
金被覆銀ナノプレートの懸濁液は、静置状態で金被覆銀ナノプレートが分散媒中に分散しているものであってもよく、静置状態では金被覆銀ナノプレートは沈降しているが振盪や超音波分散することにより分散媒中に分散するものであってもよい。
金被覆銀ナノプレートの懸濁液は、金被覆銀ナノプレートの製造方法を実施した結果生じた懸濁液をそのまま用いたものであってもよく(但し、後述の水溶性高分子濃度及びpHの要件を満たしていることを条件とする)、前記の懸濁液から金被覆銀ナノプレートを分離し、次いで分散媒中に分散させたものであってもよい。
懸濁液の製造方法に特に制限はなく、公知の製造方法や後述の実施例に記載の製造方法を用いることができる。
本発明における水溶性高分子とは、分子量が500~1,000,000、好ましくは500~100,000の水溶性物質をいう。本発明における水溶性とは、常温常圧下で高分子が水に0.001質量%以上溶解することをいう。
具体例としては、ポリビニルピロリドン(PVP)、ポリエチレングリコール、ポリアクリルアミド、ポリビニルアルコール、ポリアクリル酸、ポリメタクリル酸、ポリアリルアミン、デキストラン、ポリメタクリルアミド、ポリビニルフェノール、ポリ安息香酸ビニル、ウシ血清アルブミン(BSA)、カゼイン、ビス(p-スルホナトフェニル)フェニルホスフィンやポリスチレンスルホン酸等があげられる。これらの水溶性高分子を含有している、塗料やインクなどに用いられる市販の分散剤を、本発明の水溶性高分子として使用してもよい。また、本発明の懸濁液中の金属微粒子と化学結合する官能基である水酸基、メルカプト基、ジスルフィド基、アミノ基、カルボキシル基などで上記具体例の水溶性高分子が修飾されていてもよい。
本発明の懸濁液に含まれる水溶性高分子の種類は、該懸濁液の用途に応じて選択することができる。例えば、生体実験や細胞実験に使用する場合、生体適合性が良好なポリビニルピロリドン、ポリエチレングリコールやポリビニルアルコールなどを用いてもよい。
本発明の懸濁液が水溶性高分子を含む場合には、その懸濁液中に溶解又は分散している水溶性高分子の濃度(懸濁液1リットルあたりの水溶性高分子のモル数)は、50μM以下、好ましくは25μM以下、特に好ましくは10μM以下である。水溶性高分子の濃度が50μM以下であると、該濃度が50μMを超える場合と比較して、本発明の被験物質に対する特異的結合物質を担持した金被覆銀ナノプレートの懸濁液による該被験物質の検出感度を高めることができる。
また、別の好ましい態様では、本発明の懸濁液は水溶性高分子を含まない(すなわち0μM)。本発明の金被覆銀ナノプレートは、水溶性高分子がなくても、被験物質に対する特異的結合物質を担持することで安定な懸濁液を形成することができるので、特に被験物質の検出感度を高めるためには、水溶性高分子を含まなくてもよい。
本発明は特定の理論に拘束されるものではないが、水溶性高分子の濃度が50μM以下であると、例えば、後述する被験物質に対する特異的結合物質が金被覆銀ナノプレートに良好に担持されるか、又は、被験物質に対する特異的結合物質を担持した金被覆銀ナノプレートが該被験物質と安定した複合体を形成するため、結果として該被験物質の検出感度が高まると考えられる。
NMRの場合、金被覆銀ナノプレートの懸濁液を乾燥させ、得られた固形分を重溶媒で溶解(分散)させて測定する。重溶媒には既知濃度の内部標準物質(例えば、マレイン酸)を予め添加し、得られたスペクトル中の水溶性高分子由来のシグナルの積分値と内部標準物質由来のシグナルの積分値を比較することで水溶性高分子の濃度を算出することができる。
また、GPCの場合、初めに複数の既知濃度の水溶性高分子水溶液を分析し、得られたチャート中の水溶性高分子由来のピーク面積より検量線を作成する。次に、金被覆銀ナノプレートの懸濁液を測定し、得られたチャート中の水溶性高分子由来のピーク面積より水溶性高分子の濃度を算出することができる。
さらに、TG-DTAの場合、金被覆銀ナノプレート懸濁液の乾燥固形分を測定した際の、重量変化から水溶性高分子量を算出することができる。
本発明は特定の理論に拘束されるものではないが、懸濁液のpHが10以下であると、例えば、後述する被験物質に対する特異的結合物質が金被覆銀ナノプレートに良好に担持されるか、又は、被験物質に対する特異的結合物質を担持した金被覆銀ナノプレートが該被験物質と安定した複合体を形成するため、結果として該被験物質の検出感度が高まると考えられる。
本発明における懸濁液のpHは、本技術分野で用いられている一般的なpH測定法、例えばガラス電極法において測定された室温下(20~30℃)におけるpHをいう。
懸濁液のpHを調整するために、懸濁液へpH調整剤を添加してもよい。pH調整剤としては公知の物質を使用することができる。具体例としては、PBS緩衝液、炭酸ナトリウム、水酸化ナトリウムやクエン酸等をあげることができる。
被験物質又は被験物質に結合する物質としての核酸は、例えば、DNA、RNA、オリゴヌクレオチド、ポリヌクレオチド、又は、それらの増幅物であってもよい。
前記複合体は、被験物質の検出の分野で通常用いられる手段又は凝集物若しくは沈殿物を検出するのに通常用いられる手段を、特に制限なく利用することによって検出することができる。例えば、前記複合体の形成を、消光度測定、吸光度測定、濁度測定、粒度分布測定、粒子径測定、ラマン散乱光測定、色調変化の観察、凝集又は沈殿形成の観察、イムノクロマトグラフィー法、電気泳動、及び、フローサイトメトリーから成る群から選択される手段によって検出してもよい。
1.被験物質を含まない展開液を使用したイムノクロマト試験時の検出ラインと検出ライン以外の部分の輝度差

また、近赤外域の光を吸収する金被覆銀ナノプレートを使用したイムノクロマト試験においては、試験後に近赤外光を検出部に照射し、そのときの吸光を測定することで検出を確認することができる
(1)銀ナノプレートの種粒子の作製
2.5mMのクエン酸ナトリウム水溶液20mLに、0.5g/Lの分子量70,000ポリスチレンスルホン酸水溶液1mLと、10mMの水素化ホウ素ナトリウム水溶液1.2mLとを添加し、次いで、20mL/minで攪拌しながら、0.5mMの硝酸銀水溶液50mLを添加した。得られた溶液をインキュベーター(30℃)中で60分間静置し、銀ナノプレートの種粒子の水懸濁液を作製した。作製した水懸濁液(原液)の光学特性を図3に示す。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。最大吸収を示す波長は球状銀ナノ粒子のLSPRである396nm(消光度3.3)であった。なお、本発明の消光度とは懸濁液を分光光度計で測定した際の吸光度の値である。また、SEM観察写真を図4に示す。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。粒子径は主に3nm以上、10nm未満のプレート状粒子であった。
超純水200mLに、10mMのアスコルビン酸水溶液4.5mLを添加し、更に(1)で作製した銀ナノプレート種粒子水懸濁液4mLを添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/minで攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸ナトリウム水溶液20mLを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中で100時間静置し、銀ナノプレートの水懸濁液aを作製した。調製した懸濁液を超純水で4倍容に希釈した水懸濁液の光学特性を図5に示す。最大吸収を示す波長は526nm(消光度1.1)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。水懸濁液a中の銀ナノプレートをSEMにより観察したところ、銀ナノプレートの平均粒子径は31nmであり、平均厚さは8nmでアスペクト比は3.8であった。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。
(2)で作製した銀ナノプレート水懸濁液120mLに、0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液(以下、懸濁液A1)を作製した。
懸濁液A1の主要分散媒は水であった。
懸濁液A1に含まれる金被覆銀ナノプレートは、主面の最大長(粒子径)の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液A1に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
懸濁液A1におけるポリスチレンスルホン酸(本発明における水溶性高分子に該当)の濃度は0.98nMであった。
懸濁液A1におけるPVP(本発明における水溶性高分子に該当)の濃度は8.1μMであった。なお、金の厚みの平均は、HAADF-STEM像から任意の金被覆ナノプレート粒子10個について、各粒子の任意の部位10点について、金の厚みを測定した計100点のデータの内、上下10%を除いた80点の平均値を用いた(後述の懸濁液B1及びD1も同様)。
懸濁液A1のpHは、室温下(20℃)で4.0であった。pH測定は株式会社堀場製作所のtwinpHメータ B-212(ガラス電極法)を用いた(以下、同様)。
懸濁液A1の銀含有率は、懸濁液の総質量に対して0.0016質量%であった。
(3)で得られた懸濁液A1の1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆ナノプレート懸濁液(以下、懸濁液B1)を作製した。
懸濁液B1の主要分散媒は水であった。
懸濁液B1に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液B1に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。SEM観察写真を図6に示す。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。
懸濁液B1の3倍希釈溶液の色調はマゼンタであった。
懸濁液B1の色度座標はx=0.4276、y=0.1751であった。CIE1931xy色度図における色度座標を図1に示す。
色度座標の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び分光光度計操作用ソフトウェア「カラー測定ソフトウェア(株式会社島津製作所製)」にて照明:D65、視野:2°と設定した条件下で行われた。
懸濁液B1におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液B1におけるPVPの濃度は7.8μMであった。
懸濁液B1のpHは、室温下(20℃)で7.3であった。
懸濁液B1の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
実施例1の(4)と同様にして、実施例1の(3)で作製した懸濁液A1(pH4.0)を190mM 炭酸ナトリウム水溶液でpH5.2に調整し、懸濁液F1を作製した。懸濁液F1におけるポリスチレンスルホン酸の濃度は0.94nMであり、懸濁液F1におけるPVPの濃度は7.8μMであった。
実施例3
実施例1の(4)と同様にして、実施例1の(3)で作製した懸濁液A1(pH4.0)を190mM 炭酸ナトリウム水溶液でpH9.8に調整し、懸濁液G1を作製した。懸濁液G1におけるポリスチレンスルホン酸の濃度は0.94nMであり、懸濁液G1におけるPVPの濃度は7.8μMであった。
実施例1の(2)で作製した銀ナノプレートの水懸濁液120mLに、超純水9.1mL、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.99mLに200mMのPBS緩衝液0.01mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液I1)を作製した。
懸濁液I1の主要分散媒は水であった。
懸濁液I1に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液I1に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。

実施例1の(2)で作製した銀ナノプレートの水懸濁液120mLに、0.780mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。

実施例1の(2)で作製した銀ナノプレートの水懸濁液120mLに、0.025mMの末端がチオール基で修飾されたポリエチレングリコールであるSUNBRIGHT ME-020SH(分子量:2,000、NOF CORPORATION製)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液K1)を作製した。

銀ナノプレートの作製(色調:イエロー)
超純水200mLに、10mMのアスコルビン酸水溶液4.5mLを添加し、更に実施例1の(1)で作製した銀ナノプレート種粒子水懸濁液12mLを添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/minで攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸ナトリウム水溶液20mLを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中で100時間静置し、銀ナノプレートの水懸濁液bを作製した。調製した懸濁液を超純水で4倍容に希釈した水懸濁液の光学特性を図5に示す。最大吸収を示す波長は454nm(消光度1.1)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。水懸濁液b中の銀ナノプレートをSEMにより観察したところ、銀ナノプレートの平均粒子径は18nmであり、平均厚さは8nmでアスペクト比は2.2であった。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。
上述のように作製した銀ナノプレート水懸濁液120mLに、0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液(以下、懸濁液M1)を作製した。

懸濁液N1の3倍希釈溶液の色調はイエローであった。
懸濁液N1の色度座標はx=0.5070、y=0.4774であった。CIE1931xy色度図における色度座標を図1に示す。
また、懸濁液B1とN1を混合した混合液(B1+N1)の色調は赤色であった。混合液(B1+N1)の色度座標はx=0.6057、y=0.3317であった。

(1)銀ナノプレートの作製(色調:シアン)
超純水200mLに、10mMのアスコルビン酸水溶液4.5mLを添加し、更に実施例1の(1)で作製した銀ナノプレート種粒子水懸濁液2mLを添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/minで攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸ナトリウム水溶液20mLを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中で100時間静置し、銀ナノプレートの水懸濁液cを作製した。調製した懸濁液を超純水で4倍容に希釈した水懸濁液の光学特性を図5に示す。最大吸収を示す波長は626nm(消光度1.1)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。水懸濁液c中の銀ナノプレートをSEMにより観察したところ、銀ナノプレートの平均粒子径は50nmであり、平均厚さは10nmでアスペクト比は5.0であった。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。

混合液(B1+P1)の色度座標はx=0.1731、y=0.0675、混合液(N1+P1)の色度座標はx=0.2549、y=0.5712であった。CIE1931xy色度図における色度座標を図2に示す。
懸濁液P1におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液P1におけるPVPの濃度は7.8μMであった。

(1)銀ナノプレートの作製(色調:薄いシアン)
超純水200mLに、10mMのアスコルビン酸水溶液4.5mLを添加し、更に実施例1の(1)で作製した銀ナノプレート種粒子水懸濁液1mLを添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/minで攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸ナトリウム水溶液20mLを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中で100時間静置し、銀ナノプレートの水懸濁液dを作製した。調製した懸濁液を超純水で4倍容に希釈した水懸濁液の光学特性を図5に示す。最大吸収を示す波長は704nm(消光度1.0)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。水懸濁液d中の銀ナノプレートをSEMにより観察したところ、銀ナノプレートの平均粒子径は74nmであり、平均厚さは8nmでアスペクト比は9.2であった。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。
上述のように作製した銀ナノプレート水懸濁液120mLに、0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液(以下、懸濁液Q1)を作製した。
懸濁液Q1の主要分散媒は水であった。
懸濁液Q1に含まれる金被覆銀ナノプレートは、主面の最大長(粒子径)の平均が74nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液Q1に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
SEM観察写真を図9に示す。SEM観察写真の解析には株式会社日立製作所製の走査電子顕微鏡SU-70を用いた。
懸濁液Q1の3倍希釈溶液の色調は薄いシアンであった。
懸濁液Q1におけるポリスチレンスルホン酸(本発明における水溶性高分子に該当)の濃度は0.98nMであった。

(1)金被覆銀ナノプレートの作製
上述の実施例1の(3)金被覆銀ナノプレートの作製、における塩化金酸水溶液の濃度を0.14mMから0.42mMに変更した以外は、懸濁液A1の作製と同様にして金被覆銀ナノプレートの水懸濁液(以下、懸濁液S1)を作製した。
懸濁液S1の主要分散媒は水であった。
懸濁液S1に含まれる金被覆銀ナノプレートは、主面の最大長(粒子径)の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液S1に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.85nmであった。
懸濁液S1におけるポリスチレンスルホン酸の濃度は0.98nMであった。
懸濁液S1におけるPVPの濃度は8.1μMであった。なお、金の厚みの平均は、HAADF-STEM像から任意の金被覆ナノプレート粒子10個について、各粒子の任意の部位10点について、金の厚みを測定した計100点のデータの内、上下10%を除いた80点の平均値を用いた。
懸濁液S1のpHは、室温下(20℃)で4.0であった。pH測定は株式会社堀場製作所のtwinpHメータ B-212(ガラス電極法)を用いた。
懸濁液S1の銀含有率は、懸濁液の総質量に対して0.0016質量%であった。
(1)で得られた懸濁液S1の1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆ナノプレート懸濁液(以下、懸濁液T1)を作製した。
懸濁液T1の主要分散媒は水であった。

(1)水溶性高分子の濃度が高い例(その1)
実施例1の(2)で作製した銀ナノプレートの水懸濁液120mLに、1.25mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液D1)を作製した。

実施例1の(4)と同様にして、実施例1の(3)で作製した懸濁液A1(pH4.0)を200mM 水酸化ナトリウム水溶液でpH11.5に調整し、懸濁液H1を作製した。懸濁液H1におけるポリスチレンスルホン酸の濃度は0.94nMであり、懸濁液H1におけるPVPの濃度は7.8μMであった。
実施例1の(2)で作製した銀ナノプレートの水懸濁液120mLに、1.25mMの末端がチオール基で修飾されたポリエチレングリコールであるSUNBRIGHT ME-020SH(分子量:2,000、NOF CORPORATION製)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液L1)を作製した。

上述のように、本発明の金被覆銀ナノプレートの懸濁液及び比較例としての懸濁液を調製することができた。懸濁液A1、B1、F1、G1、I1,J1、K1、N1、P1、D1、H1、L1、又はT1中の水溶性高分子(ポリスチレンスルホン酸(PSS)、ポリビニルピロリドン(PVP)又はチオール修飾ポリエチレングリコール(PEG-SH))の濃度、pH及び色調(マゼンタ(M)、イエロー(Y)、シアン(C)又は薄いシアン(PC))を、後掲の表2にまとめた。
実施例及び比較例の各懸濁液を下記手順のイムノクロマト試験に用いて、試験結果を評価した。
(1)イムノクロマト試験に使用する展開液の作製(金被覆銀ナノプレートへの特異的結合物質の担持(検出試薬の標識))
5mMのPBS(-)緩衝液中における濃度50μg/mLのB型肝炎ウイルス抗原(HBs抗原)に対する抗体(品名:Goat anti HBsAg、製造元:Arista Biologicals,Inc.)(イムノクロマト法における検出抗体)の溶液0.2mLと、金被覆銀ナノプレート懸濁液(懸濁液A1、B1、F1、G1、I1,J1、K1、N1、P1、D1、H1、L1、又はT1)1.8mLを混合し、得られた混合物を室温下で30分間振とうした。次いで、遠心分離(25000rpm、4℃、10分間)を行い、抗体-金被覆銀ナノプレート複合体を沈殿させ、上澄み液1.85mLを除去した。その後、抗体-金被覆銀ナノプレート複合体を、4.9μMのBSAを含有する1.85mLの5mM PBS(-)緩衝液を添加して再分散させ、紫外可視分光光度計 Agilent 8453(アジレント・テクノロジー株式会社製)を使用して消光度(Extinction)2.0になるように調整し、展開液を作製した。
図10に示す手順により、B型肝炎ウイルス抗原(HBs抗原)を被験物質としたイムノクロマト試験を、抗HBs抗原抗体(捕獲抗体)を直線状(図10の検出ライン)に固定化したイムノクロマト試験紙を用いて行った。
イムノクロマト試験紙はイムノクロマト試験紙の受託作製会社(有限会社バイオデバイステクノロジー)より購入したものを使用した。捕獲抗体を直線状に固定する際、捕獲抗体を5mM PBS(-)緩衝液で濃度1g/mLに調整したものを使用した。
第1展開液(図10(a)で用いる展開液)は、5mMのPBS(-)緩衝液中におけるHBs抗原(品名:HBsAg Protein(Subtype adr)、製造元:Fitzgerald Industries International Inc.)の溶液であった。第1展開液として、HBs抗原の濃度が0.60μM、0.06μM、0.006μM、0.0006μM又は0M(ブランク)の溶液を用意した。
第2展開液は5mMのPBS(-)緩衝液であった。
第3展開液(図10(c)で用いた展開液)は、上述の展開液A1、B1、F1、G1、I1,J1、K1、N1、P1、D1、H1、L1又はT1であった。
イムノクロマト試験紙に、第1展開液15μLを展開させた(図10(a))。第1展開液の展開により、HBs抗原が試験紙の検出ライン上に固定化された捕獲抗体によって捕獲される(図10(b))。
次いで、第2展開液30μLを展開させて、イムノクロマト試験紙上の余剰抗原の洗浄を行った。
最後に、第3展開液60μLを展開させた(図10(c))。第3展開液の展開により、試験紙の検出ライン上で捕獲されたHBs抗原へ検出抗体(抗体-金被覆銀ナノプレート複合体)が結合する(図10(d))。
同一の手順をHBs濃度の異なる各第1展開液を用いて繰り返した。
第3展開液展開後の検出ラインにおける金被覆銀ナノプレートの着色(懸濁液の色調、すなわち、展開液B1を使用した際はマゼンタ、展開液N1を使用した際はイエロー、展開液P1を使用した際はシアン)を目視で確認することにより、HBs抗原の有無を判定した。結果を後掲の表2に示す。
第3展開液展開後のイムノクロマト試験紙をスキャニング(装置名:Cano Scan LiDE500F、製造元:キヤノン株式会社)し、検出ライン(捕獲抗体の固定化部分)の最低輝度と、検出ライン以外の部分の最低輝度とを画像解析ソフト(Image-J:アメリカ国立衛生研究所でWayne Rasbandが開発したオープン・ソースで公有の画像処理ソフトウェア(http://imagej.nih.gov/ij/))を用いて数値化した。最低輝度は、対象領域(検出ライン又は検出ライン以外の部分)における異なる箇所の輝度を5回測定し、得られた数値の中央値を採用した。輝度差(検出ラインの最低輝度-検出ライン以外の部分の最低輝度)を計算し、結果を次の表2及び図11~23に示す。
また、pHが10以下の懸濁液A1、B1、F1、G1、I1、J1、K1、N1、P1、D1、L1、又はT1から調製した展開液は、pHが11.5の懸濁液H1から調製した展開液よりも高い輝度差が生じたので、これよりも検出感度が高いことがわかった。
そして、水溶性高分子(PVP又はPEG-SH)の濃度が50μM以下である懸濁液B1、F1、G1、I1、J1、K1、N1、P1、又はT1から調製した展開液は、水溶性高分子の濃度が78μMである懸濁液D1又はL1から調製した展開液よりも高い輝度差が生じたので、これらよりも検出感度が高いことがわかった。特に、懸濁液I1から調製した展開液I1は、0.0006μMのHBs抗原を使用した場合にも輝度差が生じた。
1.金属微粒子懸濁液の作製
1-1.銀ナノプレートの作製
1-1-1.銀ナノプレートの種粒子の作製
2.5mMのクエン酸ナトリウム水溶液20mLに、0.5g/Lの分子量70,000ポリスチレンスルホン酸水溶液1mLと、10mMの水素化ホウ素ナトリウム水溶液1.2mLとを添加し、次いで、20mL/minで攪拌しながら、0.5mMの硝酸銀水溶液50mLを添加した。得られた溶液をインキュベーター(30℃)中に60分間静置し、銀ナノプレートの種粒子の水懸濁液を作製した。
超純水200mlに、10mMのアスコルビン酸水溶液4.5mLを添加し、上述の銀ナノプレートの種粒子の水懸濁液12mlを添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/minで攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸ナトリウム水溶液20mlを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中に100時間静置し、プレート状銀ナノ粒子の水懸濁液である銀ナノプレート懸濁液A2を調製した。
上記銀ナノプレートの種粒子の水懸濁液の添加量を12mlから4mlに変更した以外は、銀ナノプレート懸濁液A2と同様にして、プレート状銀ナノ粒子の水懸濁液である銀ナノプレート懸濁液B2を調製した。
上記銀ナノプレートの種粒子の水懸濁液の添加量を12mlから2mlに変更した以外は、銀ナノプレート懸濁液A2の調製と同様にして、プレート状銀ナノ粒子の水懸濁液である銀ナノプレート懸濁液C2を調製した。
1-2-1.金被覆銀ナノプレート懸濁液A2の作製
上記銀ナノプレート懸濁液A2の120mlに、0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、金被覆銀ナノプレート懸濁液A2を調製した。
金被覆銀ナノプレート懸濁液A2に含まれる金被覆銀ナノプレートを走査型電子顕微鏡(SEM)により観察したところ、主面の最大長(粒子径)の平均が18nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。なお、金被覆銀ナノプレートの主面の最大長(粒子径)の平均は、SEM画像(株式会社日立製作所製の走査電子顕微鏡SU-70で撮影)中の任意の金被覆銀ナノプレート100個の最大長(粒子径)を測定した計100点のデータの内、上下10%を除いた80点の平均値を用いた(後述の金被覆銀ナノプレート懸濁液B2及びCも同様)。そして、金被覆銀ナノプレート懸濁液A2に含まれる金被覆銀ナノプレートにおける金の厚さは、高角散乱環状暗視野走査透過顕微鏡法(HAADF-STEM)によれば、平均0.25nmであった。なお、金の厚みの平均は、HAADF-STEM像から任意の金被覆銀ナノプレート粒子10個について、各粒子の任意の部位10点について、金の厚みを測定した計100点のデータの内、上下10%を除いた80点の平均値を用いた(後述の金被覆銀ナノプレート懸濁液B2及びCも同様)。
また、金被覆銀ナノプレート懸濁液A2における金の濃度は0.056mg/L、銀の濃度は0.226mg/Lであり、以下の計算方法により算出された金の厚さは0.21nmであった。

して、金及び銀を王水中へ溶解させた。

金被覆銀ナノプレート懸濁液A2におけるポリスチレンスルホン酸の濃度は0.98nMであった。

上記銀ナノプレート懸濁液B2の120mlに、0.125mMのPVP(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、金被覆銀ナノプレート懸濁液B2を調製した。
金被覆銀ナノプレート懸濁液B2に含まれる金被覆銀ナノプレートをSEMにより観察したところ、主面の最大長(粒子径)の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。そして、金被覆銀ナノプレート懸濁液B2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は、HAADF-STEMによれば、0.30nmであった。
また、金被覆銀ナノプレート懸濁液B2における金の濃度は0.045mg/L、銀の濃度は0.185mg/Lであり、上述のICP発光分析結果に基づいた計算方法により算出された金の厚さは0.28nmであった。
金被覆銀ナノプレート懸濁液B2におけるポリスチレンスルホン酸の濃度は0.98nMであった。
金被覆銀ナノプレート懸濁液B2におけるPVPの濃度は8.1μMであった。
金被覆銀ナノプレート懸濁液B2のpHは、室温下(20℃)で4.0であった。
金被覆銀ナノプレート懸濁液B2の銀含有率は、懸濁液の総質量に対して0.0016質量%であった。
上記銀ナノプレート懸濁液C2の120mlに、0.125mMのPVP(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、金被覆銀ナノプレート懸濁液C2を調製した。
金被覆銀ナノプレート懸濁液C2に含まれる金被覆銀ナノプレートをSEMにより観察したところ、主面の最大長(粒子径)の平均が50nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は10nmであった。そして、金被覆銀ナノプレート懸濁液B2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は、HAADF-STEMによれば、0.45nmであった。
また、金被覆銀ナノプレート懸濁液C2における金の濃度は0.047mg/L、銀の濃度は0.186mg/Lであり、上述のICP発光分析結果に基づいた計算方法により算出された金の厚さは0.41nmであった。
金被覆銀ナノプレート懸濁液C2におけるポリスチレンスルホン酸の濃度は0.98nMであった。
金被覆銀ナノプレート懸濁液C2におけるPVPの濃度は8.1μMであった。

1-3-1.pH調整した金被覆銀ナノプレート懸濁液(懸濁液D2)の調製

懸濁液D2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が18nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。そして、懸濁液D2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は、HAADF-STEMによれば、0.25nmであった。
懸濁液D2の3倍希釈溶液の色調はイエローであった。
懸濁液D2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液D2におけるPVPの濃度は7.8μMであった。

1-2-2で得られた金被覆銀ナノプレート懸濁液B2の1.95mLに200mMのPBS緩衝液(+又は-)0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液E2)を作製した。ここで、200mMのPBS緩衝液(+)とは、二価イオンを含むPBS緩衝液(1.0mMのMg2+及び1.8mMのCa2+)のことをいい、200mMのPBS緩衝液(-)とは、二価イオンを含まないPBS緩衝液のことをいう。
懸濁液E2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。そして、懸濁液E2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は、HAADF-STEMによれば、0.30nmであった。
懸濁液E2の3倍希釈溶液の色調はマゼンタであった。
懸濁液E2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液E2におけるPVPの濃度は7.8μMであった。
懸濁液E2のpHは、室温下(20℃)で7.3であった。
懸濁液E2の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
1-2-3で得られた金被覆銀ナノプレート懸濁液C2の1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレート懸濁液(以下、懸濁液F2)を作製した。
懸濁液F2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が50nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は10nmであった。そして、懸濁液F2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は、HAADF-STEMによれば、0.45nmであった。
懸濁液F2の3倍希釈溶液の色調はシアンであった。
懸濁液F2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液F2におけるPVPの濃度は7.8μMであった。
懸濁液F2のpHは、室温下(20℃)で7.3であった。
懸濁液F2の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
2-1.金被覆銀ナノプレートへの糖鎖の担持
2-1-1.金被覆銀ナノプレート(懸濁液E2)へのメルカプトエチルマンノースの担持 上記懸濁液E2(PBS緩衝液(+)で調製したもの)4mLの入った9mL容バイヤル瓶に1-メルカプトエチルマンノース(Mw:270.274)10mgを添加、30℃下で12時間、マグネチックスターラーを使用し500rpmで撹拌し、マンノースを担持した金被覆銀ナノプレートの懸濁液G2を調製した。
なお、1-メルカプトエチルマンノースは有機化学的手法にて合成した。具体的にはマンノース(関東化学株式会社製)を原料とし、1位のアセチル化、ブロモエチル化、チオアセチル化、脱アセチル化を経て合成した。1-メルカプトエチルマンノースの化学式を以下に示す。
2-2-1.金被覆銀ナノプレートへの抗B型肝炎ウイルス抗原抗体の担持
5mMのPBS(-)緩衝液中における濃度50μg/mLのB型肝炎ウイルス抗原(HBs抗原)に対する抗体(品名:Goat anti HBsAg、製造元:Arista Biologicals,Inc.)(以下、抗HBs抗原抗体ともいう)の溶液2mLと、上記懸濁液D2の18mLを混合し、得られた混合物を室温下で30分間振とうした。次いで、遠心分離(25000rpm、4℃、10分間)を行い、抗体を担持した金被覆銀ナノプレートを沈殿させ、上澄み液18.5mLを除去した。その後、上記抗体を担持した金被覆銀ナノプレートを5mMのPBS(-)緩衝液18.5mLで再分散させ、抗HBs抗原抗体を担持した金被覆銀ナノプレートの懸濁液H2を調製した。
上記懸濁液E2(PBS緩衝液(-)で調製したもの)及び懸濁液F2についても同様に抗体を担持させ、抗HBs抗原抗体を担持した金被覆銀ナノプレートの懸濁液I2及びJ2をそれぞれ調製した。
懸濁液H2、I2及びJ2中の抗HBs抗原抗体を担持した金被覆銀ナノプレートの最大吸収波長は、それぞれ458nm、532nm及び630nmだった。
調製した懸濁液H2、I2及びJ2は、緩衝液中で安定に分散しており、分光特性もほとんど変化しないかったことから、これらはいずれも酸化に対して安定性が高いことがわかった。
3-1.糖鎖を担持した金被覆銀ナノプレートとレクチンとの相互作用
上記マンノースを担持した金被覆銀ナノプレートの懸濁液G2(PBS(+))を5本のエッペンチューブへ500μLずつ分注、5mMのPBS(+)緩衝液中における濃度10μMのコンカナバリンA(略称:ConA、品名:コンカナバリンA、製造元:株式会社オイルミルズ製)溶液5μL(ConA物質量:50pmol、添加後の濃度:0.1μM)、10μL(ConA物質量:100pmol、添加後の濃度:0.2μM)、25μL(ConA物質量:250pmol、添加後の濃度:0.5μM)、50μL(ConA物質量:500pmol、添加後の濃度:1.0μM)を各エッペンチューブへ添加、撹拌後、30℃下で1時間静置した。
マンノースを担持した金被覆銀ナノプレートの懸濁液G2にConA(50μL)を添加した後の静置中に、該懸濁液の色調変化及び該懸濁液中の沈殿形成を目視によって観察した。結果を表4に示す。
各相互作用溶液の消光度測定を株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:300-1000nmの条件下で実施した。測定結果を図24及び表5に示す。
上記抗HBs抗体を担持した金被覆銀ナノプレートの懸濁液H2(PBS(-))を2本の9mL容バイヤル瓶へ5mLずつ分注し、5mMのPBS(-)緩衝液中における濃度5μMのB型肝炎ウイルス抗原(略称:HBsAg、品名:HBsAg Protein(Subtype adr)、製造元:Fitzgerald Industries International Inc.)溶液50μL(HBs抗原物質量:250pmol、添加後の濃度:50nM)、および、5mMのPBS(-)緩衝液中における濃度10μMのB型肝炎ウイルス抗原溶液50μL(HBs抗原物質量:500pmol、添加後の濃度:100nM)をそれぞれバイヤル瓶へ添加、撹拌後、30℃下で1時間静置した。
懸濁液H2から調製した各相互作用溶液の消光度測定を、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:300-1000nmの条件下で実施した。測定結果を図25Aに示す。
また、懸濁液I2又は懸濁液J2を使用した際の結果を図25B及び図25Cに示す。
抗HBs抗原抗体を担持した金被覆銀ナノプレートの懸濁液I2にHBs抗原を添加したときの波長700nm付近における消光度を、該懸濁液の濁度としてとして測定した。測定結果を図26に示すが、これは図25Bの一部拡大図である。
1.金被覆銀ナノプレートの作製
1-1.金被覆銀ナノプレート懸濁液(懸濁液K2)の作製
実施例11の1-1-3で作製した銀ナノプレート懸濁液B2の120mlに、0.125mMのPVP(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、金被覆銀ナノプレート懸濁液K2を調製した。
懸濁液K2に含まれる金被覆銀ナノプレートは、主面の最大長(粒子径)の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液K2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
懸濁液K2におけるポリスチレンスルホン酸(本発明における水溶性高分子に該当)の濃度は0.98nMであった。
懸濁液K2におけるPVPの濃度は8.1μMであった。なお、金の厚みの平均は、HAADF-STEM像から任意の金被覆銀ナノプレート粒子10個について、各粒子の任意の部位10点について、金の厚みを測定した計100点のデータの内、上下10%を除いた80点の平均値を用いた。
懸濁液K2のpHは、室温下(20℃)で4.0であった。pH測定は株式会社堀場製作所のtwinpHメータ B-212(ガラス電極法)を用いた(以下、同様)。
懸濁液K2の銀含有率は、懸濁液の総質量に対して0.0016質量%であった。
上記1-1で作製した懸濁液K2 1.95mLに200mMのPBS緩衝液0.05mL及び190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整した金被覆銀ナノプレート懸濁液(以下、懸濁液L2)を作製した。
懸濁液L2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液L2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
懸濁液L2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液L2におけるPVPの濃度は7.8μMであった。
懸濁液L2のpHは、室温下(20℃)で7.3であった。
懸濁液L2の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
実施例11の1-1-3で作製した銀ナノプレート懸濁液B2の120mlに、1.25mMのPVP(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整した金被覆銀ナノプレート懸濁液(以下、懸濁液M2)を作製した。
懸濁液M2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液M2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
懸濁液M2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液M2におけるPVPの濃度は78μMであった。
懸濁液M2のpHは、室温下(20℃)で7.3であった。
懸濁液M2の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
実施例11の1-1-3で作製した銀ナノプレート懸濁液B2の120mlに、0.125mMのPVP(分子量:40,000)の水溶液9.1mlを添加し、0.5Mのアスコルビン酸水溶液1.6mlを添加した後、0.42mMの塩化金酸水溶液9.6mlを0.5mL/minで攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの水懸濁液を作製した。
作製した金被覆銀ナノプレートの水懸濁液1.95mLに200mMのPBS緩衝液0.05mLおよび200mMの水酸化ナトリウム水溶液を攪拌しながら添加し、pH調整した金被覆銀ナノプレート懸濁液(以下、懸濁液N2)を作製した。
懸濁液N2に含まれる金被覆銀ナノプレートは、主面の最大長の平均が31nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は8nmであった。また、懸濁液N2に含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
懸濁液N2におけるポリスチレンスルホン酸の濃度は0.94nMであった。
懸濁液N2におけるPVPの濃度は7.8μMであった。
懸濁液N2のpHは、室温下(20℃)で11.5であった。
懸濁液N2の銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
5mMのPBS(-)緩衝液中における濃度50μg/mLのアフラトキシンB1に対する抗体(品名:AFB1(Aflatoxin B1)Antibody、製造元:IMMUNE CHEM)の溶液0.2mLと、金被覆銀ナノプレート懸濁液(懸濁液K2、L2、M2又はN2)1.8mLを混合し、得られた混合物を室温下で30分間振とうした。次いで、遠心分離(25000rpm、4℃、10分間)を行い、抗体を担持した金被覆銀ナノプレートを沈殿させ、上澄み液1.85mLを除去した。その後、抗体を担持した金被覆銀ナノプレートを4.9μMのBSAを含有する5mM PBS(-)緩衝液で再分散させ、紫外可視近赤外分光光度計(装置名:紫外可視近赤外分光光度計MPC3100UV-3100PC、製造元:株式会社島津製作所)を使用して消光度(Extinction)2.0になるように調整し、懸濁液である展開液K2、L2、M2又は、N2を作製した。展開液K2、L2、M2及びN2の最大吸収波長は、532nmだった。作製された展開液と金被覆銀ナノプレート懸濁液との関係を表6に示す。
実施例1の(2)と同様の手順により、アフラトキシンB1を被験物質としたイムノクロマトグラフィーを、抗アフラトキシンB1抗体(捕獲抗体)を直線状(図10の検出ライン)に固定化したイムノクロマト試験紙を用いて行った。
イムノクロマト試験紙はイムノクロマト試験紙の受託作製会社(有限会社バイオデバイステクノロジー)より購入したものを使用した。捕獲抗体を直線状に固定する際、抗アフラトキシンB1抗体を5mM PBS(-)緩衝液で濃度1g/mLに調整した溶液を使用した。
第1展開液(図10(a)で用いる展開液)は、5mMのPBS(-)緩衝液中におけるアフラトキシンB1(品名:アフラトキシンB1、製造元:Toronto Research Chemicals Inc.)の溶液であった。第1展開液として、アフラトキシンB1の濃度が0.60μM、0.06μM、0.006μM又は0M(ブランク)の溶液を用意した。
第2展開液は5mMのPBS(-)緩衝液であった。
第3展開液(図10(c)で用いる展開液)は、上記2で作製した展開液K2、L2、M2又はN2であった。
イムノクロマト試験紙に、第1展開液15μLを展開させた(図10(a))。第1展開液の展開により、アフラトキシンB1が試験紙の検出ライン上に固定化された捕獲抗体によって捕獲される(図10(b))。
次いで、第2展開液30μLを展開させて、イムノクロマト試験紙上の余剰抗原の洗浄を行った。
最後に、第3展開液60μLを展開させた(図10(c))。第3展開液の展開により、試験紙の検出ライン上で捕獲されたアフラトキシンB1へ検出抗体(抗アフラトキシンB1抗体を担持した金被覆銀ナノプレート)が結合する(図10(d))。
同一の手順をアフラトキシンB1濃度の異なる各第1展開液を用いて繰り返した。
第3展開液展開後の検出ラインにおける金被覆銀ナノプレートの着色(マゼンタ)を目視で確認することにより、アフラトキシンB1の有無を判定した。結果を表7に示す。
第3展開液展開後のイムノクロマト試験紙をスキャニング(装置名:Cano Scan LiDE500F、製造元:キヤノン株式会社)し、検出ライン(捕獲抗体の固定化部分)の最低輝度と、検出ライン以外の部分の最低輝度とを画像解析ソフト(Image-J:アメリカ国立衛生研究所でWayne Rasbandが開発したオープン・ソースで公有の画像処理ソフトウェア(http://imagej.nih.gov/ij/))を用いて数値化した。最低輝度は、対象領域(検出ライン又は検出ライン以外の部分)における異なる箇所の輝度を5回測定し、得られた数値の中央値を採用した。輝度差は、次の式:
輝度差=検出ラインの最低輝度-検出ライン以外の部分の最低輝度
により計算した。
Claims (14)
- 金被覆銀ナノプレートの懸濁液であって、
0~50μMの水溶性高分子を含み、
pHが10以下であることを特徴とする、懸濁液。 - 前記金被覆銀ナノプレート上の金の厚みの平均が、1.0nm以下である、請求項1に記載の懸濁液。
- 前記金被覆銀ナノプレート上の金の厚みの平均が、0.1~0.7nmである、請求項1又は2に記載の懸濁液。
- 前記水溶性高分子の濃度が、0~25μMである、請求項1~3のいずれか一項に記載の懸濁液。
- pHが4~10である、請求項1~4のいずれか一項に記載の懸濁液。
- pHが5~9である、請求項1~5のいずれか一項に記載の懸濁液。
- 前記金被覆銀ナノプレートが、被験物質に対する特異的結合物質を担持していることを特徴とする、請求項1~6のいずれか一項に記載の懸濁液。
- 前記被験物質と前記特異的結合物質の組み合わせが、抗原とそれに結合する抗体、抗体とそれに結合する抗原、糖鎖又は複合糖質とそれに結合するレクチン、レクチンとそれに結合する糖鎖又は複合糖質、ホルモン又はサイトカインとそれに結合する受容体、受容体とそれに結合するホルモン又はサイトカイン、タンパク質とそれに結合する核酸アプタマー若しくはペプチドアプタマー、酵素とそれに結合する基質、基質とそれに結合する酵素、ビオチンとアビジン又はストレプトアビジン、アビジン又はストレプトアビジンとビオチン、IgGとプロテインA又はプロテインG、プロテインA又はプロテインGとIgG、及び、第1の核酸とそれに結合する第2の核酸から成る群から選択される、請求項7に記載の懸濁液。
- 請求項7又は8に記載の懸濁液を使用して、前記被験物質を検出する方法であって、
該懸濁液を該被験物質と混合して、該被験物質と前記特異的結合物質を担持した金被覆銀ナノプレートとの複合体を形成する工程、及び、
該複合体を検出する工程を含むことを特徴とする、方法。 - 前記複合体の形成を、消光度測定、吸光度測定、濁度測定、粒度分布測定、粒子径測定、ラマン散乱光測定、色調変化の観察、凝集又は沈殿形成の観察、イムノクロマトグラフィー、電気泳動、及び、フローサイトメトリーから成る群から選択される手段によって検出する、請求項9に記載の方法。
- 前記複合体の形成を、200~2500nmの範囲の前記金被覆銀ナノプレートの吸収波長における消光度測定又は吸光度測定によって検出する、請求項9又は10に記載の方法。
- 前記複合体の形成を、430~2000nmの範囲の前記金被覆銀ナノプレートの最大吸収波長における消光度測定又は吸光度測定によって検出する、請求項9~11のいずれか一項に記載の方法。
- 前記複合体の形成を、前記金被覆銀ナノプレートの最大吸収波長よりも長波長側の前記複合体の形成に依存して消光度又は吸光度が上昇する波長領域における濁度測定によって検出する、請求項9又は10に記載の方法。
- 請求項7又は8に記載の懸濁液を含む、請求項9~13のいずれか一項に記載の方法に使用するためのキット。
